Antibody identifying an antigen-bound antibody and an antigen-unbound antibody, and method for preparing the same
View Patent ↗The present invention relates to an antibody that recognizes a first antibody, the antibody specifically recognizing one of a free first antibody and an antigen-binding first antibody. More specifically, the above antibody is a domino antibody that specifically recognizes and binds to an antigen-binding first antibody, or an antibody-unlocking antibody that specifically recognizes and binds to a free first antibody.
1. A method for obtaining an antibody-unlocking antibody, comprising:
(a) immunizing a first host with an antigen,
(b) isolating from the first host at least one first antibody capable of binding to the antigen,
(c) obtaining an antibody chain or a fragment thereof from the isolated first antibody,
(d) immunizing a second host with said antibody chain,
(e) obtaining at least one hybridoma from the spleen cells of the second host, wherein the hybridoma produces monoclonal antibodies,
and
(f) selecting an antibody-unlocking antibody from the monoclonal antibodies produced by the obtained hybridoma,
wherein the antibody-unlocking antibody specifically recognizes the first antibody when the first antibody is not bound to the antigen.
2. The method according to claim 1 , wherein the antibody chain is a light chain.
3. The method according to claim 1 , wherein the first host is a mouse, a rat, a rabbit, a hamster, or a monkey.
4. The method according to claim 1 , wherein the second host is a mouse, a rat, a rabbit, a hamster, or a monkey.
