Abstract

The present invention relates to G-CSF polypeptides and their uses, particularly for therapeutic or prophylactic treatment in human subjects. The invention also relates to nucleic acids encoding said polypeptides, vectors comprising such nucleic acids and recombinant cells containing the same. The invention further discloses methods of producing such polypeptides, as well as methods and tools for detecting or dosing these polypeptides in any sample.

Claims (34)

1. An isolated nucleic acid molecule encoding a polypeptide comprising:

a) SEQ ID NO: 8;

b) a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant hinds a G-CSF receptor; or

c) a fusion protein comprising an additional amino acid domain operably linked to:

i) SEQ ID NO: 8; or

ii) a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

2. The isolated nucleic acid molecule according to claim 1 , wherein said nucleic acid molecule is a cDNA molecule.

3. The isolated nucleic acid molecule according to claim 1 , wherein said nucleic acid molecule encodes SEQ ID NO: 8.

4. The isolated nucleic acid molecule according to claim 1 , wherein said nucleic acid molecule encodes a fusion protein comprising SEQ ID NO: 8 operably linked to an additional amino acid domain.

5. The isolated nucleic acid molecule according to claim 4 , wherein said additional amino acid domain is a signal peptide, a tag, a targeting peptide, the constant domain of an immunoglobulin molecule or a biologically active protein.

6. The isolated nucleic acid molecule according to claim 3 , wherein said nucleic acid comprises SEQ ID NO: 7.

The isolated nucleic acid molecule according to claim 1 , wherein said nucleic acid molecule encodes a fusion protein comprising an additional amino acid domain operably linked to a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

8. The isolated nucleic acid molecule according to claim 1 , wherein said nucleic acid molecule encodes a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

9. A vector comprising a nucleic acid molecule according to claim 1 .

10. The vector according to claim 9 , wherein said nucleic acid molecule encodes SEQ ID NO: 8.

11. The vector according to claim 9 , wherein said nucleic acid molecule encodes a fusion protein comprising SEQ ID NO: 8 operably linked to an additional amino acid domain.

12. The vector according to claim 9 , wherein said additional amino acid domain is a signal peptide, a tag, a targeting peptide, the constant domain of an immunoglobulin molecule or a biologically active protein.

13. The vector according to claim 10 , wherein said nucleic acid comprises SEQ ID NO: 7.

14. The vector according to claim 9 , wherein said nucleic acid molecule encodes a fusion protein comprising an additional amino acid domain operably linked to a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

15. The vector according to claim 9 , wherein said nucleic acid molecule encodes a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

16. An isolated host cell comprising a vector according to claim 9 .

17. The isolated host cell according to claim 16 , wherein said host cell is a prokaryotic or eukaryotic cell.

18. The isolated host cell according to claim 16 , wherein said nucleic acid molecule encodes SEQ ID NO: 8.

19. The isolated host cell according to claim 16 , wherein said nucleic acid molecule encodes a fusion protein comprising SEQ ID NO: 8 operably linked to an additional amino acid domain.

20. The isolated host cell according to claim 16 , wherein said additional amino acid domain is a signal peptide, a tag, a targeting peptide, the constant domain of an immunoglobulin molecule or a biologically active protein.

21. The isolated host cell according to claim 18 , wherein said nucleic acid comprises SEQ ID NO: 7.

The isolated host cell according to claim 16 , wherein said nucleic acid molecule encodes a fusion protein comprising an additional amino acid domain operably linked to a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

23. The isolated host cell according to claim 16 , wherein said nucleic acid molecule encodes a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.

24. A method of producing a polypeptide comprising culturing a recombinant host cell under conditions allowing expression of the nucleic acid molecule, and recovering the polypeptide produced, wherein said recombinant host cell comprises a nucleic acid encoding:

a) SEQ ID NO: 8;

b) a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor; or

c) a fusion protein comprising an additional amino acid domain operably linked to:

i) SEQ ID NO: 8; or

ii) a variant of SEQ ID NO: 8 comprising up to 10 amino acid substitutions to SEQ ID NO: 8, wherein said variant binds a G-CSF receptor.
