Abstract

Crystals of isopropyl ester of N-(2,6-dichlorobenzoyl)-4-[6-(methylamino)methyl-1-methyl-2,4-dioxo-1,4-dihydroquinazoline-3(2H)-yl]-L-phenylalanine hydrochloride are useful as α4 integrin inhibitors.

Claims (24)

1. A crystal of the hydrochloride salt of the compound of formula (I):

which is either:

crystal form Cl 1, which has peaks at the diffraction angles (2θ) 2.9°, 7.7°, 11.6°, and 20.3° in its powder X-ray diffraction pattern;

crystal form Cl 2, which has peaks at the diffraction angles (2θ) 8.5°, 10.5°, 16.0°, 17.0°, and 23.6° in its powder X-ray diffraction pattern;

crystal form Cl 3, which has peaks at the diffraction angles (2θ) 3.6°, 8.9°, and 12.4° in its powder X-ray diffraction pattern;

crystal form Cl 4, which has peaks at the diffraction angles (2θ) 4.9° and 17.5° in its powder X-ray diffraction pattern; or

crystal form Cl 5, which has peaks at the diffraction angles (2θ) 8.2°, 11.2°, 14.3°, and 15.6° in its powder X-ray diffraction pattern.

2. A crystal according to claim 1 , which is crystal form Cl 5.

3. A method of making a crystal according to claim 2 , which comprises adding hydrogen chloride to a mixture comprising the compound of formula (I) and acetone.

4. A method of making a crystal according to claim 2 , which comprises:

heating the hydrochloride salt of the compound of formula (I) in a solvent comprising acetone and water, to obtain a first heated mixture;

adding an acetone solvent to said first heated mixture, to obtain a second heated mixture; and

cooling said second heated mixture.

A method of making a crystal according to claim 2 , which comprises mixing the hydrochloride salt of the compound of formula (I) and two or more solvents selected from the group consisting of acetone, water, tetrahydrofuran, acetonitrile, methanol, ethanol, propanol and isopropyl acetate to obtain a suspension thereof.

6. A pharmaceutical composition, which comprises a crystal according to claim 1 and a pharmaceutically acceptable carrier.

7. A crystal according to claim 1 , which is crystal form Cl 1.

8. A crystal according to claim 1 , which is crystal form Cl 2.

9. A crystal according to claim 1 , which is crystal form Cl 3.

10. A crystal according to claim 1 , which is crystal form Cl 4.

11. A pharmaceutical composition, which comprises a crystal according to claim 2 and a pharmaceutically acceptable carrier.

12. A pharmaceutical composition, which comprises a crystal according to claim 7 and a pharmaceutically acceptable carrier.

13. A pharmaceutical composition, which comprises a crystal according to claim 8 and a pharmaceutically acceptable carrier.

14. A pharmaceutical composition, which comprises a crystal according to claim 9 and a pharmaceutically acceptable carrier.

15. A pharmaceutical composition, which comprises a crystal according to claim 10 and a pharmaceutically acceptable carrier.
