Abstract

An IL-1β binding antibody or IL-1β binding fragment thereof comprising the amino acid sequence of SEQ ID NO: 8 and SEQ ID NO: 9, and related nucleic acids, vectors, cells, and compositions, as well as method of using same to treat or prevent a disease. IL-1β binding antibodies or IL-1β binding fragments thereof are provided which have desirable affinity and potency.

Claims (10)

1. An isolated IL-1β binding antibody or IL-1β binding fragment thereof comprising a heavy chain variable region of SEQ ID NO: 8 and a light chain variable region of SEQ ID NO: 9.

2. The antibody or antibody fragment of claim 1 , wherein the antibody or antibody fragment comprises a kappa light chain.

3. The antibody or antibody fragment of claim 1 , wherein the antibody or antibody fragment comprises an IgG2 region.

4. The antibody or antibody fragment of claim 1 , wherein the antibody or antibody fragment is an Fab, an F(ab′)2, an Fv, or a single-chain antibody fragment.

An isolated IL-1β binding antibody or IL-1β binding fragment thereof, wherein the antibody or fragment binds to human IL-1β with a dissociation constant less than 1 pM, and the antibody or fragment competes with the binding of an antibody comprising a light chain variable region of SEQ ID NO: 9 and a heavy chain variable region of SEQ ID NO: 8.

6. A composition comprising (a) the antibody or antibody fragment of claim 1 or 5 , and (b) a suitable carrier.

7. The composition of claim 6 , wherein the carrier is a pharmaceutically acceptable carrier.

8. The composition of claim 7 , wherein the composition is in a form suitable for intraarticular, subcutaneous, intravenous, intraperitoneal, intracerebral, intraparenchymal, intracerebroventricular, intramuscular, intraocular, intraarterial, intralesional, oral or inhalation administration.

9. The composition of claim 7 , wherein the composition comprises a lyoprotectant, a surfactant, a filler, a binder, and/or bulking agent.

10. The composition of claim 7 , wherein the composition is a controlled-release or sustained-release pharmaceutical composition.
