Abstract

Nucleic acids encoding a new family of chemokines, the CX3C family, from a mammal, reagents related thereto, including specific antibodies, and purified proteins are described. Methods of using said reagents and related diagnostic kits are also provided.

Claims (23)

1. An isolated antibody or antigen-binding fragment thereof which specifically binds to a polypeptide wherein said polypeptide a) chemoattracts peripheral blood monocytes or T-cells, b) consists of an amino acid sequence comprising at least 95% sequence identity to the amino acid sequence set forth in SEQ ID NO: 8, and c) comprises a chemokine domain identical to residues 25 to 100 of the amino acid sequence set forth in SEQ ID NO: 8.

2. The antibody or antigen-binding fragment thereof of claim 1 , which is detectably labeled.

3. The antibody or antigen-binding fragment thereof of claim 1 , which is a polyclonal antibody.

4. The antibody or antigen-binding fragment thereof of claim 1 , which is a monoclonal antibody.

5. The antibody or antigen-binding fragment thereof of claim 1 , which is a Fab.

6. The antibody or antigen-binding fragment thereof of claim 1 , which is a F(ab) 2 .

7. The antibody or antigen-binding fragment thereof of claim 1 , which is sterile.

8. The antibody or antigen-binding fragment thereof of claim 1 , which is an antibody, wherein the antibody exhibits a Kd to said polypeptide of greater than about 100 nM.

9. The antibody or antigen-binding fragment thereof of claim 1 , which is an antibody, wherein the antibody exhibits a Kd to said polypeptide of greater than about 30 nM.

10. The antibody or antigen-binding fragment thereof of claim 1 , which is an antibody, wherein the antibody exhibits a Kd to said polypeptide of greater than about 10 nM.

11. The antibody or antigen-binding fragment thereof of claim 1 , which is an antibody, wherein the antibody exhibits a Kd to said polypeptide of greater than about 3 nM.

12. A kit comprising the antibody or antigen-binding fragment thereof of claim 1 , and: a) instructional material for the antibody or antigen-binding fragment thereof; or b) a container into which the antibody or antigen-binding fragment thereof is segregated; or c) both a) and b).

13. A complex comprising an antibody or antigen-binding fragment thereof of claim 1 bound to said polypeptide.

14. A pharmaceutical composition comprising the antibody or antigen-binding fragment of claim 1 and a pharmaceutically acceptable carrier.

15. An isolated antibody or antigen-binding fragment thereof which specifically binds to a polypeptide consisting of an amino acid sequence set forth in SEQ ID NO: 8.

16. The antibody or antigen-binding fragment thereof of claim 15 , wherein said polypeptide is denatured.

17. An isolated antibody or antigen-binding fragment thereof, that specifically binds to a polypeptide consisting of Gln25 to Gly100 of the amino acid sequence set forth in SEQ ID NO: 8.

18. The antibody or antigen-binding fragment thereof of claim 17 that specifically binds to a polypeptide consisting of Gln25 to Gly100 of the amino acid sequence set forth in SEQ ID NO: 8, wherein the antibody or fragment is raised against a purified or recombinantly produced polypeptide comprising Gln25 to Gly100 of the amino acid sequence set forth in SEQ ID NO: 8.

The antibody or antigen-binding fragment thereof of claim 18 that specifically binds to a polypeptide consisting of Gln25 to Gly100 of the amino acid sequence set forth in SEQ ID NO: 8, wherein the antibody or fragment is raised against a purified or recombinantly produced polypeptide comprising a mature polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 8.

A method for detecting a polypeptide in a biological sample, comprising: a) contacting the biological sample with an antibody or antigen-binding fragment thereof of claim 1 under conditions to permit formation of an antibody or antigen-binding fragment thereof: polypeptide complex; and b) detecting the complex.

21. The method of claim 20 , wherein the biological sample is human.

22. An isolated monoclonal antibody that binds specifically to a polypeptide consisting of amino acids 25-397 of SEQ ID NO: 8.

23. A pharmaceutical composition comprising an isolated monoclonal antibody that binds specifically to a polypeptide consisting of amino acids 25-397 of SEQ ID NO: 8 and a pharmaceutically acceptable carrier.
