Abstract

A method of manufacturing a tissue matrix for implantation into a patient is disclosed. The method sets forth collecting embryonic stem cells from a placenta which has been treated to remove residual cord blood and seeding the collected stem cells onto or into a tissue matrix. The seeded tissue matrix is then implanted on or into a patient. The seeded tissue matrix made by the method of the present invention is also disclosed.

Claims (55)

1. A method of preparing a tissue matrix, comprising seeding human placental stem cells into or onto a decellularized or synthetic tissue matrix, wherein said stem cells are from a human placenta that has been drained of cord blood and perfused to remove residual blood, and comprise stem cells that are CD34 − or OCT-4 + , wherein said placental stem cells are SSEA4 − .

2. The method of claim 1 , wherein said placental stem cells are additionally CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SH2 + , SH3 + , SH4 + , or SSEA3 − .

3. The method of claim 1 , additionally comprising seeding CD34 + human placental stem cells onto said tissue matrix.

4. The method of claim 1 , wherein said stem cells are obtained from a placenta that has been perfused for at least 2 hours.

5. The method of claim 1 , wherein said stem cells are obtained from a placenta that has been perfused for at least 12 hours.

6. The method of claim 1 , wherein said stem cells are obtained from a placenta that has been perfused for at least 24 hours.

7. The method of claim 1 , wherein said decellularized tissue is contacted with serum and fibronectin prior to said seeding.

8. A decellularized or synthetic tissue matrix comprising human placental stem cells that are CD34 − or OCT-4 + , wherein said placental stem cells are SSEA4 − .

9. The tissue matrix of claim 8 , wherein said placental stem cells are additionally CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SH2 + , SH3 + , SH4 + , or SSEA3 − , or any combination of the foregoing.

10. The tissue matrix of claim 8 additionally comprising CD34 + stem cells.

11. The tissue matrix of claim 8 , wherein said tissue matrix comprises a synthetic tissue matrix.

12. The tissue matrix of claim 11 , wherein said synthetic tissue matrix comprises polylactide-co-glycolide, a hydrated collagen lattice, or a collagen cross-linked with glycosaminoglycan.

13. The tissue matrix of claim 10 , wherein said tissue matrix comprises a synthetic tissue matrix.

14. The tissue matrix of claim 13 , wherein said synthetic tissue matrix comprises polylactide-co-glycolide, a hydrated collagen lattice, or a collagen cross-linked with glycosaminoglycan.

15. The tissue matrix of claim 8 , wherein the tissue matrix comprises a decellularized tissue.

16. The tissue matrix of claim 15 , wherein said decellularized tissue is non-human.

The tissue matrix of claim 15 , wherein said tissue matrix comprises exogenous fibronectin, glycosaminoglycan, human serum, fetal bovine serum, fibroblast growth factor (FGF), platelet-derived growth factor (PDGF), transforming growth factor-β (TGF-β), fibrillar collagen, or collagen fragments.

18. The tissue matrix of claim 10 , wherein the tissue matrix comprises a decellularized tissue.

19. The tissue matrix of claim 18 , wherein said decellularized tissue is non-human.

The tissue matrix of claim 18 , wherein said tissue matrix comprises exogenous fibronectin, glycosaminoglycan, human serum, fetal bovine serum, fibroblast growth factor (FGF), platelet-derived growth factor (PDGF), transforming growth factor-β (TGF-β), fibrillar collagen, or collagen fragments.

21. The method of claim 1 , wherein said placental stem cells are seeded onto a decellularized tissue matrix.

22. The method of claim 21 , wherein said decellularized tissue matrix is decellularized heart tissue.

23. The method of claim 22 , wherein said decellularized heart tissue is a whole decellularized heart.

24. The method of claim 23 , wherein said heart is human.

25. The method of claim 23 , wherein said heart is porcine.

26. The method of claim 22 , comprising contacting said decellularized heart tissue with fibronectin or a glycosaminoglycan prior to said seeding.

