Abstract

An IL-1β binding antibody or IL-1β binding fragment thereof comprising the amino acid sequence of SEQ ID NO: 2, and related nucleic acids, vectors, cells, and compositions, as well as method of using same to treat or prevent a disease, and a method of preparing an affinity matured IL-1β binding polypeptide. IL-1β binding antibodies or IL-1β binding fragments thereof are provided which have desirable affinity and potency.

Claims (12)

1. A method of treating an IL-1β related coronary condition in a mammal comprising administering an effective amount of an IL-1β binding antibody or IL-1β binding fragment thereof, said antibody or fragment comprising a heavy chain variable region of SEQ ID NO: 15 and a light chain variable region of SEQ ID NO: 11, wherein the IL-1β related condition is selected from coronary restenosis, myocardial infarction, and coronary artery bypass graft.

2. The method of claim 1 , wherein the condition is coronary restenosis.

3. The method of claim 1 , wherein the condition is myocardial infarction.

4. The method of claim 1 , wherein the condition is coronary artery bypass graft.

5. The method of claim 1 , wherein the mammal is a human.

6. The method of claim 1 , wherein the effective amount of the antibody or antibody fragment is effective to inhibit IL-1β induced expression of serum IL-6 in an animal by at least 50% as compared to the level of serum IL-6 in the absence of the antibody.

A method of treating an IL-1β related coronary condition in a mammal comprising administering an effective amount of an IL-1β binding antibody or IL-1β binding fragment thereof, wherein the antibody or fragment binds to human IL-1β with a dissociation constant less than 1 pM, and the antibody or fragment competes with the binding of an antibody comprising the light chain variable region of SEQ ID NO: 11 and the heavy chain variable region of SEQ ID NO: 15, wherein the IL-1β related condition is selected from coronary restenosis, myocardial infarction, and coronary artery bypass graft.

8. The method of claim 7 , wherein the condition is coronary restenosis.

9. The method of claim 7 , wherein the condition is myocardial infarction.

10. The method of claim 7 , wherein the condition is coronary artery bypass graft.

11. The method of claim 7 , wherein the mammal is a human.

12. The method of claim 7 , wherein the effective amount of the antibody or antibody fragment is effective to inhibit IL-1 induced expression of serum IL-6 in an animal by at least 50% as compared to the level of serum IL-6 in the absence of the antibody.
