Abstract

This invention is directed to deimmunized antibodies that are useful as immunotherapeutic drugs against Human Immunodeficiency Virus (HIV) and CD4-mediated autoimmune disorders. More specifically, antibodies expressed by clones, Clone 7 containing the recombinant genes B4DIVHv1/VK1CHO#7, Clone 16 containing the recombinant genes B4DIVHv1/VK1#16, and clone 21 containing the recombinant genes B4DIVHv1/VK1#21, are derived from mouse monoclonal B4 antibody (mAb B4). The antibodies were produced by removing particular murine determinants recognized as foreign by the human immune system. These recombinant antibodies were generated by the chimerization and deimmunization of the Fv region of mouse monoclonal antibody (mAb) B4. For improved safety, the coding sequence may further be mutated to express an aglycosylated IgG 1 antibody that is unable to bind complement. The deimmunized antibodies retain the specificity of the murine mAb B4 for a receptor complex involving CD4 on the surface of the host T cells, and retain the characteristic ability of mAb B4 to neutralize primary isolates of HIV.

Claims (14)

1. A nucleotide of SEQ ID NO. 12-18, encoding an Fv antibody heavy chain variable region of SEQ ID NOS. 5, 6, 7 or 8 or an Fv antibody light chain variable region of SEQ ID NOS. 9, 10, or 11.

2. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 12, encoding an Fv antibody heavy chain variable region of SEQ ID NO. 5.

3. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 13, encoding an Fv antibody heavy chain variable region of SEQ ID NO. 6.

4. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 14, encoding an Fv antibody heavy chain variable region of SEQ ID NO. 7.

5. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 15, encoding an Fv antibody heavy chain variable region of SEQ ID NO. 8.

6. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 16, encoding an Fv antibody light chain variable region of SEQ ID NO. 9.

7. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 17, encoding an Fv antibody light chain variable region of SEQ ID NO. 10.

8. The nucleotide according to claim 1 , wherein the nucleotide is SEQ ID NO. 18, encoding an Fv antibody light chain variable region of SEQ ID NO. 11.

9. A polynucleotide which comprises coding regions for a deimmunized aglycosylated monoclonal antibody comprising a heavy chain variable region SEQ ID NOS. 5, 6, 7 or 8 and a light chain variable region of SEQ ID NOS. 9, 10, or 11, wherein Asn 297 of the Fc region is mutated.

10. The polynucleotide according to claim 9 , wherein Asn 297 is mutated to His 297 .

11. A isolated recombinant host cell which secretes a deimmunized monoclonal antibody comprising an Fv antibody heavy chain variable region of SEQ ID NOS. 5, 6, 7 or 8 and an Fv antibody light chain variable region of SEQ ID NOS. 9, 10, or 11.

A isolated recombinant host cell which secretes a human monoclonal antibody comprising six complementarity determining regions of the variable chains of murine antibody B4, wherein one or more of the complementarity determining regions SEQ ID NOS: 20, 22, 24, 26,28 and/or 30 of a deimmunized, a chimerized or a humanized monoclonal antibody.

13. The recombinant host cell according to claim 11 , wherein the deimmunized monoclonal antibody is a deimmunized aglycosylated monoclonal antibody, wherein Asn 297 of the Fc region is mutated.

14. The recombinant host cell according to claim 13 , wherein Asn 297 is mutated to His 297 .