27. The method of claim 22 , comprising contacting said decellularized heart tissue with fibronectin and a glycosaminoglycan prior to said seeding.

28. The method of claim 26 , wherein said glycosaminoglycan is heparin.

29. The method of claim 27 , wherein said fibronectin and said glycosaminoglycan are present in a ratio of about 10:1 fibronectin:glycosaminoglycan.

30. The method of claim 22 , wherein said decellularized heart tissue is obtained by contacting said heart tissue with a solution comprising DNase I and RNase A for a time sufficient for said DNase I and RNase A to degrade DNA and RNA in cells of the heart tissue.

31. The method of claim 30 , wherein said solution is hypotonic to said cells of the heart tissue.

32. The tissue matrix of claim 15 , wherein said decellularized tissue is heart tissue.

33. The tissue matrix of claim 32 , wherein said heart tissue is a whole heart.

34. The tissue matrix of claim 33 , wherein said heart is human.

35. The tissue matrix of claim 33 , wherein said heart is porcine.

36. A tissue comprising a decellularized organ and a plurality of CD34 − or OCT-4 + human placental stem cells, wherein said placental stem cells are SSEA4 − .

37. The tissue of claim 36 , wherein said placental stem cells are from a human placenta that has been drained of cord blood and perfused to remove residual blood.

38. The tissue of claim 36 , wherein said decellularized organ is a heart.

39. The tissue of claim 36 , wherein said stem cells are allogenic to said decellularized organ.

40. A method of producing a tissue comprising contacting a decellularized organ with a plurality of CD34 − or OCT-4 + human placental stem cells, wherein said placental stem cells are SSEA4 − .

41. A method of transplanting a tissue into a recipient, comprising transplanting said tissue into a recipient, wherein said tissue comprises a decellularized organ and a plurality of CD34 − or OCT-4 + human placental stem cells, wherein said placental stem cells are SSEA4 − , and wherein said placental stem cells are from a human placenta that has been drained of cord blood and perfused to remove residual blood.

42. The method of claim 41 , wherein said stem cells are allogenic to said recipient.

43. The method of claim 41 , wherein said tissue is allogenic or xenogenic to said recipient.

44. A tissue matrix comprising isolated CD34 − , CD10 + , SH2 + multipotent placental cells.

45. The tissue matrix of claim 44 wherein said multipotent placental cells are additionally CD90 + or CD45 − .

46. The tissue matrix of claim 45 wherein said multipotent placental cells are additionally CD90 + and CD45 − .

47. The tissue matrix of claim 45 wherein said multipotent placental cells are additionally one or more of CD29 + , CD38 − , CD44 + , CD54 + , SH3 + or SH4 + .

48. The tissue matrix of claim 46 wherein said multipotent placental cells are additionally one or more of CD29 + , CD38 − , CD44 + , CD54 + , SH3 + or SH4 + .

49. The tissue matrix of claim 47 wherein said multipotent placental cells are CD44 + .

50. The tissue matrix of claim 48 wherein said multipotent placental cells are CD44 + .

51. The tissue matrix of claim 44 wherein said multipotent placental cells are tissue culture plastic adherent.

52. The tissue matrix of claim 44 wherein said multipotent placental cells have the potential to differentiate into a cell expressing one or more characteristics of neural cells.

53. The tissue matrix of claim 44 wherein said multipotent placental cells have the potential to differentiate into a cell expressing one or more characteristics of osteogenic or chondrogenic cells.

54. The tissue matrix of claim 2 , wherein said placental stem cells are CD29 + , CD90 + , SH2 + , SH3 + , SH4 + , CD34 − , and CD45 − .

55. A tissue matrix comprising isolated human multipotent placental cells, wherein said placental stem cells are CD29 + , CD90 + , SH2 + , SH3 + , SH4 + , CD34 − , and CD45 − .
