Humanized GM-CSF antibodies
View Patent ↗Chimeric antibodies, as well as fusion proteins which comprise chimeric antibodies, are disclosed. The antibodies bind to GM-CSF, CD-30, and G250 antigen. The fusion proteins include biologically active portions of tumor necrosis factor, or full length tumor necrosis factor. Expression vectors adapted for production of the antibodies, as well as methods for manufacturing these, are also disclosed.
1. An isolated nucleic acid molecule which encodes a chimerized, GM-CSF specific antibody light chain, the amino acid sequence of which consists of the amino acid encoded by nucleotides 1357-1752 of SEQ ID NO: 35, concatenated to the amino acid sequence encoded by nucleotides 1886-2203 of SEQ ID NO: 35.
2. An isolated nucleic acid molecule which encodes a chimerized, GM-CSF specific antibody heavy chain, the amino acid sequence of which consists of the amino acid encoded by nucleotides 1357-1764 of SEQ ID NO: 36, concatenated to the amino acid sequence encoded by nucleotides 1839-2825 of SEQ ID NO: 36.
3. An expression vector which comprises the isolated nucleic acid molecule of claim 1 , operably linked to a promoter.
4. An expression vector which comprises the isolated nucleic acid molecule of claim 2 , operably linked to a promoter.
5. An expression vector of claim 3 , consisting of the nucleotide sequence of SEQ ID NO: 35.
6. An expression vector of claim 4 , consisting of the nucleotide sequence of SEQ ID NO: 36.
7. A recombinant cell, transformed or transfected with the isolated nucleic acid molecule of claim 1 .
8. A recombinant cell, transformed or transfected with the isolated nucleic acid molecule of claim 2 .
9. The isolated nucleic acid molecule of claim 1 comprising nucleotides 1357-1752 and 1886-2203 of SEQ ID NO: 35.
10. The isolated nucleic acid molecule of claim 2 comprising nucleotides 1357-1764 and 1839-2825 of SEQ ID NO: 36.
11. A recombinant cell transformed or transfected with the expression vector of claim 3 .
12. A recombinant cell transformed or transfected with the expression vector of claim 4 .
