Abstract

A method of producing a modified acellular tissue matrix (mATM) from an acellular tissue matrix (ATM), where the mATM has a reduced stretchiness relative to the ATM, without substantially compromising its associated structural or functional integrity. The method includes providing an acellular tissue matrix (ATM) and exposing the ATM to elastase for a period of time.

Claims (55)

1. A method comprising:

decellularizing a dermal tissue to produce an acellular tissue matrix (ATM), wherein decellularizing includes contacting the tissue with a processing solution containing one or more agents to prevent cross-linking; and

exposing the ATM to a solution containing elastase for a period of time, the exposure resulting in a modified ATM (mATM), and

controlling the exposure time and concentration of elastase in the solution to obtain a percent extension of the mATM under an amount of tensile force,

wherein the percent extension of the mATM in a toe region of the stress-strain curve resulting from an amount of tensile force is less than the percent extension of the ATM resulting from the same amount of tensile force.

2. The method of claim 1 , wherein the percent extension of the mATM is from about 14% to 24%.

3. The method of claim 1 , wherein the concentration of elastase ranges from about 0.1 units/milliliter to 0.5 units/milliliter.

4. The method of claim 3 , wherein the concentration of elastase ranges from about 0.2 units/milliliter to 0.25 units/milliliter.

5. The method of claim 1 , wherein the period of time ranges from about 12 hours to 24 hours.

6. The method of claim 5 , wherein the period of time ranges from 18 hours to 24 hours.

7. The method of claim 1 , further comprising agitating the ATM and the solution while exposing the ATM to elastase for a period of time.

8. The method of claim 1 , wherein the ATM is made from human dermal tissue.

9. The method of claim 1 , wherein the ATM is made from non-human mammalian dermal tissue.

10. The method of claim 9 , wherein the non-human mammalian dermal tissue is porcine dermal tissue.

11. The method of claim 9 , wherein the non-human mammalian dermal tissue is bovine dermal tissue.

12. A method comprising:

providing the modified acellular tissue matrix (mATM) of claim 1 ;

identifying a vertebrate subject as having an organ or tissue in need of repair or amelioration; and

placing the mATM in or on the tissue or organ.

13. A method of treating a group of acellular tissue matrices, the method comprising:

decellularizing a group of dermal tissues to provide a group of acellular tissue matrices (ATMs), wherein when an equal amount of tensile force is applied to each of the ATMs in the group, at least some of the ATMs in the group have a different percent extension than other ATMs in the group; and

wherein deceullularizing the group of tissues includes contacting the group of tissues with a solution containing one or more agents to prevent cross-linking;

exposing one or more ATMs of the group to a solution containing elastase for a period of time, and

controlling the exposure time and concentration of elastase in the solution to obtain a percent extension of the mATM under an amount of tensile force, the exposure resulting in one or more modified ATMs (mATMs),

wherein the percent extension of the one or more mATMs in a toe region of the stress-strain curve resulting from an amount of tensile force is less than or equal to the percent extension of the corresponding ATMs resulting from the same amount of tensile force,

wherein variations in the percent extension across the mATMs is less than variations in the percent extension across the ATMs.

14. The method of claim 13 , wherein the elastase is provided in a solution containing a concentration of elastase.

15. The method of claim 14 , wherein the concentration of elastase ranges from about 0.1 units/milliliter to 0.5 units/milliliter.

16. The method of claim 15 , wherein the concentration of elastase ranges from about 0.2 units/milliliter to 0.25 units/milliliter.

17. The method of claim 13 , wherein the percent extension of a plurality of the one or more of the mATMs ranges from 14% to 24%, and said amount of tensile force is about 5 newtons/cm.

18. The method of claim 13 , wherein the percent extension of a plurality of the one or more of the mATMs is about 19%, and said amount of tensile force is about 5 newtons/cm.

19. The method of claim 13 , wherein said period of time ranges between 12 to 24 hours.

20. The method of claim 19 , wherein said period of time ranges between 18 hours to 24 hours.

21. The method of claim 13 , further comprising agitating the one or more ATMs of the group and the solution while exposing one or more ATMs of the group to elastase for a period of time.

22. The method of claim 13 , wherein one or more of the ATMs are made from human dermal tissue.

23. The method of claim 13 , wherein one or more of the ATMs are made from non-human mammalian dermal tissue.

24. The method of claim 23 , wherein the non-human mammalian dermal tissue is porcine tissue.

25. The method of claim 23 , wherein the non-human mammalian dermal tissue is bovine tissue.

26. A method comprising:

providing the group of modified acellular tissue matrices (mATMs) of claim 13 ;

identifying a vertebrate subject as having an organ or tissue in need of repair or amelioration; and

placing one or more of the mATMs in or on the tissue or organ.

27. A method comprising:

decellularizing a dermal tissue to produce an acellular tissue matrix (ATM);

exposing the ATM to a solution containing elastase for a period of time, the exposure resulting in a modified ATM (mATM), and

controlling the exposure time and concentration of elastase in the solution to obtain a percent extension of the mATM under an amount of tensile force, and

wherein the percent extension of the mATM in a toe region of the stress-strain curve resulting from an amount of tensile force is less than or equal to the percent extension of the ATM resulting from the same amount of tensile force.

28. The method of claim 27 , wherein the percent extension of the mATM resulting from a tensile force of about 5 newtons/cm ranges from about 14% to 24%.

29. The method of claim 27 , wherein the ATM is made from human tissue.

30. The method of claim 27 , wherein the dermal tissue is porcine tissue.

31. A method comprising:

decellularizing a dermal tissue to produce an acellular tissue matrix (ATM);

exposing the ATM to a solution containing elastase for a period of time, the exposure resulting in a modified ATM (mATM), and

controlling the exposure time and concentration of elastase in the solution to obtain a percent elongation of the mATM under an amount of tensile force, and

wherein the percent elongation of the mATM in a toe region of the stress-strain curve resulting from an amount of tensile force is less than or equal to the percent elongation of the ATM resulting from the same amount of tensile force.
