Abstract

The present invention relates to 3-substituted-2-(arylalkyl)-1-azabicycloalkanes, methods of preparing the compounds, and methods of treatment using the compounds. The azabicycloalkanes generally are azabicycloheptanes, azabicyclooctanes, or azabicyclononanes. The compounds exhibit activity at nicotinic acetylcholine receptors (nAChRs), particularly the α7 nAChR subtype, and are useful towards modulating neurotransmission and the release of ligands involved in neurotransmission. Methods for preventing or treating conditions and disorders, including central nervous system (CNS) disorders, which are characterized by an alteration in normal neurotransmission, are also disclosed. Also disclosed are methods for treating inflammation, autoimmune disorders, pain and excess neovascularization, such as that associated with tumor growth.

Claims (239)

1. A radiolabeled compound having a structure of the formula:

wherein:

m and n individually are 1 or 2;

p is 1, 2, 3, or 4;

X is oxygen or NR′;

Y is oxygen or sulfur;

Z is NR′, a covalent bond, or a linker species, A;

A is selected from the group —CR′R″—, —CR′R″—CR′R″—, —CR′═CR′—, and —C≡C—, provided that when Z is a covalent bond or A, X must be nitrogen;

Ar is an unsubstituted or substituted, carbocyclic or 3- to 10-membered heterocyclic, monocyclic or fused polycyclic aryl group;

Cy is a pyridinyl;

the depicted wavy lines indicate variable stereochemistry; and

substituents are selected from the group consisting of alkyl, alkenyl, 3- to 10-membered heterocyclyl, cycloalkyl, 5- or 6-membered carbocyclic or heterocyclic aromatic ring which may include monocyclic or fused polycyclic rings, substituted aryl, arylalkyl wherein the aryl is a 5- or 6-membered carbocyclic or heterocyclic aromatic ring which may include monocyclic or fused polycyclic rings, substituted arylalkyl wherein the aryl is a 5- or 6-membered carbocyclic or heterocyclic aromatic ring which may include monocyclic or fused polycyclic rings, halo, —OR′, —NR′R″, —CF 3 , —CN, —NO 2 , —C≡CR′, —SR′, —N 3 , —C(═O)NR′R″, —NR′C(═O)R″, —C(═O)R′, —C(═O)OR′, —OC(═O)R′, —O(CR′R″) r C(═O)R′, —O(CR′R″) r NR″C(═O)R′, —O(CR′R″) r NR″SO 2 R′, —OC(═O)NR′R″, —NR′C(═O)OR″, —SO 2 R′, —SO 2 NR′R″, and —NR′SO 2 R″,

where R′ and R″ are individually hydrogen, straight chain or branched C 1 -C 8 alkyl, C 3-8 cycloalkyl, 3- to 10-membered heterocyclyl, aryl, or arylalkyl wherein the aryl is a 5- or 6-membered carbocyclic or heterocyclic aromatic ring which may include monocyclic or fused polycyclic rings, or R′ and R″ can combine with the nitrogen atom to which the R′ and R″ are attached to form a 3- to 10-membered cyclic functionality, and

r is an integer from 1 to 6;

or a salt thereof,

wherein the compound includes a radiolabel selected from the group consisting of 11 C, 18 F , 76 Br, 123 I, and 125 I.

2. The compound of claim 1 , wherein Cy is 3-pyridinyl.

3. The compound of claim 1 , wherein X and Y are O and Z is NR′.

4. The compound of claim 1 , wherein X is N and Y is O.

5. The compound of claim 1 , wherein the azabicyclic ring is a 1-azabicyclo[2.2.1]heptane.

6. The compound of claim 1 , wherein the azabicyclic ring is a 1-azabicyclo[3.2.1]octane.

7. The compound of claim 1 , wherein the azabicyclic ring is a 1-azabicyclo[2.2.2]octane.

8. The compound of claim 1 , wherein the azabicyclic ring is a 1-azabicyclo[3.2.2]nonane.

9. A radiolabeled compound selected from the group consisting of radiolabeled versions of:

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-phenylcarbamate,

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-(4-fluorophenyl)carbamate,

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-(4-chlorophenyl)carbamate,

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-(4-bromophenyl)carbamate,

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-(3-fluorophenyl)carbamate,

(R,R; R,S; S,R; and S,S)-2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl N-(3-chlorophenyl)carbamate,

10. A radiolabeled compound selected from the group consisting of radiolabeled versions of:

(R,R; R,S; S,R; and S,S)—N-phenyl-N′-(2-((3-pyridinyl)methyl)-1-azabicyclo[2.2.2]oct-3-yl)urea,

13. The diagnostic composition of claim 12 , wherein Cy is 3-pyridinyl.

14. The diagnostic composition of claim 12 , wherein X and Y are O, and Z is NR′.

15. The diagnostic composition of claim 12 , wherein X is N and Y is O.

16. The diagnostic composition of claim 12 , wherein the azabicyclic ring is a 1-azabicyclo[2.2.1]heptane.

17. The diagnostic composition of claim 12 , wherein the azabicyclic ring is a 1-azabicyclo[3.2.1]octane.

18. The diagnostic composition of claim 12 , wherein the azabicyclic ring is a 1-azabicyclo[2.2.2]octane.

19. The diagnostic composition of claim 12 , wherein the azabicyclic ring is a 1-azabicyclo[3.2.2]nonane.

20. A diagnostic composition comprising a compound of claim 9 and a carrier.

21. A diagnostic composition comprising a compound of claim 10 and a carrier.

22. A diagnostic composition comprising a compound of claim 11 and a carrier.
