Abstract

Disclosed is a method for producing cardiomyocytes in vivo by administering to the heart of an individual a cardiomyocyte producing amount of mesenchymal stem cells. These cells can be administered as a liquid injectable or as a preparation of cells in a matrix which is or becomes solid or semi-solid. The cells can be genetically modified to enhance myocardial differentiation and integration. Also disclosed is a method for replacing cells ex vivo in a heart valve for implantation.

Claims (22)

1. A method for producing cardiomyocytes in the heart of an individual, comprising:

administering directly to the heart of the individual a cardiomyocyte producing amount of allogeneic mesenchymal stem cells, wherein the mesenchymal stem cells differentiate into cardiomyocytes in the heart of the individual.

2. The method of claim 1 , wherein the mesenchymal stem cells are administered in a suspension containing about 10 to about 40×10 6 of the mesenchymal stem cells/ml.

3. The method of claim 2 , wherein the cell suspension is at least about 500 μl.

4. The method of claim 1 , wherein the mesenchymal stem cells are administered in a pharmaceutically acceptable liquid injectable carrier.

5. The method of claim 1 wherein the mesenchymal stem cells are human.

6. The method of claim 1 , wherein the mesenchymal stem cells are administered by intracardiac injection.

7. The method of claim 1 , wherein the individual has cardiac muscle that has been damaged through disease.

8. The method of claim 1 , wherein the individual has experienced a myocardial infarction.

9. The method of claim 1 , wherein the mesenchymal stem cells are administered directly to at least one damaged portion of heart tissue.

10. The method of claim 1 , wherein the mesenchymal stem cells are administered during an open surgical procedure.

11. A method for producing cardiomyocytes in the heart of an individual, comprising:

administering directly to the heart of the individual a cardiomyocyte producing amount of autologous mesenchymal stem cells, wherein the mesenchymal stem cells differentiate into cardiomyocytes in the heart of the individual.

12. The method of claim 11 , wherein the mesenchymal stem cells are administered as a suspension containing about 10 to about 40×10 6 of the mesenchymal stem cells/ml.

13. The method of claim 12 , wherein the cell suspension is at least about 500 μl.

14. The method of claim 11 , wherein the mesenchymal stem cells are administered in a pharmaceutically acceptable liquid injectable carrier.

15. The method of claim 11 wherein the mesenchymal stem cells are human.

16. The method of claim 11 , wherein the mesenchymal stem cells are administered by intracardiac injection.

17. The method of claim 11 , wherein the individual has cardiac muscle that has been damaged through disease.

18. The method of claim 11 , wherein the individual has experienced a myocardial infarction.

19. The method of claim 11 , wherein the mesenchymal stem cells are administered directly to at least one damaged portion of heart tissue.

20. The method of claim 11 , wherein the mesenchymal stem cells are administered during an open surgical procedure.
