Abstract

Efficient sequence specific gene silencing is possible through the use of siRNA technology. By selecting particular siRNAs by rational design, one can maximize the generation of an effective gene silencing reagent, as well as methods for silencing genes. Methods, compositions, and kits generated through rational design of siRNAs are disclosed including those directed to CTGF.

Claims (20)

1. A method for inhibiting the expression of a gene in a cell, the method comprising introducing into the cell in vitro a double stranded ribonucleic acid, wherein said double stranded ribonucleic acid consists of (i) a sense region and an antisense region, wherein said sense region and said antisense region together form a duplex region consisting of 19-30 base pairs and said antisense region comprises a sequence that is the complement of SEQ ID NO: 484 and (ii) either zero overhang regions or at least one overhang region, wherein each overhang region comprises six of fewer bases, wherein the gene is connective tissue growth factor.

2. The method according to claim 1 , wherein said duplex region has 19-25 base pairs.

3. The method according to claim 2 , wherein said duplex region has 19 base pairs.

4. The method according to claim 1 , wherein the sense region comprises a sequence that is the same as SEQ ID NO: 484.

5. The method according to claim 2 , wherein the sense region comprises a sequence that is the same as SEQ ID NO: 484.

6. The method according to claim 3 , wherein the sense region comprises a sequence that is the same as SEQ ID NO: 484.

7. The method according to claim 1 , wherein there are no overhang regions.

8. The method according to claim 2 , wherein there are no overhang regions.

9. The method according to claim 3 , wherein there are no overhang regions.

10. The method according to claim 1 , wherein there is one overhang region.

11. The method according to claim 2 , wherein there is one overhang region.

12. The method according to claim 3 , wherein there is one overhang region.

13. The method according to claim 1 , wherein there are two overhang regions.

14. The method according to claim 2 , wherein there are two overhang regions.

15. The method according to claim 3 , wherein there are two overhang regions.

16. The method according to claim 1 , wherein the cell is a human cell.

17. The method according to claim 2 , wherein the cell is a human cell.

18. The method according to claim 3 , wherein the cell is a human cell.

19. The method according to claim 1 , wherein said introducing is via transfection.

20. The method according to claim 1 , wherein said introducing is via passive uptake.
