Abstract

Various methods of use for surface enhanced spectroscopy-active composite nanoparticles (SACN's) are provided. SACN's are submicron-sized particles or labels can be covalently or non-covalently affixed to entities of interest for the purpose of quantification, location, identification, tracking, and diagnosis. The various methods include administering a SACN nanoparticle imaging agent to a patient, scanning the patient using a system that can perform spectral imaging; and generating a spectrum or image of an internal region of the patient. A method, for diagnosing an abnormal pathology as well as a method for labeling an animal with a SACN are also provided.

Claims (17)

1. A method comprising,

a) administering a Surface Enhanced Spectroscopy-Active Nanoparticle (SACN) imaging agent to a patient,

b) performing a spectral imaging of the patient; and

c) generating a spectrum or image of an internal region of the patient by detecting said SACN,

wherein said SACN comprises a metal nanoparticle core, a raman-active reporter, and an encapsulant.

The method of claim 1 , wherein said spectral imaging is conducted by a spot scanning confocal microscope, a line scanning system, an Optical Coherence tomographic system, a system that detects only over a single wavelength band, a fluorescence imaging system comprising an excitation light source or filtered image detection.

3. The method of claim 1 , wherein said internal region of a patient is selected from the group consisting of the entire internal region of a patient, the vasculature, heart, liver, and spleen, the gastrointestinal region.

4. The method of claim 1 , wherein the system that can perform spectral imaging is a portable system.

The method of claim 1 , wherein said SACN is non-covalently associated with an imaging probe, wherein said administering comprises introducing the imaging probe into said internal region of said patient, and wherein said SACNs disassociate into the tissue of said patient or a bodily fluid of said patient.

6. A method, comprising:

a) introducing a plurality of Surface Enhanced Spectroscopy-Active Composite Nanoparticles (SACNs) targeted to a molecule involved in an abnormal pathology into a patient with the abnormal pathology, wherein the SACNs become associated to said molecule involved with the abnormal pathology; and

b) obtaining an imaging of the associated SACNs, whereby an abnormal pathology may be diagnosed,

wherein each SACN of said plurality comprises a metal nanoparticle core, a ramam-active reporter, and an encapsulant.

7. The method of claim 6 , wherein said the location of the abnormal pathology is selected from the group consisting of the gastro-intestinal tract, heart, lung, liver cervix, breast, and colon.

A method for labeling an animal with a Surface Enhanced Spectroscopy-Active Composite Nanoparticle (SACN), comprising the steps of: introducing a SACN into an animal; detecting the SACN or labeled animal, wherein said introducing is selected from the group consisting of subcutaneous implantation and intravenous introduction; and wherein said SACN comprises a metal nanoparticle core, a raman-active reporter, and an encapsulant,

wherein said introducing is selected from the group consisting of subcutaneous implantation and intravenous introduction,

wherein said SACN comprises a metal nanoparticle core, a raman-active reporter, and an encapsulant.
