Compositions employing alternative reading frame polypeptides for the treatment of cancer and infectious disease
View Patent ↗Provided are alternative reading frame (ARF) polypeptides as well as antigen presenting cell (APC) and dendritic cell (DC) based compositions that comprise ARF polypeptides. ARF polypeptides and ARF polypeptide-based compositions are useful in the treatment of cancer and infectious disease.
1. A polypeptide conjugate comprising an N-terminal moiety and a C-terminal moiety, wherein said N-terminal moiety comprises one or more ARF polypeptides and said C-terminal moiety comprises a polypeptide moiety that facilitates binding of said ARF polypeptide to an antigen presenting cell (APC),
and wherein each said ARF polypeptide comprises at least 9 amino acids of an ARF polypeptide encoded by a human tyrosine kinase receptor (hHER-2) polynucleotide represented by SEQ ID NO: 2 and has a sequence selected from SEQ ID NOs: 71-95 and fragments thereof comprising at least 9 amino acids.
2. The polypeptide conjugate of claim 1 , wherein said polypeptide moiety that facilitates binding is selected from the group consisting of GM-CSF, IL-1, TNF, IL-4, CD40L, CTLA4, CD28, FLT-3 ligand, and a functional fragment, derivative or variant thereof.

An immunogenic composition comprising an antigen presenting cell (APC) pulsed ex vivo with a fusion protein comprising two or more isolated ARF polypeptides, wherein each said ARF polypeptide comprises at least 9 amino acids of an ARF polypeptide encoded by a human tyrosine kinase receptor (hHER-2) polynucleotide represented by SEQ ID NO: 2, and has a sequence selected from SEQ ID NOs: 71-95 and fragments thereof comprising at least 9 amino acids.

An immunogenic composition comprising an antigen presenting cell (APC) pulsed ex vivo with a protein conjugate comprising an N-terminal moiety and a C-terminal moiety, wherein said N-terminal moiety comprises one or more ARF polypeptides and said C-terminal moiety comprises a polypeptide moiety that facilitates binding of said ARF polypeptide to an antigen presenting cell (APC),
wherein each said ARF polypeptide comprises at least 9 amino acids of an ARF polypeptide encoded by a human tyrosine kinase receptor (hHER-2) polynucleotide represented SEQ ID NO: 2 and has a sequence selected from SEQ ID NOs: 71-95 and fragments thereof comprising at least 9 amino acids.
