Abstract

The invention provides peptides that bind Tissue Factor Pathway Inhibitor (TFPI), including TFPI-inhibitory peptides, and compositions thereof. The peptides may be used to inhibit a TFPI, enhance thrombin formation in a clotting factor-deficient subject, increase blood clot formation in a subject, and/or treat a blood coagulation disorder in a subject.

Claims (28)

1. A peptide that binds TFPI, comprising the following structure of formula (I):

X1001-X1002-X1003-X1004-X1005-X1006-X1007-X1008-X1009-X1010-X1011-X1012-X1013-X1014-X1015-X1016-X1017-X1018-X1019-X1020  (I) (SEQ ID NO: 3116):

wherein X1001 is an amino acid selected from the group consisting of β-Homophenylalanine (Bhf), C, D, F, G, H, I, K, L, M, N, (S)—N-Methyl-phenylalanine (Nmf), Q, R, T, V, W and Y;

wherein X1002 is an amino acid selected from the group consisting of G and Q;

wherein X1003 is an amino acid selected from the group consisting of A, 2-Amino-isobutyric acid (Aib), β-Homoserine (Bhs), C, D, E, F, G, H, I, K, L, M, N, P, Q, R, S, T, V, W and Y;

wherein X1004 is an amino acid selected from the group consisting of A, Aib, β-Homolysine (Bhk), C, D, E, F, G, H, I, K, k, L, M, N, (S)—N-Methyl-lysine (Nmk), P, Q, R, S, T, V, W and Y;

wherein X1005 is an amino acid selected from the group consisting of a, A, Aib, β-Alanine (Bal), C, D, D-Aspartic Acid (d), E, F, G, H, K, k, L, M, N, N-Methyl-glycine (Nmg), D-Proline (p), Q, R, S, T, V, W and Y;

wherein X1006 is an amino acid selected from the group consisting of A, Aib, β-Homoasparagin (Btq), C, D, E, F, G, H, I, K, L, M, N, Q, R, S T, V, W and Y;

wherein X1007 is an amino acid selected from the group consisting of A, F, I, K, L, (S)—N-Methyl-valine (Nmv), P, Q, S, V, W and Y;

wherein X1008 is an amino acid selected from the group consisting of F, H, K, W and Y;

wherein X1009 is an amino acid selected from the group consisting of A, Aib, D-Phenylalanine (f), I, K, S, T and V;

wherein X1010 is an amino acid selected from the group consisting of A, Aib, C, D, E, F, G, H, I, K, L, M, N, Nmf, P, Q, R, S, T, V, W and Y;

wherein X1011 is an amino acid selected from the group consisting of Aib, C, K, G and Nmg;

wherein X1012 is Y;

wherein X1013 is an amino acid selected from the group consisting of A, Aib, C, E, F, G, H, K, L, M, Q, R, W and Y;

wherein X1014 is an amino acid selected from the group consisting of A, Aib, β-Homoglutamatic acid (Bhe), C, D, E, F, G, H, I, K, L, M, N, P, Q, R, S, T, V, W and Y;

wherein X1015 is an amino acid selected from the group consisting of (omega-methyl)-R, D, E, K and R;

wherein X1016 is L;

wherein X1017 is an amino acid selected from the group consisting of (omega-methyl)-R, A, Aib, β-Homoarginine (Bhr), C, (S)-Cyclohexylalanine (Cha), (S)-Citrullin (Cit), D, (S)-2,4-Diaminobutyric acid (Dab), (S)-Diaminopropionic acid (Dap), E, (S)-2-Propargylglycine (Eag), (S)—N(omega)-nitro-arginine (Eew), F, G, H, (S)-Homo-arginine (Har), (S)-Homo-citrulline (Hci), (S)-2-Amino-5-methyl-hexanoic acid (Hle), I, K, L, M, N, (S)-Norleucine (Nle), (S)-2-Amino-pentanoic acid (Nva), (S)-2-Pyridyl-alanine (Opa), (S)-Ornithine (Orn), Q, R, S, T, V, W and Y;

wherein X1018 is an amino acid selected from the group consisting of A, Bal, C, D, E, F, G, H, I, K, L, M, N, Q, R, S, T, V, W and Y;

wherein X1019 is an amino acid selected from the group consisting of Bhk, K, R and V; and

wherein X1020 is either present or absent, whereby in case X1020 is present it is an amino acid selected from the group consisting of Aib, β-Homoleucine (Bhl), C, F, G, H, I, K, L, (S)—N-Methyl-leucine (Nml), Q, R, S, T, V, W and Y.

2. A peptide that binds TFPI, comprising the following structure of formula (III):

X1001-Q-X1003-X1004-X1005-X1006-I/V-X1008-V-X1010-G-Y-C/F-X1014-R-L-X1017-X1018-K-K/L  (III) (SEQ ID NO: 3117)

wherein X1001, X1003, X1004, X1005, X1006, X1008, X1010, X1014, X1017 and X1018 are each independently selected from any amino acid.

The peptide of claim 1 , the peptide consisting of the amino acid sequence selected from the group consisting of SEQ ID NOs: 16-123, 125-127, 129-178, 183-188, 190-252, 256-270, 272-284, 286-405, 407-422, 424-428, 430-434, 436-452, 455-458, 461, 468, 471, 472, 474, 475, 478-480, 482-484, 486-488, 490, 491, 493-498, 500-502, 505-507, 509, 511-519, 523, 524, 527, 530, 532, 533, 536, 538-540, 542, 545, 548-550, 553-559, 561, 563-565, 568-572, 576, 577, 579-582, 584-628, 630, 632, 633, 635-648, 650-658, 660-671, 690-693, 698, 700-709, 720, 724, 725, 731, 736-740, 747, 749, 765, 767-769, 774, 783, 785-788, 791, 795, 807, 811, 817, 818, 823, 829, 835, 851, 858, 872-874, 876, 893, 926, 938, 944, 952, 953, and 972, 973, and 975-978.

4. A method for treating a blood coagulation disorder in a subject, the method comprising administering to the subject a peptide that inhibits TFPI activity, wherein the peptide is administered in an amount sufficient to treat the blood coagulation disorder in the subject.

5. The method according to claim 4 , wherein the peptide is a peptide as defined in claim 1 .
