Compositions and methods for binding lysophosphatidic acid
View Patent ↗Compositions and methods for making and using anti-LPA agents, for example, monoclonal antibodies, are described. Variable domain and complementarity determining region amino acid sequences of several monoclonal antibodies against LPA are disclosed, as is a consensus anti-LPA monoclonal antibody variable domain sequence.
1. A diagnostic reagent comprising a derivatized lysophosphatidic acid (LPA) conjugated to a solid support or carrier, wherein the derivatized LPA is selected from the group consisting of 18:2, 18:1, 18:0, 16:0 and 20:4 LPA, and wherein the hydrocarbon chain of the derivatized LPA includes a reactive group at the most distal carbon position of the hydrocarbon chain.
2. A diagnostic reagent according to claim 1 wherein the LPA is covalently conjugated to the solid support or carrier.
3. A diagnostic reagent according to claim 2 wherein the carrier is selected from the group consisting of polyethylene glycol, colloidal gold, adjuvant, a silicone bead, and a protein.
4. A diagnostic reagent according to claim 3 wherein the carrier is a protein selected from the group consisting of keyhole limpet hemocyanin, albumin, bovine thyroglobulin, and soybean trypsin inhibitor.
5. A diagnostic reagent according to claim 1 wherein the reactive group is selected from the group consisting of a sulfhydryl (thiol) group, a carboxylic acid group, a cyano group, an ester group, a hydroxyl group, an alkene group, an alkyne group, an acid chloride group, and a halogen atom group.
6. A diagnostic reagent according to claim 1 wherein the reactive group comprises a sulfhydryl (thiol) group.
