Abstract

An apparatus may include a flow cell for a surface plasmon resonance test apparatus and a fluid delivery system operatively coupled to the flow cell and configured to deliver a plurality of fluids to the flow cell substantially continuously. The fluid delivery system may include two pumps for pumping the fluids.

Claims (57)

1. A surface plasmon resonance test system, comprising:

a microarray including a plurality of test patches configured for specific association with one or more molecules or biomolecules;

a flow cell configured to flow fluids, at least some of which potentially contain the one or more molecules or biomolecules across the microarray; and

a fluid delivery system including:

at least one holding coil;

a first pump configured to push or pull fluids into the holding coil; and

second pump configured to push fluids through the flow cell.

2. The surface plasmon resonance test system of claim 1 , further comprising:

a surface plasmon resonance coupling surface adjacent to the microarray of test patches; and

an optical subsystem configured to illuminate the surface plasmon resonance coupling surface and detect changes in light reflected from locations corresponding to the test patches resulting from association and dissociation of the molecules or biomolecules.

3. The surface plasmon resonance test system of claim 1 , wherein the fluid delivery system further includes an injection valve including a first port coupled to receive fluid pushed by the first pump;

a second port coupled to receive fluid pushed by the second pump;

a third port coupled to the flow cell; and

fourth and fifth ports coupled to the holding coil.

4. The surface plasmon resonance test system of claim 3 , wherein the injection valve is configured to receive a control signal and to responsively switch between two positions;

wherein the first position includes first fluid coupling between the second pump, the holding coil, and the flow cell; and

wherein the second position includes a second fluid coupling between the second pump and the flow cell and a third fluid coupling between the first pump and the holding coil.

5. The surface plasmon resonance test system of claim 4 , wherein the injection valve includes a sixth port coupled to a waste receptacle;

wherein the first position includes a fourth fluid coupling between the first pump and the waste receptacle; and

wherein the third fluid coupling includes a coupling between the first pump, the holding coil, and the waste receptacle.

6. The surface plasmon resonance test system of claim 1 , wherein the fluid delivery system includes tubing having hydrophilic surfaces.

7. The surface plasmon resonance test system of claim 6 , wherein the tubing includes silica-coated surfaces or tubing treated with a surfactant.

8. The surface plasmon resonance test system of claim 1 , wherein the flow cell has one or more hydrophilic surfaces.

9. The surface plasmon resonance test system of claim 1 , wherein the second pump is configured to push fluids from the holding coil through the flow cell.

10. The surface plasmon resonance test system of claim 1 , wherein the fluid delivery system further includes:

an injection valve configured to couple either the first or the second pump to the holding coil.

11. The surface plasmon resonance test system of claim 10 , wherein the fluid delivery system further includes:

a selection valve configured to selectively couple the first pump to one of a plurality of fluid sources or the injection valve.

12. The surface plasmon resonance test system of claim 11 , wherein at least one of the plurality of fluid sources includes an autosampler.

13. The surface plasmon resonance test system of claim 12 , further comprising:

a controller operatively coupled to the selection valve.

14. The surface plasmon resonance test system of claim 11 , further comprising:

a controller operatively coupled to the first pump, the second pump, and the injection valve.

15. The surface plasmon resonance test system of claim 1 , wherein the fluids include at least one analyte, and further comprising:

a controller configured to prevented the first and second pumps from being contaminated by the analyte.

16. The surface plasmon resonance test system of claim 1 , wherein the fluid delivery system further includes:

a plurality of fluid reservoirs; and

wherein the fluids include a plurality of analytes, a buffer solution, and at least one regeneration fluid.

17. The surface plasmon resonance test system of claim 16 , further comprising:

a surface plasmon resonance coupling surface adjacent to the microarray of test patches;

an optical subsystem configured to illuminate the surface plasmon resonance coupling surface and detect changes in light reflected from locations corresponding to the test patches resulting from association and dissociation of the chemicals or biochemicals;

wherein the fluid delivery system includes at least on regeneration fluid reservoir; and

a controller configured to measure a parameter corresponding to the microarray of test patches and automatically determine when the at least one regeneration fluid is delivered to the flow cell.

18. A method for analyzing at least one molecule or biomolecule in a fluid, comprising:

pumping a first fluid including a buffer through a flow cell of a surface plasmon resonance test apparatus;

simultaneously loading a holding coil with a fluid analyte containing at least one molecule or biomolecule for analysis; and

after the holding coil is loaded, actuating a valve to a first position to stop the flow of the first fluid while substantially simultaneously starting the flow of the fluid analyte from the holding coil through the flow cell.

19. The method for analyzing at least one molecule or biomolecule in a fluid of claim 18 , further comprising:

while the valve is in the first position, flushing fluid analyte from tubing other than the holding coil to a waste receptacle.

20. The method for analyzing at least one molecule or biomolecule in a fluid of claim 18 , further comprising:

after at least a portion of the fluid analyte from the holding coil is pumped through the flow cell, actuating the valve to a second position to stop the flow of the fluid analyte through the flow cell while substantially simultaneously starting to pump the first fluid through the flow cell.

21. The method for analyzing at least one molecule or biomolecule in a fluid of claim 18 , further comprising:

sensing a parameter corresponding to a microarray coupled to the flow cell; and

pumping a regeneration fluid through the flow cell when the value of the parameter corresponds to a regeneration indication.

22. The method for analyzing at least one molecule or biomolecule in a fluid of claim 18 , wherein substantially simultaneously switching flow includes no more than about 100 mS delay between the flow of the first fluid and the flow of the fluid analyte.

23. The method for analyzing at least one molecule or biomolecule in a fluid of claim 22 , wherein substantially simultaneously switching flow includes no more than about 10 mS delay between the flow of the first fluid and the flow of the fluid analyte.

24. The method for analyzing at least one molecule or biomolecule in a fluid of claim 23 , wherein substantially simultaneously switching flow includes no more than about 1 mS delay between the flow of the first fluid and the flow of the fluid analyte.
