Abstract

The invention generally relates to compositions and methods for transporting substances across mucosal barriers. The invention also relates to methods of making and using such substances.

Claims (33)

1 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface that reduce mucoadhesion of the particle, wherein said particle diffuses through human cervicovaginal mucus at a diffusivity that is greater than 1/1000 the diffusivity that the particle diffuses through water at a time scale of 1 s.

2 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface, whereby the particle moves in human cervicovaginal mucus at a diffusivity of more than 4×10 −2 μm 2 /s at a time scale of 1 s.

3 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface that reduce mucoadhesion of the particle, wherein the particle has a zeta potential lower than 10 mV.

4 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface that reduce mucoadhesion of the particle, wherein the mass of the surface-altering moiety makes up at least 1/3400 of the mass of the particle.

5 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface that reduce mucoadhesion of the particle, wherein the particle adsorbs less than 10% of the amount of fluorescently labeled avidin that is adsorbed by a corresponding particle lacking the surface-altering moiety, as calculated by average maximum fluorescent intensity.

6 . A particle comprising an outer surface and one or more surface-altering moieties disposed on the outer surface that reduce mucoadhesion of the particle, wherein the surface-altering moiety is present on the outer surface at a density of greater than 0.01 units per nanometer squared.

12 . A particle of claim 1 , wherein the particle comprises a core.

13 . A particle of claim 12 , wherein the core comprises a polymer.

14 . A particle of claim 13 , wherein the polymer comprises surface-altering moieties disposed on the outer surface.

16 . A particle of claim 1 , wherein the particle is a liposome, metal or metal oxide, or a quantum dot.

17 . A particle of claim 1 , wherein one or more bioactive agents are associated with the particle.

22 . The particle of claim 1 , wherein said particle is larger than 1 nm in diameter.

26 . The particle of claim 17 , wherein the bioactive agent is an imaging agent or a therapeutic agent.

30 . The particle of claim 1 , further comprising one or more targeting moieties.

31 . The particle of claim 1 , wherein the one or more surface-altering moieties enhance hydrophilicity of the surface of the particle.

34 . A pharmaceutical composition comprising the particle of claim 1 and one or more pharmaceutically acceptable carriers.

35 . An inhaler comprising the particle of claim 1 .

36 . A method for treating, preventing, or diagnosing a condition in a patient, comprising administering to the patient one or more particles of claim 1 .

42 . A composition comprising a plurality of particles, wherein at least 10% of the particles in the composition are particles as defined in claim 1 .

60 . A method for decreasing the mucoadhesion of a substance, comprising identifying a substance in need of increased diffusivity through mucus, modifying the substance with a surface-altering moiety, and formulating the modified substance in a pharmaceutical preparation suitable for topical administration to a mucosal surface.

68 . A pharmaceutical preparation suitable for topical administration to a mucus membrane, comprising a bioactive agent associated with a surface-altering moiety.

An enveloped virus comprising a surface-altering moiety, wherein said virus diffuses through human cervicovaginal mucus at a diffusivity at a time scale of 1 second that is more than 20-fold greater than the diffusivity at which a corresponding virus lacking the surface-altering moiety diffuses through human cervicovaginal mucus.
