Abstract

The present invention relates to canine COX-1 and COX-2 proteins; to canine COX-1 and COX-2 nucleic acid molecules, including those that encode such COX-1 and COX-2 proteins, respectively; to antibodies raised against such proteins; and to compounds that inhibit the activity of such proteins. The present invention also includes methods to obtain such proteins, nucleic acid molecules, antibodies, and inhibitory compounds. The present invention also includes therapeutic compositions comprising such inhibitory compounds, particularly those that specifically inhibit COX-2 activity, as well as the use of such therapeutic compositions to treat animals.

Claims (30)

1. An isolated protein encoded by a nucleic acid sequence that hybridizes with a polynucleotide sequence consisting of SEQ ID NO:3, SEQ ID NO:6 or SEQ ID NO:8, under conditions comprising (i) hybridizing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 37° C. and (ii) washing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 81° C., wherein said isolated protein has cyclooxygenase activity.

2. The isolated protein of claim 1 , wherein said nucleic acid sequence is at least 95% identical to SEQ ID NO:1, SEQ ID NO:4 or SEQ ID NO:7.

3. The isolated protein of claim 1 , wherein said nucleic acid sequence is at least 98% identical to SEQ ID NO:1, SEQ ID NO:4 or SEQ ID NO:7.

4. The isolated protein of claim 1 , wherein said nucleic acid sequence is identical to SEQ ID NO:1, SEQ ID NO:4 or SEQ ID NO:7.

5. An isolated protein selected from the group consisting of:

(a) an isolated protein comprising an amino acid sequence at least 95% identical to SEQ ID NO:2 or SEQ ID NO:5; and

(b) an isolated protein comprising at least 115 contiguous amino acids from SEQ ID NO:5;

wherein said isolated protein has cyclooxygenase activity.

6. The isolated protein of claim 5 , wherein said protein comprises at least 300 contiguous amino acids from SEQ ID NO:5.

7. The isolated protein of claim 5 , wherein said amino acid sequence is at least 98% identical to SEQ ID NO:2 or SEQ ID NO:5.

8. The isolated protein of claim 5 , wherein said amino acid sequence is identical to SEQ ID NO:2 or SEQ ID NO:5.

9. A method to detect an inhibitor of cyclooxygenase activity, said method comprising:

(a) contacting an isolated protein with a putative inhibitory compound, wherein said isolated protein is encoded by a nucleic acid sequence that hybridizes with a polynucleotide sequence consisting of SEQ ID NO:3, SEQ ID NO:6 or SEQ ID NO:8, under conditions comprising (i) hybridizing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 37° C. and (ii) washing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 81° C., wherein said isolated protein has cyclooxygenase activity; and

(b) determining if said putative inhibitory compound inhibits said cyclooxygenase activity.

10. The method of claim 9 , wherein said isolated protein is selected from the group consisting of:

(a) an isolated protein comprising an amino acid sequence at least 95% identical to SEQ ID NO:2 or SEQ ID NO:5; and

(b) an isolated protein comprising at least 115 contiguous amino acids from SEQ ID NO:5.

11. The method of claim 9 , wherein said isolated protein is selected from the group consisting of:

(a) an isolated protein comprising an amino acid sequence at least 98% identical to SEQ ID NO:2 or SEQ ID NO:5; and

(b) an isolated protein comprising at least 300 contiguous amino acids from SEQ ID NO:5.

12. The method of claim 9 , wherein said isolated protein comprises SEQ ID NO:2 or SEQ ID NO:5.

13. A kit for performing the method of claim 10 , said kit comprising:

(a) an isolated protein having cyclooxygenase activity, wherein said protein is encoded by a nucleic acid sequence that hybridizes with a polynucleotide sequence consisting of SEQ ID NO:3, SEQ ID NO:6 or SEQ ID NO:8, under conditions comprising (i) hybridizing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 37° C. and (ii) washing in a solution comprising 1×SSC in the absence of helix destabilizing compounds, at a temperature of about 81 20 C., and

(b) a means for detecting the inhibition of said cyclooxygenase activity.

14. The kit of claim 13 , wherein said isolated protein having cyclooxygenase activity is selected from the group consisting of:

(a) an isolated protein comprising an amino acid sequence at least 95% identical to SEQ ID NO:2 or SEQ ID NO:5; and

(b) an isolated protein comprising at least 115 contiguous amino acids from SEQ ID NO:5.

15. The kit of claim 13 , wherein said isolated protein having cyclooxygenase activity is selected from the group consisting of:

(a) an isolated protein comprising an amino acid sequence at least 98% identical to SEQ ID NO:2 or SEQ ID NO:5; and

(b) an isolated protein comprising at least 300 contiguous amino acids from SEQ ID NO:5.
