Abstract

The present invention encompasses influenza vaccines, in particular canine influenza vaccines. The vaccine may be a recombinant poxvirus vaccine or an inactivated vaccine. The invention also encompasses recombinant poxvirus vectors encoding and expressing influenza antigens, epitopes or immunogens which can be used to protect animals, in particular dogs, against influenza.

Claims (20)

1 . A Canidae vaccine comprising an effective amount of an expression vector that contains and expresses in vivo in a canine a polynucleotide encoding equine influenza antigen comprising equine influenza H3 operatively linked to a promoter and optionally to an enhancer, and a pharmaceutically or veterinarily acceptable carrier, excipient, or vehicle.

2 . The vaccine of claim 1 , wherein the polynucleotide comprises H3N8 codon optimized HA.

3 . The vaccine according to claim 1 , wherein the polynucleotide is isolated from a canine infected with equine influenza.

4 . The vaccine according to claim 3 , wherein the polynucleotide is isolated from a broncho alveolar lavage of the infected canine.

5 . The vaccine according to claim 3 , wherein the polynucleotide is isolated from lung tissue of the infected canine.

6 . The vaccine according to claim 1 , wherein the polynucleotide is isolated from an equine influenza virus isolate.

7 . The vaccine according to claim 6 , wherein the equine influenza isolate is selected from the group consisting of an Ohio equine influenza virus isolate, a Kentucky equine influenza virus isolate, a Newmarket equine influenza virus isolate or mixtures thereof.

8 . The vaccine according to claim 6 , wherein the polynucleotide is isolated from an Ohio equine influenza virus isolate.

9 . The vaccine according to claim 6 , wherein the polynucleotide is isolated from a Kentucky equine influenza virus isolate.

10 . The vaccine according to claim 6 , wherein the polynucleotide is isolated from a Newmarket equine influenza virus isolate.

11 . The vaccine according to claim 1 , wherein the expression vector is a poxvirus.

12 . The vaccine according to claim 11 , wherein the poxvirus is an avipox virus.

13 . The vaccine according to claim 12 , wherein the avipox virus is a canarypox virus.

14 . The vaccine according to claim 13 , wherein the canarypox virus is ALVAC.

15 . The vaccine according to claim 13 , wherein the canarypox virus is selected from CP1529, CP1533, or CP2242.

A method of vaccinating a Canidae comprising administering to said Canidae an effective amount of a vaccine comprising an effective amount of an expression vector that contains and expresses in vivo in a canine a polynucleotide encoding equine influenza antigen comprising equine influenza H3 operatively linked to a promoter and optionally to an enhancer, and a pharmaceutically or veterinarily acceptable carrier, excipient, or vehicle.

17 . The method of claim 16 , wherein the vaccine further comprises an adjuvant.

18 . The method of claim 16 , wherein the polynucleotide is isolated from an equine influenza virus isolate selected from the group consisting of an Ohio equine influenza virus isolate, a Kentucky equine influenza virus isolate, a Newmarket equine influenza virus isolate or mixtures thereof.

19 . The method of claim 17 , wherein the adjuvant is a cationic lipid containing a quaternary ammonium salt.

20 . The method of claim 16 , wherein the administering is subcutaneously or intramuscularly.
