Abstract

Endogenous LINGO-1 is a negative regulator for neuronal survival, axon regeneration, oligodendrocyte differentiation and myelination. Molecules that block endogenous LINGO-1 function, such anti-LINGO-1 antibodies can be used as therapeutics for the treatment of neuron and oligodendrocyte dysfunction. The present invention provides antibodies specific for LINGO-1, and methods of using such antibodies as antagonists of endogenous LINGO-1 function. The invention further provides specific hybridoma and phage library-derived monoclonal antibodies, nucleic acids encoding these antibodies, and vectors and host cells comprising these antibodies. The invention further provides methods of promoting oligodendrocyte survival and myelination in a vertebrate, comprising administering to a vertebrate in need of such treatment an effective amount of an anti-LINGO-1 antibody.

Claims (17)

1. An isolated antibody or antigen-binding fragment thereof that can specifically bind to a LINGO-1 polypeptide, wherein the antibody or antigen-binding fragment thereof comprises an immunoglobulin heavy chain variable region (VH) wherein the VH CDR1, CDR2, and CDR3 comprise the amino acids of SEQ ID NOs: 2, 3, and 30, respectively.

2. The isolated antibody or antigen-binding fragment thereof of claim 1 , wherein the VH comprises the sequence of SEQ ID NO: 66.

The isolated antibody or antigen-binding fragment thereof of claim 1 further comprising a VL polypeptide comprising VL CDR1, CDR2, and CDR3 regions, wherein the VL CDR1, CDR2, and CDR3 regions are identical, respectively, to the reference VL CDR1, CDR2, and CDR3 sequences SEQ ID NOs: 10, 11, and 12.

4. An isolated antibody or antigen-binding fragment thereof that can specifically bind to a LINGO-1 polypeptide, wherein the VL comprises VL CDR1, CDR2, and CDR3 regions identical, respectively, to SEQ ID NOs: 10, 11, and 12.

5. The isolated antibody or antigen-binding fragment thereof of claim 4 , wherein the VL comprises the sequence of SEQ ID NO:9.

6. The isolated antibody or antigen-binding fragment thereof of claim 5 , further comprising a VH polypeptide comprising VH CDR1, CDR2, and CDR3 regions, wherein the VH CDR1, CDR2, and CDR3 sequences are identical, respectively, to SEQ ID NOs: 2, 3, and 30.

7. The isolated antibody or antigen-binding fragment thereof of claim 6 , wherein the VH comprises the sequence of SEQ ID NO:66.

8. The isolated antibody or antigen-binding fragment thereof of claim 4 , further comprising a VH polypeptide comprising the sequence of SEQ ID NO: 66.

9. The antibody or fragment thereof of claim 4 , which is an antagonist of LINGO-1 mediated neuronal cell death.

10. The antibody or fragment thereof of claim 4 , which is an antagonist of LINGO-1-mediated myelination inhibition.

11. The antibody or fragment thereof of claim 4 , which is an antagonist of LINGO-1 mediated oligodendrocyte differentiation inhibition.

12. The antibody or fragment thereof of claim 4 , further comprising a heterologous polypeptide fused thereto.

13. The antibody or fragment thereof of claim 4 , wherein the antibody is conjugated to an agent selected from the group consisting of a therapeutic agent, a prodrug, a peptide, a protein, an enzyme, a virus, a lipid, a biological response modifier, a pharmaceutical agent, or PEG.

14. A composition comprising the antibody or fragment thereof of claim 4 and a carrier.

15. An isolated anti-LINGO-1 antibody, or antigen-binding fragment thereof, produced by a method comprising

culturing a host cell comprising a composition comprising a VH encoding polynucleotide and a VL encoding polynucleotide, wherein the VH encoding polynucleotide and the VL encoding polynucleotide, respectively, comprise polynucleotides encoding the amino acid sequences of reference polypeptides SEQ ID NO:66 and SEQ ID NO:9; and

recovering the antibody.
