Abstract

The invention discloses the nanoparticles composed of chitosan, poly-glutamic acid, and at least one protein drug or bioactive agent characterized with a positive surface charge and their enhanced permeability for paracellular protein drug and bioactive agent delivery.

Claims (20)

1. A method of enhancing permeability of at least one bioactive agent through epithelial cells in an animal subject comprising administering into the animal subject bioactive nanoparticles with a dosage effective for enhancing said permeability, wherein said bioactive nanoparticles consist of positively charged chitosan, a negatively charged substrate, inactive ingredients, and said at least one bioactive agent, wherein a shell portion of said nanoparticles is dominated by said chitosan, and wherein a core portion contains said negatively charged substrate that is at least partially neutralized with a portion of said chitosan.

2. The method of claim 1 , wherein said at least one bioactive agent is an interleukin.

3. The method of claim 1 , wherein said at least one bioactive agent is an interferon.

4. The method of claim 1 , wherein said at least one bioactive agent is a tumor necrosis factor.

5. The method of claim 1 , wherein said at least one bioactive agent is a monoclonal antibody.

6. The method of claim 1 , wherein said at least one bioactive agent is a glucagon.

7. The method of claim 1 , wherein said nanoparticles are in a spheroidal shape having a mean particle size of about 50 nanometers to 400 nanometers.

8. The method of claim 1 , wherein said bioactive nanoparticles are treated with a coating.

9. The method of claim 1 , wherein said method comprises administering said nanoparticles into a blood vessel of the animal subject.

10. The method of claim 9 , wherein said at least one bioactive agent comprises an antagonist for treating Alzheimer's diseases.

11. The method of claim 1 , wherein said bioactive nanoparticles are encapsulated in capsules.

12. The method of claim 11 , wherein said capsules are treated with an enteric coating.

13. The method of claim 11 , wherein said bioactive nanoparticles are freeze-dried prior to being loaded in said capsules.

14. The method of claim 13 , wherein said bioactive nanoparticles are mixed with trehalose prior to being freeze-dried.

15. The method of claim 1 , wherein said method of administering said bioactive nanoparticles is via an oral route.

16. The method of claim 1 , wherein said at least one bioactive agent is a vaccine.

17. The method of claim 1 , wherein said chitosan is N-trimethyl chitosan or a chitosan derivative.

18. The method of claim 1 , wherein one of said inactive ingredients comprises at least one transport enhancer.

19. The method of claim 1 , wherein one of said inactive ingredients comprises magnesium sulfate or TPP.

20. The method of claim 1 , wherein said at least one bioactive agent is a non-viral gene.
