Abstract

The invention relates to novel binding domain-immunoglobulin fusion proteins that feature a binding domain for a cognate structure such as an antigen, a counterreceptor or the like, a wild-type IgG1, IGA or IgE hinge region polypeptide or a mutant IgG1 hinge region polypeptide having either zero, one or two cysteine residues, and immunoglobulin CH2 and CH3 domains, and that are capable of ADCC and/or CDC while occurring predominantly as polypeptides that are compromised in their ability to form disulfide-linked multimers. The fusion proteins can be recombinantly produced at high express levels. Also provided are related compositions and methods, including cell surface forms of the fusion proteins and immunotherapeutic applications of the fusion proteins and of polynucleotides encoding such fusion proteins.

Claims (33)

1. A single chain binding domain-immunoglobulin fusion protein that binds human CD37, comprising from N-terminus to C-terminus:

(a) a human CD37-specific single chain Fv (scFv) binding domain polypeptide;

(b) a human IgG1 hinge region polypeptide comprising a mutated human IgG1 hinge peptide as set forth in amino acids 269-283 of SEQ ID NO:397, 398, 582, or 586; and

(c) human IgG1 heavy chain constant region domains CH2 and CH3.

2. The fusion protein of claim 1 , wherein the scFv binding domain polypeptide comprises a heavy chain variable region of CD37-specific monoclonal antibody G28-1.

3. The fusion protein of claim 1 , wherein the scFv binding domain polypeptide comprises a light chain variable region of CD37-specific monoclonal antibody G28-1.

4. The fusion protein of claim 1 , wherein the scFv binding domain polypeptide comprises a light chain variable region and a heavy chain variable region of CD37-specific monoclonal antibody G28-1.

5. The fusion protein of claim 4 , wherein the scFv binding domain polypeptide comprises amino acids 21-259 shown in SEQ ID NO:13.

6. The fusion protein of claim 1 , wherein the scFv variable domains are joined by a linker polypeptide comprising an amino acid sequence of Gly-Gly-Gly-Gly-Ser (SEQ ID NO:39).

7. The fusion protein of claim 6 , wherein the linker polypeptide comprises three repeats of the amino acid sequence as set forth in SEQ ID NO:39.

8. The fusion protein of claim 1 , wherein the scFv binding domain polypeptide is humanized.

9. The fusion protein of claim 8 , wherein the scFv binding domain polypeptide comprises a humanized heavy chain variable region of CD37-specific monoclonal antibody G28-1.

10. The fusion protein of claim 8 , wherein the scFv binding domain polypeptide comprises a humanized light chain variable region of CD37-specific monoclonal antibody G28-1.

11. The fusion protein of claim 8 , wherein the scFv binding domain polypeptide comprises a humanized light chain variable region and a humanized heavy chain variable region of CD37-specific monoclonal antibody G28-1.

12. The fusion protein of claim 8 , wherein the scFv binding domain polypeptide is a humanized amino acid sequence of amino acids 21-259 shown in SEQ ID NO:13.

13. The fusion protein of claim 1 , wherein the human IgG1 hinge region polypeptide comprises the mutated human IgG1 hinge peptide as set forth in amino acids 269 to 283 of SEQ ID NO:586.

14. The fusion protein of claim 1 , wherein the heavy chain constant region domains CH2 and CH3 comprise amino acids 284 to 500 of SEQ ID NO:586.

15. A composition comprising a fusion protein according to any one of claims 1 - 14 and a pharmaceutically acceptable excipient.

16. A single chain binding domain-immunoglobulin fusion protein that binds human CD37, comprising from N-terminus to C-terminus:

(a) a human CD37-specific single chain Fv (scFv) binding domain polypeptide, wherein the scFv binding domain polypeptide comprises an immunoglobulin light chain variable region as set forth in amino acids 21-127 of SEQ ID NO:13 and an immunoglobulin heavy chain variable region as set forth in amino acids 144-259 of SEQ ID NO:13;

(b) a human IgG1 hinge region polypeptide comprising amino acids 269-283 of SEQ ID NO:586;

(c) an immunoglobulin heavy chain CH2 constant region polypeptide as set forth in amino acids 284-393 of SEQ ID NO:586-, and

(d) an immunoglobulin heavy chain CH3 constant region polypeptide as set forth in amino acids 394-500 of SEQ ID NO:586.

17. The fusion protein of claim 16 , wherein the scFv binding domain polypeptide comprises amino acids 21-259 of SEQ ID NO:13.

18. A single chain binding domain-immunoglobulin fusion protein that binds human CD37, comprising from N-terminus to C-terminus:

(a) a human CD37-specific single chain Fv (scFv) binding domain polypeptide, wherein the scFv binding domain polypeptide comprises a sequence comprising complementary determining regions of amino acids 44-54, 70-76, and 109-117 of an immunoglobulin light chain variable region as set forth in amino acids 21-127 of SEQ ID NO:13, fused via a linker polypeptide to a sequence comprising complementary determining regions of amino acids 174-178, 195-211, and 244-250 of an immunoglobulin heavy chain variable region as set forth in amino acids 144-259 of SEQ ID NO:13, wherein the linker polypeptide comprises amino acids 128-142 of SEQ ID NO:13,

(b) a human IgG1 hinge region polypeptide comprising amino acids 269-283 of SEQ ID NO:586;

(c) an immunoglobulin heavy chain CH2 constant region polypeptide as set forth in amino acids 284-393 of SEQ ID NO:586, and

(d) an immunoglobulin heavy chain CH3 constant region polypeptide as set forth in amino acids 394-500 of SEQ ID NO:586.

19. A composition comprising a fusion protein of claim 16 or 17 and a pharmaceutically acceptable excipient.

20. The fusion protein of claim 1 , wherein the human IgG1 hinge region polypeptide comprises the mutated human IgG1 hinge peptide as set forth in amino acids 269 to 283 of SEQ ID NO:397.

21. The fusion protein of claim 1 , wherein the human IgG1 hinge region polypeptide comprises the mutated human IgG1 hinge peptide as set forth in amino acids 269 to 283 of SEQ ID NO:398.

22. The fusion protein of claim 1 , wherein the human IgG1 hinge region polypeptide comprises the mutated human IgG1 hinge peptide as set forth in amino acids 269 to 283 of SEQ ID NO:582.
