Abstract

This invention provides tandem fluorescent protein construct including a donor fluorescent protein moiety, an acceptor fluorescent protein moiety and a linker moiety that couples the donor and acceptor moieties. The donor and acceptor moieties exhibit fluorescence resonance energy transfer which is eliminated upon cleavage. The constructs are useful in enzymatic assays.

Claims (26)

1. A method for determining whether a sample contains an enzyme comprising:

contacting the sample with a recombinant tandem fluorescent protein construct comprising:

a polypeptide chain having a donor fluorescent protein moiety,

an acceptor fluorescent protein moiety, and

a peptide linker moiety that couples the donor and acceptor moieties,

wherein the donor and acceptor moieties exhibit fluorescence resonance energy transfer when the donor moiety is excited, wherein the donor moiety and acceptor moiety are Aequorea fluorescent protein moieties, wherein the linker moiety consists of between 5 amino acids and 50 amino acids, wherein the donor and acceptor Aequorea fluorescent protein moieties together have a contiguous sequence of 150 amino acids with at least 85% sequence identity with a contiguous amino acid sequence of 150 amino acids from the wild type Aequorea green fluorescent protein of SEQ ID NO:2, wherein at least one of the donor and acceptor protein moieties comprises a mutation selected from the group consisting of F64L, S65G, V68L, S72A, N149K, I167T, T203I, and T203Y, and wherein said linker moiety does not substantially emit light to excite said donor fluorescent protein moiety;

exciting the donor moiety; and

determining the degree of fluorescence resonance energy transfer in the sample, whereby a degree of fluorescence resonance energy transfer that is lower than an expected amount indicates the presence of an enzyme.

The method of claim 1 , wherein determining the degree of fluorescence resonance energy transfer in the sample comprises determining the degree at a first and second time after contacting the sample with a tandem fluorescent protein construct, and determining the difference in the degree of fluorescence resonance energy transfer, whereby the difference in the degree of fluorescence resonance energy transfer reflects the amount of enzyme in the sample.

3. The method of claim 1 , wherein the linker moiety consists of between 12 amino acids and 30 amino acids.

4. The method of claim 1 , wherein the linker moiety contains a cleavage site.

5. The method of claim 4 , wherein the cleavage site comprises an amino acid sequence selected from SEQ ID NOS:3 to 14.

6. The method of claim 1 , wherein both the donor and acceptor protein moieties comprise a mutation selected from the group consisting of F64L, S65G, V68L, S72A, N149K, I167T, T203I, and T203Y.

7. The method of claim 1 , wherein at least one of the donor and acceptor protein moieties comprises S65G, V68L, V72A, and T203Y mutations.

8. The method of claim 1 , wherein at least one of the donor and acceptor protein moieties comprises F64L, S65T, Y66W, N146I, M153T, V163A, and N212K mutations.

9. The method of claim 1 , wherein at least one of the donor and acceptor protein moieties comprises S65T, S72A, N149K, M153T, and I167T mutations.

10. The method of claim 1 , wherein at least one of the donor and acceptor protein moieties comprises S72A, Y145F, and T203I mutations.

The method of claim 1 , wherein the donor and acceptor Aequorea fluorescent protein moieties together have a contiguous sequence of 150 amino acids with at least 95% sequence identity with a contiguous amino acid sequence of 150 amino acids from the wild type Aequorea green fluorescent protein of SEQ ID NO:2.

12. The method of claim 11 , wherein the linker moiety consists of between 12 amino acids and 30 amino acids.

13. The method of claim 11 , wherein the linker moiety contains a cleavage site.

14. The method of claim 13 , wherein the cleavage site comprises an amino acid sequence selected from SEQ ID NOS:3 to 14.

15. The method of claim 11 , wherein both the donor and acceptor protein moieties comprise a mutation selected from the group consisting of F64L, S65G, V68L, S72A, N149K, I167T, T203I, and T203Y.

16. The method of claim 11 , wherein at least one of the donor and acceptor protein moieties comprises S65G, V68L, V72A, and T203Y mutations.

17. The method of claim 11 , wherein at least one of the donor and acceptor protein moieties comprises F64L, S65T, Y66W, N146I, M153T, V163A, and N212K mutations.

18. The method of claim 11 , wherein at least one of the donor and acceptor protein moieties comprises S65T, S72A, N149K, M153T, and I167T mutations.

19. The method of claim 11 , wherein at least one of the donor and acceptor protein moieties comprises S72A, Y145F, and T203I mutations.
