Abstract

This invention concerns 5-(hydroxymethylene and aminomethylene)pyrimidine derivatives having HIV (Human Immunodeficiency Virus) replication inhibiting properties, to the preparation thereof and to pharmaceutical compositions comprising these compounds.

Claims (40)

1. A compound of formula

a stereochemically isomeric form thereof, a pharmaceutically acceptable addition salt thereof, or a N-oxide thereof, wherein

each R 1 independently is hydrogen; aryl; formyl; C 1-6 alkylcarbonyl; C 1-6 alkyl; C 1-6 alkyloxycarbonyl;

R 2 , R 3 , R 7 and R 8 independently are hydrogen; hydroxy; halo; C 3-7 cycloalkyl; C 1-6 alkyloxy; carboxyl; C 1-6 alkyloxycarbonyl; cyano; nitro; amino; mono- or di(C 1-6 alkyl)-amino; polyhaloC 1-6 alkyl; polyhaloC 1-6 alkyloxy; —C(═O)R 10 ; C 1-6 alkyl optionally substituted with halo, cyano or —C(═O)R 10 ; C 2-6 alkenyl optionally substituted with halo, cyano or —C(═O)R 10 ; C 2-6 alkynyl optionally substituted with halo, cyano or —C(═O)R 10 ;

R 4 and R 9 independently are hydroxy; halo; C 3-7 cycloalkyl; C 1-6 alkyloxy; carboxyl; C 1-6 alkyloxycarbonyl; formyl; cyano; nitro; amino; mono- or di(C 1-6 alkyl)amino; polyhaloC 1-6 alkyl; polyhaloC 1-6 alkyloxy; —C(═O)R 10 ; cyano; —S(═O) r R 10 ; —NH—S(═O) r R 10 ; —NHC(═O)H; —C(═O)NHNH 2 ; —NHC(═O)R 10 ; Het; —Y-Het; C 1-12 alkyl optionally substituted with halo, cyano, amino, mono- or di(C 1-6 alkyl)-amino, —C(═O)—R 10 , Het or with C 1-6 alkyloxy; C 2-12 alkenyl optionally substituted with halo, cyano, amino, mono- or di(C 1-6 alkyl)amino, —C(═O)—R 10 , Het or with C 1-6 alkyloxy; C 2-12 alkynyl optionally substituted with halo, cyano, amino, mono- and di(C 1-6 alkyl)amino, —C(═O)—R 10 , Het or with C 1-6 alkyloxy;

R 5 is C 1-6 alkyl optionally substituted with Ar or with Het; C 2-6 alkenyl optionally substituted with Ar or with Het; C 2-6 alkynyl optionally substituted with Ar or with Het; C 3-7 cycloalkyl; Ar; Het;

R 6 is H, C 1-6 alkyl, Het;

Y is —OR 11 , —NR 12 R 13 ;

each R 10 independently is C 1-6 alkyl, amino, mono- or di(C 1-6 alkyl)amino or polyhalo-C 1-6 alkyl;

R 11 is hydrogen or C 1-6 alkyl optionally substituted with hydroxy, C 1-6 alkyloxy, phenyl or pyridyl;

R 12 is hydrogen or C 1-6 alkyl;

R 13 is hydrogen or C 1-6 alkyl; or R 12 and R 13 taken together with the nitrogen atom to which they are substituted form pyrrolidinyl; piperidinyl; morpholinyl; piperazinyl; piperazinyl optionally substituted with C 1-6 alkyl or C 1-6 alkylcarbonyl; imidazolyl;

X is —NR 1 —, —O—, —C(═O)—, —CH 2 —, —CHOH—, —S—, —S(═O) r —;

each Y independently is —NR 1 —, —O—, —C(═O)—, —S—, —S(═O) r —;

each r independently is 1 or 2;

