Abstract

The present invention provides protein and gene expression profiles indicative of whether a patient afflicted with non-small cell lung cancer is likely to be responsive to treatment with a therapeutic compound that is a EGFR-TK inhibitor. By identifying such responsiveness, a treatment provider may determine in advance those patients who would benefit from such treatment, as well as identify alternative therapies for non-responders. The present invention further provide methods of using the gene and protein expression profiles, and assays for identifying the presence of a gene or protein expression profile in a patient sample.

Claims (7)

1. A method of determining if a patient diagnosed with non-small cell lung cancer (NSCLC) is a responder to treatment with the EGFR-TK inhibitor gefitinib comprising:

a. obtaining lung cancer tissue from the patient diagnosed with non-small cell lung cancer;

b. determining the level of expression in the lung cancer tissue or lung cancer cells of the following proteins: p70S6K comprising the amino acid sequence of SEQ ID NO. 17, phopho-S6 comprising the amino acid sequence of SEQ ID NO. 18, phospho-AKT comprising the amino acid sequence of SEQ ID NO. 19, phospho-mTOR comprising the amino acid sequence of SEQ ID NO. 20, phospho-pTEN comprising the amino acid sequence of SEQ ID NO. 21, phospho-MEK comprising the amino acid sequence of SEQ ID NO. 22, phospho-MAPK comprising the amino acid sequence of SEQ ID NO. 23, phospho-IGFR/InR comprising the amino acid sequence of SEQ ID NO. 24, EGFR comprising the amino acid sequence of SEQ ID NO. 25, phospho-HER2(ErbB2) comprising the amino acid sequence of SEQ ID NO. 26, phospho-ER comprising the amino acid sequence of SEQ ID NO. 27, phospho-AR comprising the amino acid sequence of SEQ ID NO. 28, AIK comprising the amino acid sequence of SEQ ID NO. 29, osteopontin comprising the amino acid sequence of SEQ ID NO. 30, MMP11 comprising the amino acid sequence of SEQ ID NO. 31 and GFAP comprising the amino acid sequence of SEQ ID NO. 32; and

c. determining that the patient is a responder to treatment with an EGFR-TK inhibitor if the levels of the p70S6K, phospho-S6, phospho-mTOR, phospho-pTEN, EGFR, phospho-ER, phospho-AR, AIK, osteopontin, MMP11 and GFAP are elevated in the lung cancer tissue or lung cancer cells as compared to the corresponding levels in normal lung tissue or normal lung cells, and the levels of phospho-MEK, phospho-MAPK, phospho-IGFR/InR, and phospho-HER2(ErbB2) are lower in the lung cancer tissue or lung cancer cells as compared to the corresponding levels in the normal lung tissue or normal lung cells.

2. The method of claim 1 further comprising determining the level of expression of at least one reference protein in said lung cancer tissue or lung cancer cells and in said normal lung tissue or normal lung cells.

3. The method of claim 1 wherein step (b) is performed using immunohistochemistry.

4. The method of claim 2 wherein the reference protein is selected from the group consisting of: ACTB, GAPD, GUSB, RPLP0 and TRFC.
