Abstract

The present invention is directed to antagonists of CS1 that bind to and neutralize at least one biological activity of CS1. The invention also includes a pharmaceutical composition comprising such antibodies or antigen-binding fragments thereof. The present invention also provides for a method of preventing or treating disease states, including autoimmune disorders and cancer, in a subject in need thereof, comprising administering into said subject an effective amount of such antagonists.

Claims (16)

1. A monoclonal antibody or antigen binding fragment that binds a protein encoded by SEQ ID NO:1 and inhibits immunoglobulin secretion, wherein said antibody or antigen binding fragment decreases by at least 50% in a competitive binding assay the binding to a protein encoded by SEQ ID NO:1 of a control antibody obtainable from a hybridoma bearing ATCC designation No. PTA-5091.

2. The monoclonal antibody or antigen binding fragment of claim 1 which comprises the amino acid sequences corresponding to SEQ ID NO:9, SEQ ID NO:10, SEQ ID NO:11, SEQ NO:12, SEQ NO:13 and SEQ ID NO:14.

3. The monoclonal antibody or antigen binding fragment of claim 2 which comprises a VH region comprising the amino acid sequence corresponding to SEQ ID NO:3, and a VL region comprising the amino acid sequence corresponding to SEQ ID NO:4.

4. The monoclonal antibody or antigen binding fragment of claim 1 which comprises the amino acid sequences corresponding to SEQ ID NO:21, SEQ ID NO:22, SEQ ID NO:23, SEQ NO:24, SEQ NO:25 and SEQ ID NO:26.

5. The monoclonal antibody or antigen binding fragment of claim 4 which comprises a VH region comprising the amino acid sequence corresponding to SEQ ID NO:7, and a VL region comprising the amino acid sequence corresponding to SEQ ID NO:8.

6. The monoclonal antibody or antigen binding fragment of claim 1 which inhibits immunoglobulin secretion by at least 40%.

7. The monoclonal antibody or antigen binding fragment of claim 1 which is an IgG 1 .

8. The monoclonal antibody or antigen binding fragment of claim 1 which is a human or humanized antibody.

9. A conjugate compound comprising a monoclonal antibody or antigen binding fragment according claim 1 linked to an effector moiety or a detectable label.

10. The conjugate compound of claim 9 in which the antibody or antigen binding fragment is linked to a detectable label.

11. The conjugate compound of claim 10 in which the detectable label is a radioactive compound, a fluorescent compound, an enzyme, a substrate, an epitope tag or a toxin.

12. The conjugate compound of claim 9 in which the antibody or antigen binding fragment is linked to an effector moiety.

13. The conjugate compound of claim 12 in which the effector moiety is a cytotoxic agent.

14. The conjugate compound of claim 12 in which the effector moiety is a detection moiety, an activatable moiety, a chemotherapeutic agent, a lipase, an antibiotic, a chemoattracting moiety, an immune modulator, or a radioisotope.

15. A pharmaceutical composition comprising the antibody or antigen binding fragment of claim 1 .

16. A pharmaceutical composition comprising the conjugate compound of claim 9 .
