Abstract

Aryl sulfone compounds of formula I and II are described and have therapeutic utility, particularly in the treatment of diabetes, obesity and related conditions and disorders:

Claims (35)

1. A compound having the formula:

and pharmaceutically acceptable salts, or stereoisomers, or prodrugs thereof, wherein:

A is a thiazole ring;

R 11 is a member selected from the group consisting of —OH, halogen, (C 1 -C 8 )alkoxy and (C 1 -C 8 )haloalkyl;

R 12 and R 13 are members independently selected from the group consisting of halogen, (C 1 -C 8 )alkyl, (C 2 -C 8 )alkenyl, (C 2 -C 8 )alkynyl, (C 1 -C 8 )alkoxy, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl and (C 3 -C 8 )cycloalkyl, wherein no more than two of R 11 , R 12 and R 13 are halogen;

R 14 is H or halogen;

the subscript n is an integer of from 0 to 4;

each R 15 is independently selected from the group consisting of H, halogen, —CN, —NO 2 , (C 1 -C 8 )alkyl, (C 2 -C 8 )alkenyl, (C 2 -C 8 )alkynyl, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl, (C 3 -C 8 )cycloalkyl, heterocycloalkyl, (C 3 -C 8 )cycloalkyl(C 1 -C 6 )alkyl, (C 3 -C 8 )cycloalkyl(C 1 -C 6 )alkoxy, heterocycloalkyl(C 1 -C 6 )alkyl, aryl, heteroaryl, heteroaryl(C 1 -C 6 )alkyl, aryl(C 1 -C 6 )alkyl, —C(O)R′, —C(O)OR′, —NR′C(O)OR″, —OR′, —OC(O)R′, —C(O)N(R′) 2 , —S(O)R″, —SO 2 R″, —SO 2 N(R′) 2 , —N(R′) 2 , —NR′C(O)R′, —X—C(O)R′, —X—C(O)OR′, —X—NR′C(O)OR″, —X—OR″, —X—OC(O)R′, —X—C(O)N(R′) 2 , —X—S(O)R″, —X—SO 2 R″, —X—SO 2 N(R′) 2 , —X—N(R′) 2 and —X—NR′C(O)R′; and optionally two of R 15 when attached to adjacent carbon atoms are combined to form a fused 5-, 6- or 7-membered ring optionally having from one to three heteroatoms as ring members, and

any fused ring portion, cycloalkyl portion, heterocycloalkyl portion, aryl or heteroaryl portion is optionally substituted with from one to four members selected from the group consisting of H, halogen, —CN, —NO 2 , (C 1 -C 8 )alkyl, (C 2 -C 8 )alkenyl, (C 2 -C g )alkynyl, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl, —C(O)OR′, —NR′C(O)OR″, —OR′, —SR′, —OC(O)R′, —C(O)N(R′) 2 , —S(O)R″, —SO 2 R″, —SO 2 N(R′) 2 , —N(R′) 2 and —NR′C(O)R′;

wherein X is a branched or straight chain (C 1 -C 8 )alkylene group;

each occurrence of R′ is independently H or an unsubstituted member selected from the group consisting of (C 1 -C 8 )alkyl, (C 2 -C g )alkenyl, (C 2 -C 8 )alkynyl, (C 1 -C 4 )alkoxy(C 1 -C 4 )alkyl, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl, (C 3 -C 8 )cycloalkyl, heterocycloalkyl, heteroaryl, aryl, (C 3 -C 8 )cycloalkyl(C 1 -C 6 )alkyl, heterocyclyl(C 1 -C 6 )alkyl, heteroaryl(C 1 -C 6 )alkyl, aryl(C 1 -C 6 )alkyl, or two R′ groups, when attached to the same nitrogen atom, can be combined with the nitrogen atom to which they are attached to form a heterocycle or heteroaryl group; and

each occurrence of R″ is independently an unsubstituted member selected from the group consisting of (C 1 -C 8 )alkyl, (C 2 -C g )alkenyl, (C 2 -C 8 )alkynyl, (C 1 -C 4 )alkoxy(C 1 -C 4 )alkyl, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl, (C 3 -C 8 )cycloalkyl, heterocycloalkyl, heteroaryl, aryl, (C 3 -C 8 )cycloalkyl(C 1 -C 6 )alkyl, heterocyclyl(C 1 -C 6 )alkyl, heteroaryl(C 1 -C 6 )alkyl or aryl(C 1 -C 6 )alkyl.

