Abstract

The present invention provides a monoclonal antibody which specifically recognizes CD27 containing an O-linked sugar chain to which galactose is not bound and binds to its extracellular region, or a method for using the same. The present invention can provide a monoclonal antibody or an antibody fragment thereof, which specifically recognizes a polypeptide encoded by CD27 gene containing an O-linked sugar chain to which galactose is not bound, and binds to its extracellular region; a hybridoma which produces the antibody; a DNA which encodes the antibody; a vector which comprises the DNA; a transformant obtainable by transforming the vector; a process for producing an antibody or an antibody fragment thereof using the hybridoma or the transformant; and a diagnostic agent or a therapeutic agent comprising the antibody or the antibody fragment thereof as an active ingredient.

Claims (24)

1. A monoclonal antibody which does not recognize an extracellular region of a CD27 containing an O-linked sugar chain to which galactose is bound, but recognizes and binds to a polypeptide extracellular region of CD27, said polypeptide extracellular region containing an O-linked sugar chain to which galactose is not bound, or an antigen-binding fragment thereof.

2. The monoclonal antibody or the antigen-binding fragment thereof according to claim 1 , wherein the O-linked sugar chain to which galactose is not bound is at least one O-linked sugar chain selected from a sialyl Tn antigen and a Tn antigen.

3. The monoclonal antibody or the antigen-binding fragment thereof according to claim 2 , wherein the O-linked sugar chain to which galactose is not bound is the Tn antigen.

4. The monoclonal antibody or the antigen-binding fragment thereof according to claim 1 , wherein the monoclonal antibody is a monoclonal antibody which competes with a monoclonal antibody KM4030.

5. The monoclonal antibody or the antigen-binding fragment thereof according to claim 1 , wherein the monoclonal antibody is a monoclonal antibody which binds to an epitope to which a monoclonal antibody KM4030 binds.

6. The monoclonal antibody or the antigen-binding fragment thereof according to claim 1 , wherein the monoclonal antibody is a monoclonal antibody which is produced by a hybridoma deposited under Accession No. FERM BP-10976.

7. The antibody or the antigen-binding fragment thereof according to claim 1 , wherein the monoclonal antibody is a recombinant antibody.

8. The antibody or the antigen-binding fragment thereof according to claim 7 , wherein the recombinant antibody is an antibody selected from a human chimeric antibody, a humanized antibody and a human antibody.

9. The antigen-binding fragment according to claim 1 , which is an antigen-binding fragment selected from the group consisting of Fab, Fab′, F(ab′) 2 , a single chain antibody (scFv), a diabody, and a disulfide stabilized Fv (dsFv).

10. A hybridoma which produces the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

11. The hybridoma according to claim 10 , wherein the hybridoma is a hybridoma deposited under Accession No. FERM BP-10976.

A process for producing the antibody or the antigen-binding fragment thereof according to claim 1 , comprising culturing the hybridoma described in claim 11 in a medium to form and accumulate the monoclonal antibody or the antibody fragment thereof described in claim 2 in the culture, and then collecting the antibody or the antibody fragment thereof from the culture.

A method for immunologically detecting or measuring CD27 containing an O-linked sugar chain to which galactose is not bound, comprising detecting or measuring the CD27 or a cell expressing the CD27 which are bound in a sample to the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

14. A reagent for detecting CD27 containing an O-linked sugar chain to which galactose is not bound, comprising the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

15. A diagnostic agent for a disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound, comprising the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

16. The diagnostic agent according to claim 15 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is IgA nephropathy.

17. The diagnostic agent according to claim 15 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is cancer.

18. A therapeutic agent for a disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound, comprising the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 as an active ingredient.

19. The therapeutic agent according to claim 18 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is IgA nephropathy.

20. The therapeutic agent according to claim 18 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is cancer.

21. A diagnostic method for a disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound, comprising detecting or measuring a cell expressing CD27 containing an O-linked sugar chain to which galactose is not bound, wherein said cell is bound in a sample to the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

A diagnostic method for a disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound, comprising detecting or measuring CD27 containing an O-linked sugar chain to which galactose is not bound, wherein the CD27 containing an O-linked sugar chain to which galactose is not bound is bound in a sample to the monoclonal antibody or the antigen-binding fragment thereof according to claim 1 .

23. The diagnostic method according to claim 21 or 22 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is IgA nephropathy.

24. The diagnostic method according to claim 21 or 22 , wherein the disease relating to CD27 containing an O-linked sugar chain to which galactose is not bound is cancer.
