Abstract

The present invention relates to compounds of formula I: and pharmaceutically acceptable salts or tautomers thereof which are inhibitors of poly(ADP-ribose)polymerase (PARP) and thus useful for the treatment of cancer, inflammatory diseases, reperfusion injuries, ischaemic conditions, stroke, renal failure, cardiovascular diseases, vascular diseases other than cardiovascular diseases, diabetes mellitus, neurodegenerative diseases, retroviral infections, retinal damage, skin senescence and UV-induced skin damage, and as chemo- or radiosensitizers for cancer treatment.

Claims (39)

1. A compound of formula II:

wherein:

the sum of a and j is 1 or 2;

b is 0, 1, 2 or 3;

t is 0,1, 2 or 3;

when t is 0 then B is CH 2 ;

when t is 1, 2 or 3 then B is CH 2 , NH or O;

w is 0, 1, 2 or 3;

X is N;

each R 1 is independently C 1-6 alkyl, haloC 1-6 alkyl, halogen or cyano;

each R 2 is independently hydroxy, halogen, cyano, nitro, C 1-6 alkyl, haloC 1-6 alkyl, C 1-6 alkoxy, haloC 1-6 alkoxy or NR a R b ;

each A is independently a direct bond, O, C═O, (C═O)NR 7 , NR 7 (C═O), (C═O)O, O(C═O), (C═S)NR 7 , NR 7 or S(O) r ;

each q is independently 0, 1, 2, 3, or 4;

r is 0, 1 or 2;

each R 6 is independently hydroxy, cyano, halogen, nitro, C 1-6 alkyl, C 2-10 alkenyl, haloC 1-6 alkyl, NR a R b or a ring which is: C 3-10 cycloalkyl; C 6-10 aryl; C 6-10 aryloxy; 4 membered saturated heterocyclic ring containing one N atom; a 5, 6 or 7 membered saturated or partially saturated heterocyclic ring containing one, two or three atoms independently selected from N, O and S; a 5 membered heteroaromatic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S, not more than one heteroatom of which is O or S; a 6 membered heteroaromatic ring containing 1, 2 or 3 nitrogen atoms; or a 7-10 membered unsaturated or partially saturated heterocyclic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S; any of which rings being optionally substituted by one or more groups independently selected from R 8 ;

R 7 is hydrogen or R 6 ;

each R 8 is independently hydroxy, oxo, cyano, halogen, nitro, C 1-6 alkyl, C 1-6 alkoxy, C 2-10 alkenyl, haloC 1-6 alkyl, haloC 1-6 alkoxy, —O(C═O)C 1-6 alkyl, —(C═O)OC 1-6 alkyl, NR a R b or a ring which is: C 3-10 cycloalkyl, C 6-10 arylC 1-6 alkyl, C 6-10 arylC 1-6 alkoxycarbonyl, C 6-10 aryl, a 5 membered heteroaromatic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S, not more than one heteroatom of which is O or S or a 6 membered heteroaromatic ring containing 1, 2 or 3 nitrogen atoms; any of which rings being optionally substituted by one or more groups independently selected from halogen, C 1-6 alkyl or haloC 1-6 alkyl;

each of R 9 and R 10 is independently hydrogen, halogen, C 1-6 alkyl or haloC 1-6 alkyl;

each of R a and R b is independently selected from hydrogen, C 1-6 alkyl, C 3-10 cycloalkyl, C 3-10 cycloalkyl, C 3-10 cycloalkylC 1-6 alkyl, C 1-6 aryl or C 6-10 arylC 1-6 alkyl;

or a pharmaceutically acceptable salt or tautomer thereof.

2. A compound of formula VII:

wherein:

the sum of a and j is 1 or 2;

b is 0, 1, 2 or 3;

i is 0 or 1;

X is N;

each R 1 is independently C 1-6 alkyl, haloC 1-6 alkyl, halogen or cyano;

each R 2 is independently hydroxy, halogen, cyano, nitro, C 1-6 alkyl, haloC 1-6 alkyl, C 1-6 alkoxy, haloC 1-6 alkoxy or NR a R b ;

R 4 is hydrogen or C 1-6 alkyl;

R 5 is a 4 membered saturated heterocyclic ring containing one N atom, a 5 or 6 membered saturated or partially saturated heterocyclic ring containing one, two or three N atoms and zero or one O atom or a 7-15 membered partially saturated or saturated heterocyclic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S; any of which rings being optionally substituted by one or more groups independently selected from A-(CR 9 R 10 ) q R 6 ;

each A is independently a direct bond, O, C═O, (C═O)NR 7 , NR 7 (C═O), (C═O)O, O(C═O), (C═S)NR 7 , NR 7 or S(O) r ;

each q is independently 0, 1, 2, 3, or 4;

r is 0, 1 or 2;

each R 6 is independently hydroxy, cyano, halogen, nitro, C 1-6 alkyl, C 2-10 alkenyl, haloC 1-6 alkyl, NR a R b or a ring which is: C 3-10 cycloalkyl; C 6-10 aryl; C 6-10 aryloxy; 4 membered saturated heterocyclic ring containing one N atom; a 5, 6 or 7 membered saturated or partially saturated heterocyclic ring containing one, two or three atoms independently selected from N, O and S; a 5 membered heteroaromatic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S, not more than one heteroatom of which is O or S; a 6 membered heteroaromatic ring containing 1, 2 or 3 nitrogen atoms; or a 7-10 membered unsaturated or partially saturated heterocyclic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S; any of which rings being optionally substituted by one or more groups independently selected from R 8 ;

R 7 is hydrogen or R 6 ;

each R 8 is independently hydroxy, oxo, cyano, halogen, nitro, C 1-6 alkyl, C 1-6 alkoxy, C 2-10 alkenyl, haloC 1-6 alkyl, haloC 1-6 alkoxy, —O(C═O)C 1-6 alkyl, —(C═O)OC 1-6 alkyl, NR a R b or a ring which is: C 3-10 cycloalkyl, C 6-10 arylC 1-6 alkyl, C 6-10 arylC 1-6 alkoxycarbonyl, C 6-10 aryl, a 5 membered heteroaromatic ring containing 1, 2, 3 or 4 heteroatoms independently selected from N, O and S, not more than one heteroatom of which is O or S or a 6 membered heteroaromatic ring containing 1, 2 or 3 nitrogen atoms; any of which rings being optionally substituted by one or more groups independently selected from halogen, C 1-6 alkyl or haloC 1-6 alkyl;

each of R 9 and R 10 is independently hydrogen, halogen, C 1-6 alkyl or haloC 1-6 alkyl;

each of R a and R b is independently selected from hydrogen, C 1-6 alkyl, C 3-10 cycloalkyl, C 3-10 cycloalkyl, C 3-10 cycloalkylC 1-6 alkyl, C 1-6 aryl or C 6-10 arylC 1-6 alkyl;

or a pharmaceutically acceptable salt or tautomer thereof.
