Compositions of prokaryotic phenylalanine ammonia-lyase and methods of treating cancer using compositions thereof
View Patent ↗The present invention is directed to phenylalanine ammonia-lyase (PAL) variants produced by prokaryotes, wherein such prokaryotic PAL variant has a greater phenylalanine-converting activity and/or a reduced immunogenicity as compared to a wild-type PAL. The invention provides compositions of prokaryotic PAL and biologically active fragments, mutants, variants or analogs thereof, as well as methods for the production, purification, and use of such compositions for therapeutic purposes, including the treatment of cancer.
1. A method for treating a subject having cancer, comprising administering to the subject a therapeutically effective amount of a pharmaceutical composition comprising (a) an Anabaena variabilis phenylalanine ammonia-lyase (AvPAL) variant, wherein the cysteine residue at position 503 of said AvPAL variant has been substituted by a seine residue (SEQ ID NO:9), and wherein said AvPAL variant further comprises a polyethylene glycol, and (b) a pharmaceutically acceptable carrier.

A method for treating a subject having cancer, comprising administering to the subject a therapeutically effective amount of a pharmaceutical composition comprising (a) an AvPAL variant, wherein the cysteine residue at position 565 of said AvPAL variant has been substituted by a seine residue (SEQ ID NO:10), and wherein said AvPAL variant further comprises a polyethylene glycol, and (b) a pharmaceutically acceptable carrier.
3. A method for treating a subject having cancer, comprising administering to the subject a therapeutically effective amount of a pharmaceutical composition comprising (a) an AvPAL variant, wherein the cysteine residues at positions 503 and 565 of said AvPAL variant have been substituted by serine residues (SEQ ID NO:11), and wherein said AvPAL variant further comprises a polyethylene glycol, and (b) a pharmaceutically acceptable carrier.
4. The method of claim 3 , wherein the ratio of the AvPAL variant to the polyethylene glycol is about 1:3.
5. The method of claim 3 , wherein the effective amount decreases blood, serum or plasma phenylalanine concentration of the subject to a range from below the level of detection to between 20 μM and 60 μM.
6. The method of claim 3 , wherein the pharmaceutically acceptable carrier comprises a stabilizer.
7. The method of claim 6 , wherein the stabilizer is L-phenylalanine or structural analog thereof.
8. The method of claim 6 , wherein the stabilizer is L-phenylalanine.
9. The method of claim 6 , wherein the stabilizer is trans-cinnamic acid.
10. The method of claim 6 , wherein the stabilizer is benzoic acid.
11. The method of claim 3 , wherein the cancer is a lung cancer, brain cancer, central nervous system cancer, colon cancer, prostate cancer, renal cancer, head and neck cancer, ovarian cancer, uterine cancer, bladder cancer, breast cancer, pancreatic cancer, acute myeloid leukemia, acute lymphoblastic leukemia, myeloma, pediatric cancer or a resistant cancer.
12. The method of claim 3 , wherein the cancer is metastatic melanoma.
13. The method of claim 3 , wherein the proliferation and/or survival of cells derived from the cancer is sensitive to phenylalanine restriction or depletion.
14. The method of claim 3 , further comprising administering a therapeutically effective amount of a pharmaceutical composition comprising a cancer therapeutic agent or a targeted cancer therapeutic agent.
15. The method of claim 3 , further comprising administering to the subject a protein-restricted diet.
16. The method of claim 3 , wherein the effective amount decreases blood, serum or plasma phenylalanine concentration of the subject to a range from below the level of detection to less than about 20 μM.
17. The method of claim 3 , wherein the effective amount decreases blood, serum or plasma phenylalanine concentration of the subject to a range from below the level of detection to less than about 10 μM.
18. The method of claim 3 , wherein the subject is a human.
19. A method for decreasing blood, serum or plasma phenylalanine concentration of a subject having cancer, comprising administering to the subject a therapeutically effective amount of a pharmaceutical composition comprising (a) an AvPAL variant, wherein the cysteine residues at positions 503 and 565 of said AvPAL variant have been substituted by serine residues (SEQ ID NO:11), and wherein said AvPAL variant further comprises a polyethylene glycol, and (b) a pharmaceutically acceptable carrier.
20. The method of claim 19 , wherein the ratio of the AvPAL variant to the polyethylene glycol is about 1:3.
21. The method of claim 19 , wherein the effective amount decreases blood, serum or plasma phenylalanine concentration of the subject to a range from below the level of detection to between 20 μM and 60 μM.
22. The method of claim 19 , wherein the pharmaceutically acceptable carrier comprises a stabilizer.
23. The method of claim 22 , wherein the stabilizer is L-phenylalanine or structural analog thereof.
24. The method of claim 22 , wherein the stabilizer is L-phenylalanine.
25. The method of claim 22 , wherein the stabilizer is trans-cinnamic acid.
26. The method of claim 22 , wherein the stabilizer is benzoic acid.
