Rapid setting high strength calcium phosphate cements comprising cyclodextrins
View Patent ↗Rapid setting high strength calcium phosphate cements and methods of using the same are provided. Aspects of the cements include fine and coarse calcium phosphate particulate reactants and a cyclodextrin which, upon combination with a setting fluid, produce a flowable composition that rapidly sets into a high strength product. The flowable compositions find use in a variety of different applications, including the repair of hard tissue defects, e.g., bone defects such as fractures.
1. A method of producing a flowable composition that sets into a calcium phosphate containing product, the method comprising:
combining:
(a) a dry reactant component comprising:
(i) a first tricalcium phosphate particulate calcium phosphate reactant having a mean particle size of 8 μm or less;
(ii) a second calcium phosphate reactant having a tricalcium phosphate coarse particulate composition comprising tricalcium phosphate particles that range in size from 30 to 250 μm, wherein the tricalcium phosphate particles comprise sodium in an amount ranging from 1500 to 2500 ppm;
(b) a α-cyclodextrin in an amount ranging from 0.05 to 2.0% w/w of the dry reactants; and
(c) a setting fluid;
in a ratio sufficient to produce the flowable composition that sets into a calcium phosphate containing product.
2. The method according to claim 1 , wherein the mass ratio of the first to second particulate calcium phosphate reactants in the dry reactant component ranges from 1 to 10.
3. The method according to claim 1 , wherein the first particulate calcium phosphate reactant has a mean particle size of 4 μm.
4. The method according to claim 1 , wherein the α-cyclodextrin and the setting fluid are mixed prior to being mixed with the dry reactant component.
5. The method according to claim 1 , wherein the α-cyclodextrin and the dry reactant component are mixed prior to being mixed with the setting fluid.
6. The method according to claim 1 , wherein the flowable composition has a 4-minute setting strength of 1000 N or greater.
7. The method according to claim 1 , wherein the calcium phosphate product has a 24-hour tensile strength of 5 MP or greater.
8. The method according to claim 1 , wherein the setting fluid is a solution of a soluble silicate.
9. A method of repairing a hard tissue defect, the method comprising:
(a) combining:
(i) a dry reactant component comprising:
a first tricalcium phosphate particulate calcium phosphate reactant having a mean particle size of 8 μm or less; and
a second calcium phosphate reactant having a tricalcium phosphate coarse particulate composition comprising tricalcium phosphate particles that range in size from 30 to 250 μm, wherein the tricalcium phosphate particles comprise sodium in an amount ranging from 1500 to 2500 ppm;
(ii) a cyclodextrin in an amount ranging from 0.05 to 2.0% w/w of the dry reactants; and
(iii) a setting fluid;
in a ratio sufficient to produce flowable composition that sets into a calcium phosphate containing product; and
(b) applying the flowable composition to the defect in a manner sufficient to repair the hard tissue defect.
10. The method according to claim 1 , wherein the tricalcium phosphate particles of the second calcium phosphate reactant comprise phosphate, potassium, calcium, magnesium, zinc, copper, manganese, iron, sulfur and sodium.