Het is a 5- or 6-membered completely unsaturated ring wherein one, two, three or four ring members are hetero atoms, each independently selected from nitrogen, oxygen and sulfur, and wherein the remaining ring members are carbon atoms; and, where possible, any nitrogen ring member may optionally be substituted with C 1-6 alkyl; which 5- or 6-membered ring may optionally be annelated with a benzene, thiophene or pyridine ring; and wherein any ring carbon atom, including any carbon of an optionally annelated benzene, thiophene or pyridine ring, may, each independently, optionally be substituted with a substituent selected from halo, hydroxy, mercapto, cyano, C 1-6 alkyl, hydroxyC 1-4 alkyl, carboxyC 1-4 alkyl, C 1-4 alkyloxyC 1-4 alkyl, C 1-4 alkyloxycarbonylC 1-4 alkyl, cyanoC 1-4 alkyl, mono- and di(C 1-4 alkyl)aminoC 1-4 alkyl, Het 1 C 1-4 alkyl, arylC 1-4 alkyl, polyhaloC 1-4 alkyl, C 3-7 cycloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl-C 2-4 alkenyl, C 1-4 alkyloxy, —OCONH 2 , polyhaloC 1-4 alkyloxy, aryloxy, amino, mono- and di-C 1-4 alkylamino, pyrrolidinyl, piperidinyl, morpholinyl, piperazinyl, 4-C 1-6 alkylpiperazinyl, C 1-4 alkylcarbonylamino, formyl, C 1-4 alkylcarbonyl, C 1-4 alkyloxy-carbonyl, aminocarbonyl, mono- and diC 1-4 alkylaminocarbonyl, aryl, Het 1 ;

Het 1 is pyridyl, thienyl, furanyl, oxazolyl, isoxazolyl, imidazolyl, pyrazolyl, thiazolyl, thiadiazolyl, oxadiazolyl quinolinyl, benzothienyl, benzofuranyl; which each may be optionally substituted with one or two C 1-4 alkyl radicals;

each aryl independently is phenyl or phenyl substituted with one, two, three, four or five substituents each independently selected from halo, hydroxy, mercapto, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, hydroxyC 1-6 alkyl, aminoC 1-6 alkyl, mono and di(C 1-6 alkyl)-amino-C 1-6 alkyl, C 1-6 alkylcarbonyl, C 3-7 cycloalkyl, C 1-6 alkyloxy, phenylC 1-6 alkyloxy, C 1-6 alkyloxycarbonyl, aminosulfonyl, C 1-6 alkylthio, cyano, nitro, polyhaloC 1-6 alkyl, polyhaloC 1-6 alkyloxy, aminocarbonyl, phenyl, Het and —Y-Het.

2. A compound according to claim 1 wherein the compound is of formula

wherein X, Y, R 1 , R 4 , R 5 , R 6 , R 7 , R 8 and R 9 are as defined in claim 1 .

3. A compound according to claim 1 wherein R 1 is hydrogen.

4. A compound according to claim 1 wherein R 2 , R 3 , R 7 and R 8 independently are hydrogen; hydroxy; halo; C 1-6 alkyl; C 1-6 alkyloxy; cyano; amino; mono-or di(C 1-6 alkyl)amino; polyhaloC 1-6 alkyl.

5. A compound according to claim 1 wherein R 4 and R 9 independently are cyano; C 1-6 alkyl substituted with cyano; C 2-6 alkenyl substituted with cyano.

6. A compound according to claim 1 wherein R 5 is C 1-6 alkyl substituted with Ar or with Het, C 2-6 alkenyl, C 2-6 alkynyl optionally substituted with Het,

C 3-7 cycloalkyl, Ar, Het;

R 6 is H, C 1-6 alkyl, Het;

Y is OR 11 , NR 12 R 13 ;

R 11 is hydrogen or C 1-6 alkyl optionally substituted with hydroxy, C 1-6 alkyloxy, phenyl or pyridyl;

R 12 is hydrogen or C 1-6 alkyl;

R 13 is hydrogen or C 1-6 alkyl; or R 12 and R 13 taken together with the nitrogen atom to which they are substituted form imidazolyl.

7. A compound according to claim 1 wherein R 5 is C 2-6 alkenyl,

C 2-6 alkynyl substituted with Het, Ar, Het;

R 6 is H, C 1-6 alkyl, Het;

Y is OR 11 , NR 12 R 13 ;

R 11 is hydrogen or C 1-6 alkyl optionally substituted with hydroxy, C 1-6 alkyloxy;

R 12 is hydrogen or C 1-6 alkyl;

R 13 is hydrogen.

8. A compound according to claim 1 wherein each Het independently is pyridyl, thienyl, thiazolyl, furanyl, each of which may be optionally substituted with a radical selected from C 1-6 alkyl.

9. A compound according to claim 1 wherein each aryl independently may be phenyl optionally substituted with C 1-6 alkyl, amino, mono- or diC 1-6 alkyl-amino, C 1-6 alkyloxy, aminosulfonyl, or thiadiazolyl.

10. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and as active ingredient a therapeutically effective amount of a compound as claimed in claim 1 .
