Abstract

The present invention is directed to a composition comprising a matrix suitable for implantation in humans, comprising defatted, shredded, allogeneic human muscle tissue that has been combined with an aqueous carrier and dried in a predetermined shape. Also disclosed is a tissue graft or implant comprising a matrix suitable for implantation in humans, comprising defatted, shredded, allogeneic human muscle tissue that has been combined with an aqueous carrier and dried in a predetermined shape. The composition and/or tissue graft or implant of the invention is usable in combination with seeded cells, a tissue growth factor, and/or a chemotactic agent to attract a desired cell.

Claims (43)

1. An implant comprising a matrix suitable for implantation in humans, said matrix comprising a dried slurry of allogeneic muscle tissue, wherein said implant further comprises a particulate component selected from the group consisting of demineralized bone matrix (DBM), mineralized bone matrix, cortical cancellous chips (CCC), crushed cancellous chips, tricalcium phosphate, hydroxyapatite, biphasic calcium phosphate and combinations thereof.

2. The implant of claim 1 , wherein said muscle tissue is striated muscle tissue.

3. The implant of claim 2 , wherein said striated muscle tissue is skeletal muscle.

4. The implant of claim 1 , wherein said muscle tissue is defatted.

5. The implant of claim 1 , wherein said muscle tissue is lyophilized.

6. The implant of claim 1 , wherein the particulate component constitutes from 50% to 99% of the dry weight of the composition.

7. The implant of claim 6 , wherein the particulate component is sandwiched between layers of the matrix.

The implant of claim 1 , further comprising a bone morphogenic protein (BMP), a LIM mineralization protein, a RUNX-2 protein, a transforming growth factor-α, a transforming growth factor-β, a fibroblast growth factor, a platelet derived growth factor, an epidermal growth factor, an insulin-like growth factor, or a combination thereof.

9. The implant of claim 8 , wherein said BMP is selected from the group consisting of BMP-2, BMP-4 and BMP-7.

10. The implant of claim 1 , wherein said muscle tissue is shredded.

11. The implant of claim 1 , wherein said implant is seeded with dermatocytes, fibroblasts, melanocytes, or osteogenic cells.

12. The implant of claim 1 , wherein the dried slurry is made from drying a solution of aqueous carrier and allogeneic muscle tissue.

13. The implant of claim 12 , wherein the aqueous carrier is acidic, basic or neutral.

14. The implant of claim 1 , further comprising collagen fibers.

15. The implant of claim 14 , wherein the dry weight ratio of the collagen fibers to muscle tissue is 10:90 to 90:10.

16. The implant of claim 14 , wherein the dry weight ratio of the collagen fibers to muscle tissue is 25:75 to 75:25.

17. The implant of claim 14 , wherein source of collagen fibers is tendon, fascia, or dermis.

18. An implantable sheet suitable for use in humans comprising a matrix of allogeneic muscle tissue that has been combined with a carrier and dried into a predetermined shape, wherein said implantable sheet further comprises a particulate component selected from the group consisting of demineralized bone matrix (DBM), mineralized bone matrix, cortical cancellous chips (CCC), crushed cancellous chips, tricalcium phosphate, hydroxyapatite, biphasic calcium phosphate and combinations thereof.

19. The implantable sheet of claim 18 , wherein said muscle tissue is striated muscle tissue.

20. The implantable sheet of claim 19 , wherein said striated muscle tissue is skeletal muscle.

21. The implantable sheet of claim 18 , wherein said muscle tissue is defatted.

22. The implantable sheet of claim 18 , wherein said muscle tissue is lyophilized.

23. The implantable sheet of claim 18 , wherein the particulate component constitutes from 50% to 99% of the dry weight of the composition.

24. The implantable sheet of claim 23 , wherein the particulate component constitutes from 90% to 99% of the dry weight of the implant.

The implantable sheet of claim 18 , further comprising a bone morphogenic protein (BMP), a LIM mineralization protein, or RUNX-2 protein, a transforming growth factor-α, a transforming growth factor-β, a fibroblast growth factor, a platelet derived growth factor, an epidermal growth factor, an insulin-like growth factor, or a combination thereof.

26. The implantable sheet of claim 18 , wherein said muscle tissue is also shredded.

27. The implantable sheet of claim 18 , wherein said implant is seeded with dermatocytes, fibroblasts, melanocytes, or osteogenic cells.

28. The implantable sheet of claim 18 , wherein the particulate component is uniformly distributed throughout the matrix.

29. The implantable sheet of claim 18 , wherein the particulate component is layered within said matrix.

30. The implantable sheet of claim 18 , wherein the shape of the implant is a sheet having dimensions of about 20 mm to 300 mm by 10 mm to 100 mm wide, and 1 mm to 10 mm thick.

31. The implantable sheet of claim 18 , wherein the shape of the implant is a sheet having dimensions of about 30 mm to 150 mm by 20 mm to 70 mm wide, and 1.5 mm to 8 mm thick.

32. The implantable sheet of claim 18 , wherein the shape of the implant is a sheet having dimensions of about 50 mm to 100 mm long, by 25 mm to 50 mm wide, and 2 mm to 5 mm thick.

33. The implantable sheet of claim 18 , wherein the shape of the implant is a sheet having dimensions of about 70 mm long, by 70 mm wide, and 0.5 mm thick.

34. The implantable sheet of claim 18 , wherein the sheet is made from drying a solution of aqueous carrier and allogeneic muscle tissue.

35. The implantable sheet of claim 34 , wherein the aqueous carrier is acidic, basic or neutral.

36. The implantable sheet of claim 34 , wherein the solution and the muscle tissue are in a ratio of volume of solution in milliliters to dry weight of muscle in grams in the range of 100:1 to 10:1.

37. The implantable sheet of claim 34 , wherein the solution and the muscle tissue are in a ratio of volume of solution in milliliters to dry weight of muscle in grams in the range of 70:1 to 30:1.

38. The implantable sheet of claim 18 , further comprising collagen fibers.

39. The implantable sheet of claim 38 , wherein the dry weight ratio of the collagen fibers to muscle tissue is 10:90 to 90:10.

40. The implantable sheet of claim 38 , wherein the dry weight ratio of the collagen fibers to muscle tissue is 25:75 to 75:25.

41. The implantable sheet of claim 38 , wherein source of collagen fibers is tendon, fascia, or dermis.

42. An intermediate composition for producing a matrix for an implant, the matrix comprising lyophilized animal muscle tissue and a biocompatible acid; wherein said intermediate composition further comprises a particulate component, wherein said particulate component is selected from the group consisting of demineralized bone matrix (DBM), mineralized bone matrix, cortical cancellous chips (CCC), crushed cancellous chips, tricalcium phosphate, hydroxyapatite, biphasic calcium phosphate and combinations thereof.

43. The composition of claim 42 , wherein the biocompatible acid is selected from the group consisting of acetic acid, citric acid, formic acid, hydrochloric acid, lactic acid, phosphoric acid, and sulfuric acid.
