Abstract

The invention relates to a process for the culturing of cells, preferably E1-immortalized HER cells, more preferably PER.C6 cells in a reactor in suspension in a cell culture medium, wherein the cells produce a biological substance, preferably an antibody, wherein at least one cell culture medium component is fed to the cell culture and wherein the cell culture comprising the cells, the biological substance and cell culture medium is circulated over a separation system and wherein the separation system separates the biological substance from substances having a lower molecular weight than the biological substance and wherein the biological substance is retained in or fed back into the reactor. Preferably part of the substances of lower molecular weight is continuously removed from the cell culture.

Claims (18)

1. Process for culturing of cells in a reactor in suspension in a cell culture medium, comprising:

circulating a cell culture through a separation system comprising a filter, wherein the cell culture comprises eukaryotic cells, a biological substance produced by the cells, and a cell culture medium, wherein at least one cell culture medium component is fed to the cell culture; wherein the cell culture is circulated through the filter in an alternating tangential flow; and wherein the separation system separates the biological substance from substances having a lower molecular weight than the biological substance;

retaining or feeding back the biological substance into the reactor, wherein the cells are cultured to have a viable cell density of at least 10×10 6 cells/mL; and

harvesting the biological substance from the cells and/or from the cell culture;

wherein the biological substance is present in the cell culture at a concentration of at least 5 g/L prior to the harvesting.

2. Process according to claim 1 , wherein the cell culture is continuously circulated through the separation system, and wherein part of the substances of lower molecular weight is continuously removed from the cell culture.

3. Process according to claim 1 , wherein the cells are mammalian cells.

4. Process according to claim 1 , wherein the biological substance is a protein.

5. Process according to claim 1 , wherein the separation system is a hollow fiber filter.

6. Process according to claim 1 wherein cell culture is removed at least once from the reactor and cell culture medium is added to the reactor to compensate for the cell culture removal.

7. Process according to claim 1 , wherein cell culture conditions are chosen such that cell growth rate and/or specific productivity of the cells is not limited.

8. Process according to claim 7 , wherein the cell culture conditions are chosen such that the concentration of at least one of the components of the cell culture medium remains constant.

9. Process according to claim 1 , wherein at least the depleted nutrients are supplemented by feeding these nutrients to the reactor.

10. The process of claim 1 , wherein the biological substance is present in the cell culture at a concentration of at least 10 g/L prior to the harvesting.

11. The process of claim 10 , wherein the cells are cultured to have a viable cell density of at least 50×10 6 cells/mL.

12. The process of claim 4 , wherein the harvesting comprises harvesting the protein from the flow which contents are kept in or fed back into the reactor, from cell culture that is removed from the reactor, or both.

13. The process of claim 12 , wherein the harvesting comprises subjecting the flow which contents are kept in or fed back into the reactor, the cell culture that is removed from the reactor, or both to affinity chromatography and/or ion exchange chromatography.

14. The process of claim 12 , wherein the protein is present in the cell culture at a concentration of at least 10 g/L prior to the harvesting.
