Abstract

Methods of treating or preventing an IL-1 related disease or condition in a mammal comprising administering an effective amount of an IL-1β binding antibody or IL-1β binding fragment thereof. An IL-1β binding antibody or IL-1β binding fragment thereof is provided comprising the amino acid sequence of SEQ ID NO:28, and related nucleic acids, vectors, cells, and compositions, and a method of preparing an affinity matured IL-1β binding polypeptide is provided. IL-1β binding antibodies or IL-1β binding fragments thereof are provided which have desirable affinity and potency.

Claims (22)

1. A method of treating an IL-1 related inflammatory disease or condition in a mammal comprising administering an effective amount of an IL-1β binding antibody or IL-1β binding fragment thereof, said antibody or fragment comprising a heavy chain variable region of SEQ ID NO: 15 and a light chain variable region of SEQ ID NO: 11, wherein the IL-1 related inflammatory disease or condition is rheumatoid arthritis, osteoarthritis, CIAS1 Associated Periodic Syndromes (CAPS), Neonatal Onset Multisystem Inflammatory Disorder (NOMID), Muckle-Wells Syndrome (MWS), Familial Cold Autoinflammatory Syndrome (FCAS), systemic onset juvenile idiopathic arthritis or Stills disease.

2. The method of claim 1 , wherein the IL-1 related disease or condition is rheumatoid arthritis.

3. The method of claim 1 , wherein the IL-1 related disease or condition is osteoarthritis.

4. The method of claim 1 , wherein the IL-1 related disease or condition is CIAS1 Associated Periodic Syndromes (CAPS).

5. The method of claim 1 , wherein the IL-1 related disease or condition is Neonatal Onset Multisystem Inflammatory Disorder (NOMID).

6. The method of claim 1 , wherein the IL-1 related disease or condition is Muckle-Wells Syndrome (MWS).

7. The method of claim 1 , wherein the IL-1 related disease or condition is Familial Cold Autoinflammatory Syndrome (FCAS).

8. The method of claim 1 , wherein the IL-1 related disease or condition is systemic onset juvenile idiopathic arthritis.

9. The method of claim 1 , wherein the IL-1 related disease or condition is Stills disease.

10. The method of claim 1 , wherein the mammal is a human.

11. The method of claim 1 , wherein the effective amount of the antibody or antibody fragment is effective to inhibit IL-1β induced expression of serum IL-6 in an animal by at least 50% as compared to the level of serum IL-6 in the absence of the antibody.

A method of treating an IL-1 related inflammatory disease or condition in a mammal comprising administering an effective amount of an IL-1β binding antibody or IL-1β binding fragment thereof, wherein the antibody or fragment binds to human IL-1β with a dissociation constant less than 1 pM, and the antibody or fragment competes with the binding of an antibody comprising the light chain variable region of SEQ ID NO: 11 and the heavy chain variable region of SEQ ID NO: 15, wherein the IL-1 related inflammatory disease or condition is rheumatoid arthritis, osteoarthritis, CIAS1 Associated Periodic Syndromes (CAPS), Neonatal Onset Multisystem Inflammatory Disorder (NOMID), Muckle-Wells Syndrome (MWS), Familial Cold Autoinflammatory Syndrome (FCAS), systemic onset juvenile idiopathic arthritis or Stills disease.

13. The method of claim 12 , wherein the IL-1 related disease or condition is rheumatoid arthritis.

14. The method of claim 12 , wherein the IL-1 related disease or condition is osteoarthritis.

15. The method of claim 12 , wherein the IL-1 related disease or condition is CIAS1 Associated Periodic Syndromes (CAPS).

16. The method of claim 12 , wherein the IL-1 related disease or condition is Neonatal Onset Multisystem Inflammatory Disorder (NOMID).

17. The method of claim 12 , wherein the IL-1 related disease or condition is Muckle-Wells Syndrome (MWS).

18. The method of claim 12 , wherein the IL-1 related disease or condition is Familial Cold Autoinflammatory Syndrome (FCAS).

19. The method of claim 12 , wherein the IL-1 related disease or condition is systemic onset juvenile idiopathic arthritis.

20. The method of claim 12 , wherein the IL-1 related disease or condition is Stills disease.

21. The method of claim 12 , wherein the mammal is a human.

22. The method of claim 12 , wherein the effective amount of the antibody or antibody fragment is effective to inhibit IL-1β induced expression of serum IL-6 in an animal by at least 50% as compared to the level of serum IL-6 in the absence of the antibody.
