Abstract

The present invention relates to novel compounds that act as chemical uncouplers. Compounds of the invention are useful, inter alia, in the treatment, including prevention, of obesity, diabetes and a number of diseases or conditions associated therewith.

Claims (26)

1. A compound, which is a compound selected from the group consisting of:

6-tert-Butyl-4-chloro-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol;

5-tert-Butyl-3-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-4-hydroxy-2-methyl-benzonitrile;

N-[5-tert-Butyl-3-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thia-diazin-3-yl)-4-hydroxy-2-methyl-phenyl]-methanesulfonamide;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-4-trifluoromethylsulfanyl-phenol;

6-tert-Butyl-4-chloro-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]-thiadiazin-3-yl)-3-ethyl-phenol;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-4-methylsulfanyl-phenol;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-4-methanesulfinyl-3-methyl-phenol;

(R)-6-tert-Butyl-2-(1,1-dioxo-6-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thia-diazin-3-yl)-4-methanesulfinyl-3-methyl-phenol;

(S)-6-tert-Butyl-2-(1,1-dioxo-6-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thia-diazin-3-yl)-4-((S)-methanesulfinyl)-3-methyl-phenol;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-4-methanesulfonyl-3-methyl-phenol;

6-tert-Butyl-4-fluoro-2-(7-methanesulfonyl-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-4-fluoro-3-methyl-phenol;

6-tert-Butyl-4-chloro-2-(7-methanesulfonyl-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol;

N-[5-tert-Butyl-3-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thia-diazin-3-yl)-4-hydroxy-2-methyl-phenyl]-methanesulfonamide;

6-tert-Butyl-2-(1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-isopropyl-phenol;

6-tert-Butyl-2-(5-chloro-1,1-dioxo-7-trifluoromethanesulfonyl-1,2-dihydro-1λ 6 -benzo[1,2,4]-thiadiazin-3-yl)-4-fluoro-3-methyl-phenol;

6-tert-Butyl-2-(7-difluoromethylsulfanyl-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-4-fluoro-3-methyl-phenol;

6-tert-Butyl-4-fluoro-2-[7-(heptafluoropropane-1-sulfonyl)-1,1-dioxo-1,2-dihydro-1λ 6 -benzo-[1,2,4]thiadiazin-3-yl]-3-methyl-phenol;

4-Bromo-6-tert-butyl-2-(6-chloro-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol; and

6-tert-Butyl-4-chloro-2-(6-chloro-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol;

or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 , where the compound is 4-bromo-6-tert-butyl-2-(6-chloro-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol or a pharmaceutically acceptable salt thereof.

3. The compound of claim 1 , where the compound is 6-tert-butyl-4-chloro-2-(6-chloro-1,1-dioxo-1,2-dihydro-1λ 6 -benzo[1,2,4]thiadiazin-3-yl)-3-methyl-phenol or a pharmaceutically acceptable salt thereof.

4. A pharmaceutical composition comprising a compound of claim 1 and a pharmaceutically acceptable carrier or diluent.

5. A method of treating obesity comprising administering to a human subject a compound of claim 1 .
