Methods and compositions for targeting HEPSIN
Anti-HEPSIN monoclonal antibodies, and methods for using the antibodies, are provided.
1. An isolated antibody comprising (a) the heavy chain hypervariable region (HC-HVR) 1, HC-HVR2, and HC-HVR3 sequences, and (b) the light chain hypervariable region (LC-HVR) 1, LC-HVR2 and LC-HVR3 sequences of an antibody produced by hybridoma cell line deposited at American Type Culture Collection (ATCC) under Accession No. PTA-7469, PTA-7470, PTA-7471, PTA-7472 or PTA-7473, wherein said isolated antibody binds native human sequence HEPSIN polypeptide.
2. An isolated antibody comprising (a) the heavy chain variable domain sequence and/or (b) the light chain variable domain sequence of an antibody produced by hybridoma cell line deposited at American Type Culture Collection (ATCC) under Accession No. PTA-7469, PTA-7470, PTA-7471, PTA-7472 or PTA-7473, wherein said isolated antibody binds native human sequence HEPSIN polypeptide.
3. The antibody of claim 1 , wherein the full length IgG form of the antibody specifically binds native human sequence HEPSIN polypeptide with a EC 50 binding affinity of 150 nM or less.
4. The antibody of claim 3 wherein the EC 50 binding affinity is 120 nM or less.
5. The antibody of claim 3 wherein the EC 50 binding affinity is 100 nM or less.
6. The antibody of claim 3 wherein the EC 50 binding affinity is 75 nM or less.
7. The antibody of claim 3 wherein the EC 50 binding affinity is 50 nM or less.
8. The antibody of claim 1 , wherein the antibody specifically binds native sequence HEPSIN polypeptide that is in native, partially denatured or denatured form.
9. The antibody of claim 1 , wherein the antibody binds soluble or cell-attached native sequence HEPSIN polypeptide.
10. An anti-HEPSIN antibody encoded by the antibody coding sequence of hybridoma cell line deposited at the American Type Culture Collection (ATCC) under Accession No. PTA-7469, PTA-7470, PTA-7471, PTA-7472 or PTA-7473.
11. A nucleic acid molecule comprising a nucleic acid sequence encoding the antibody of claim 1 .
12. A host cell comprising the nucleic acid molecule of claim 11 .
13. A cell line capable of producing the antibody of claim 1 .
14. A method of producing the antibody of claim 1 , comprising culturing a host cell comprising a nucleic acid encoding the antibody under conditions wherein the antibody is produced.
15. A composition comprising the antibody of claim 1 , and a carrier.
16. A method of detecting presence of native sequence HEPSIN polypeptide in a sample, comprising contacting the sample with an antibody of claim 1 and diagnosing presence of native sequence HEPSIN polypeptide by detecting binding of the antibody to HEPSIN polypeptide, wherein detection of binding indicates the presence of native sequence HEPSIN polypeptide.