2. A compound of claim 1 , wherein R 11 is —OH; and R 12 and R 13 are each independently selected from the group consisting of (C 1 -C 4 )alkyl and (C 1 -C 4 )haloalkyl.

3. A compound of claim 1 , wherein R 11 is fluoro; and R 12 and R 13 are each independently selected from the group consisting of (C 1 -C 4 )alkyl and (C 1 -C 4 )haloalkyl.

4. A compound of claim 1 , wherein R 11 is —OH; R 12 is (C 1 -C 4 )alkyl and R 13 is (C 1 -C 4 )haloalkyl.

5. A compound of claim 3 , wherein R 11 is —OH; R 12 is —CH 3 ; and R 13 is —CF 3 .

A compound of claim 3 , wherein n is 0 to 2 and each R 15 , when present, is independently selected from the group consisting of halogen, —CN, —NO 2 , (C 1 -C 8 )alkyl, (C 2 -C 8 )alkenyl, (C 2 -C 8 )alkynyl, (C 1 -C 8 )haloalkyl, (C 2 -C 8 )hydroxyalkyl, (C 3 -C 8 )cycloalkyl, heterocycloalkyl, (C 3 -C 8 )cycloalkyl(C 1 -C 6 )alkyl, heterocycloalkyl(C 1 -C 6 )alkyl, heteroaryl(C 1 -C 6 )alkyl, aryl(C 1 -C 6 )alkyl, —C(O)R′, —C(O)OR′, —NR′C(O)OR″, —OR″, —OC(O)R′, —C(O)N(R′) 2 , —SO 2 R″, —SO 2 N(R′) 2 , —N(R′) 2 and —NR′C(O)R′.

7. A compound selected from the group consisting of:

2-[4-(2-bromo-4-methylthiazol-5-ylsulfonyl)phenyl]-1,1,1-trifluoropropan-2-ol,

2-{4-[5-ethyl-2-(piperidin-1-yl)thiazol-4-ylsulfonyl]phenyl}-1,1,1-trifluoropropan-2-ol,

1,1,1-trifluoro-2-[4-(4-methyl-2-phenylthiazol-5-ylsulfonyl)phenyl]propan-2-ol,

2-{4-[5-chloro-2-(pyrrolidin-1-yl)thiazol-4-ylsulfonyl]phenyl}-1,1,1-trifluoropropan-2-ol,

2-(4-(5-chloro-2-(2-methylpyrrolidin-1-yl)thiazol-4-ylsulfonyl)phenyl)-1,1,1-trifluoropropan-2-ol,

2-[4-(4-cyclopropyl-2-phenylthiazol-5-ylsulfonyl)phenyl]-1,1,1-trifluoropropan-2-ol,

4-cyclopropyl-5-(4-(1,1,1-trifluoro-2-hydroxypropan-2-yl)phenylsulfonyl)thiazole-2-carbonitrile,

2-[4-(4-cyclopropylthiazol-5-ylsulfonyl)phenyl]-1,1,1-trifluoropropan-2-ol,

1,1,1-trifluoro-2-{4-[2-(pyrrolidin-1-yl)-4-(trifluoromethyl)thiazol-5-ylsulfonyl]phenyl}propan-2-ol,

2-(4-(4-chloro-2-(1H-pyrazol-1-yl)thiazol-5-ylsulfonyl)phenyl)-1,1,1-trifluoropropan-2-ol,

2-[4-(4-cyclopropyl-2-isopropoxythiazol-5-ylsulfonyl)phenyl]-1,1,1-trifluoropropan-2-ol,

2-[4-(4-tert-butyl-2-phenylthiazol-5-ylsulfonyl)phenyl]-1,1,1-trifluoropropan-2-ol, and

2-(4-(4-cyclopropyl-2-(pyridin-4-yl)thiazol-5-ylsulfonyl)phenyl)-1,1,1-trifluoropropan-2-ol.

8. A pharmaceutical composition comprising the compound of claim 1 , and a pharmaceutically acceptable carrier.

9. A pharmaceutical composition comprising the compound of claim 1 , and an additional therapeutic agent.

10. A pharmaceutical composition of claim 9 , wherein the additional therapeutic agent is useful for treating a condition or disorder selected from the group consisting of diabetes and obesity.

11. A method for treating a condition or disorder selected from the group consisting of diabetes and obesity, comprising claim 1 .
