Abstract

The present disclosure provides isolated monoclonal antibodies, particularly human monoclonal antibodies that specifically bind to Fucosyl-GM1 with high affinity. Nucleic acid molecules encoding the antibodies of this disclosure, expression vectors, host cells and methods for expressing the antibodies of this disclosure are also provided. Immunoconjugates, bispecific molecules and pharmaceutical compositions comprising the antibodies of this disclosure are also provided. This disclosure also provides methods for detecting Fucosyl-GM1, as well as methods for treating various diseases, including cancer, using anti-Fucosyl-GM1 antibodies.

Claims (19)

1. An isolated monoclonal antibody, or antigen-binding portion thereof, comprising:

(a) a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:4; and

(b) a light chain variable region comprising the amino acid sequence of SEQ ID NO:10.

2. An isolated human monoclonal antibody, or antigen-binding portion thereof, wherein the antibody, or antigen-binding portion thereof, cross-competes for binding to Fucosyl-GM1 with the antibody, or antigen-binding portion thereof, of claim 1 .

3. An isolated monoclonal antibody, or antigen-binding portion thereof, which comprises:

(a) a heavy chain variable region CDR1 domain comprising SEQ ID NO:16;

(b) a heavy chain variable region CDR2 domain comprising SEQ ID NO:22;

(c) a heavy chain variable region CDR3 domain comprising SEQ ID NO:28;

(d) a light chain variable region CDR1 domain comprising SEQ ID NO:34;

(e) a light chain variable region CDR2 domain comprising SEQ ID NO:40; and

(f) a light chain variable region CDR3 domain comprising SEQ ID NO:46.

4. An immunoconjugate comprising the antibody, or antigen-binding portion thereof, of any one of claims 2 , 3 , or 1 , linked to a therapeutic agent.

5. The immunoconjugate of claim 4 , wherein the therapeutic agent is a cytotoxin or a radioactive agent.

6. The antibody, or antigen-binding portion thereof, of any one of claims 2 , 3 , or 1 , wherein the antibody is nonfucosylated.

7. The antibody, or antigen-binding portion thereof, of claim 3 or 1 , which is a human antibody.

8. The antibody, or antigen-binding portion thereof, of claim 3 or 1 , which is a chimeric antibody or a humanized antibody.

9. The antibody, or antigen-binding portion thereof, of claim 3 or 1 , which is a full-length antibody of an IgG1 isotype.

10. The antibody, or antigen-binding portion thereof, of claim 3 or 1 , which is a full-length antibody of an IgG4 isotype.

11. A pharmaceutical composition comprising the antibody, or antigen-binding portion thereof, of any one of claims 2 , 3 , or 1 , and a pharmaceutically acceptable carrier.
