Abstract

Polyols stabilize polymorphous form of rifaximin, in particular the β form. When polyols having at least two hydroxy groups are added to rifaximin powder, polymorph β is stable and remains stable in time independently from the environment humidity. In this invention a method to prepare formulations constituted by pure and stable polymorphous forms able to give a pharmaceutical product is described.

Claims (45)

1. A method of stabilizing rifaximin in polymorphous form β comprising

i) obtaining a solid polymorphous form of rifaximin;

ii) adding a liquid mixture comprising one or more polyol compounds bearing at least two hydroxy groups to obtain a mixture comprising the polymorphous form of rifaximin and the one or more polyol compounds; and

iii) drying the mixture, wherein the rifaximin in polymorphous β has a residual water content lower than 4.5%.

2. The method of claim 1 , wherein the one or more polyols are selected from the group consisting of polyols containing two to seven carbon atoms and two to seven hydroxy groups, monosaccharides, disaccharides, polysaccharides, xantan gum, di-hydroxy acids and poly-hydroxy acids.

3. The method of claim 1 , wherein the one or more polyols are compounds of formula I:

H—[O—CH—(X)—CH 2 ] n —OH  (I)

wherein X is hydrogen or lower alkyl, and n is between 1 and 20.

4. The method of claim 1 , wherein the polyol is 1,2,3-propanetriol.

5. The method of claim 1 , wherein the polyol is a compound having the general formula H—[O—CH 2 —CH 2 ] n —OH, wherein n is between 2 and 14.

6. The method of claim 1 , wherein the polyol is 1,2-propanediol.

7. The method of claim 2 , wherein the polysaccharide is selected from the group consisting of starch, cellulose or derivatives thereof, dextrin and maltodextrin.

8. A stabilized polymorphous rifaximin β in solid state obtained by the process of:

i) obtaining a solid polymorphous form of rifaximin;

ii) adding a liquid mixture comprising one or more polyol compounds bearing at least two hydroxy groups to obtain a mixture comprising the polymorphous form of rifaximin and the one or more polyol compounds; and

iii) drying the mixture to achieve a water content of below 4.5%.

A pharmaceutical composition for the treatment of pathologies that need antibiotic therapy, said composition comprising rifaximin in the polymorphous β form and an excipient, wherein said polymorphous β form is stabilized by one or more polyols bearing at least two hydroxy groups, and has a water content of below 4.5%.

10. The pharmaceutical composition of claim 9 , wherein the rifaximin is in the form of microgranules of gastroresistant rifaximin β.

11. The pharmaceutical composition of claim 10 , wherein the composition is in thermo-welded bags.

12. The pharmaceutical composition of claim 9 , wherein the composition is in tablet form.

13. The pharmaceutical composition of claim 9 , wherein the excipient is selected from the group consisting of diluents, ligands, lubricants, disintegrants, dyes, flavors and sweeteners.

14. The pharmaceutical composition of claim 9 , wherein the one or more polyols are compounds of formula I:

H—[O—CH—(X)—CH 2 ] n —OH  (I)

wherein X is hydrogen or lower alkyl, and n is between 1 and 20.

15. The pharmaceutical composition of claim 14 , wherein the polyol is selected from the group consisting of a compound having the general formula H—[O—CH 2 —CH 2 ] n —OH, wherein n is between 2 and 14, 1,2,3-propanetriol, 1,2-propanediol, and a mixture thereof.

The pharmaceutical composition of claim 9 , wherein the one or more polyols are selected from the group consisting of polyols containing two to seven carbon atoms and two to seven hydroxy groups, monosaccharides, disaccharides, polysaccharides, xantan gum, di-hydroxy acids and poly-hydroxy acids.

17. The pharmaceutical composition of claim 9 , wherein the polyol is 1,2,3-propanetriol.

18. The pharmaceutical composition of claim 14 , wherein n is between 2 and 14.

19. The pharmaceutical composition of claim 9 , wherein the polyol is 1,2-propanediol.

20. The pharmaceutical composition of claim 9 , wherein the polyol is a polysaccharide selected from the group consisting of starch, cellulose, hydroxypropyl cellulose, hydroxyethyl cellulose, carboxymethyl cellulose, dextrin and maltodextrin.

21. A stabilized polymorphous β form of rifaximin in solid state comprising one or more polyols and having water content of less than 4.5%.

22. The rifaximin of claim 21 , wherein the one or more polyols are compounds of formula I:

H—[O—CH—(X)—CH 2 ] n —OH  (I)

wherein X is hydrogen or lower alkyl, and n is between 1 and 20.

23. The rifaximin of claim 22 , wherein the polyol is selected from the group consisting of a compound having the general formula H—[O—CH 2 —CH 2 ] n —OH, wherein n is between 2 and 14, 1,2,3-propanetriol, and 1,2-propanediol.

24. The rifaximin of claim 21 , wherein the one or more polyols are selected from the group consisting of polyols containing two to seven carbon atoms and two to seven hydroxy groups, monosaccharides, disaccharides, polysaccharides, xantan gum, di-hydroxy acids and poly-hydroxy acids.

25. The rifaximin of claim 21 , wherein the polyol is 1,2,3-propanetriol.

26. The rifaximin of claim 22 , wherein n is between 2 and 14.

27. The rifaximin of claim 21 , wherein the polyol is 1,2-propanediol.

28. A pharmaceutical composition for the treatment of pathologies that need antibiotic therapy, said composition comprising rifaximin in the polymorphous β form and an excipient, wherein said polymorphous β form is stabilized by one or more polyols bearing at least two hydroxy groups and has a water content of below 4.5% and further wherein said polyol stabilized polymorphous β form is obtained by the process of:

i) obtaining a solid polymorphous form of rifaximin;

ii) adding a liquid mixture comprising one or more polyol compounds bearing at least two hydroxy groups to obtain a mixture comprising the polymorphous form of rifaximin and the one or more polyol compounds; and

iii) drying the mixture to achieve a water content of below 4.5.

The pharmaceutical composition of claim 28 , wherein the rifaximin in polymorphous form β is prepared by a process comprising contacting rifaximin in solid state with an aqueous solution of one or more polyols at a concentration of 5% to 59% (w/w), at a temperature between 30° C. and 90° C., for a period of time of between 1 and 24 hours, to obtain a mixture of rifaximin and polyols, and wherein the drying step comprises drying the mixture of rifaximin and polyols at a temperature of between 30 and 80° C., at ambient pressure or under vacuum, for a period of time of between 2 and 72 hours.

The pharmaceutical composition of claim 29 , wherein the contacting step comprises spraying the polyol aqueous solution at concentrations of from 5 to 50% (w/w) on the rifaximin β in solid state in a fluid bed apparatus at an inlet temperature of between 40° C. and 90° C., and wherein the drying step comprises drying the mixture under an air flow at a temperature of between 40 and 90° C.
