Abstract

The invention provides a novel additive for improved analysis by mass spectrometry. More specifically, ascorbic acid has been found to reduce or eliminate the presence of adducts commonly present in mass spectra. The improved processes and compositions of the invention allow for increased accuracy, sensitivity and throughput for samples analyzed by mass spectrometry.

Claims (16)

1. A method of mass spectrometric analysis, comprising:

(a) providing a composition comprising an analyte and an additive that includes ascorbic acid, or a salt, tautomer or analog thereof, and

(b) analyzing the analyte by mass spectrometry, wherein the additive in the composition is in an amount that reduces the amount of an adduct formed under mass spectrometry conditions.

2. The method of claim 1 , wherein the additive consists essentially of ascorbic acid, or a salt, tautomer or analog thereof.

3. The method of claim 1 , wherein the additive consists of ascorbic acid, or a salt, tautomer or analog thereof.

4. The method of claim 1 , wherein the additive consists essentially of ascorbic acid.

5. The method of claim 1 , wherein the additive consists of ascorbic acid.

6. The method of claim 1 , wherein the additive further comprises ammonium oxalate.

7. The method of claim 1 , wherein the analyte is a nucleic acid.

8. The method of claim 7 , wherein the nucleic acid is deoxyribonucleic acid.

9. The method of claim 7 , wherein the nucleic acid is ribonucleic acid.

The method of claim 1 , wherein the mass spectrometry is selected from the group consisting of: Matrix-Assisted Laser Desorption/Ionization Time-of-Flight (MALDI-TOF) Mass Spectrometry, Laser Desorption Mass Spectrometry (LDMS), Electrospray (ES) Mass Spectrometry, Ion Cyclotron Resonance (ICR) Mass Spectrometry, and Fourier Transform Mass Spectrometry.

11. The method of claim 1 , wherein the composition comprises 5 mM to 50 mM ascorbic acid, or a salt, tautomer or analog thereof.

12. The method of claim 1 , wherein the composition comprises a mass spectrometry matrix.

13. The method of claim 12 , wherein the mass spectrometry matrix comprises 3-hydroxy picolinic acid (3-HPA).

14. The method of claim 12 , wherein the mass spectrometry matrix comprises di-ammonium citrate (DAC).
