Abstract

Peptide-peptidase inhibitor conjugate molecules and methods of manufacture thereof are disclosed. These conjugate molecules are useful as agents for the treatment and prevention of metabolic and cardiovascular diseases, disorders, and conditions. Such diseases, conditions and disorders include, but are not limited to, hypertension, dyslipidemia, cardiovascular disease, eating disorders, insulin-resistance, obesity, and diabetes mellitus of any kind, and other diabetes-related disorders.

Claims (30)

1. A method for synthesizing a peptide-peptidase inhibitor conjugate having peptide comprising the structure: peptide-linker-peptidase inhibitor, wherein the linker is optional; wherein the method comprises:

a) coupling a peptidase inhibitor to a resin under conditions suitable to form a resin-bound peptidase inhibitor, wherein the peptidase inhibitor comprises a peptidase inhibitor core and an initial linker covalently hound thereto, wherein the peptidase inhibitor comprises a first amine and a second amine, and wherein the peptidase inhibitor is coupled to the resin through the first amine;

b) optionally coupling at the second amine an extended linker to form the optional linker, wherein the extended linker comprises an amine which replaces the second amine;

c) coupling the peptide, one or more amino acids, or one or more fragments of the peptide at the second amine under conditions suitable to form the peptide-peptidase inhibitor conjugate; and

d) cleaving the peptide-peptidase inhibitor conjugate from the resin, thereby providing the peptide-peptidase inhibitor conjugate; wherein the peptide comprises the amino acid sequence of any one of SEQ ID NOs:1-189, 190-232, 268, 269, and 274.

2. The method of claim 1 , wherein the initial linker is coupled to an extended linker prior to coupling with the peptidase inhibitor core, wherein the extended linker comprises an amine which replaces the second amine of the peptidase inhibitor, thereby providing the optional linker.

The method according to claim 1 , wherein the peptidase inhibitor is selected from the group consisting of an inhibitor of angiotensin converting enzyme, vasopeptidase, neuraminadase, plasma peptidase, endothelial converting enzyme, human neutral endopeptidase, dipeptidyl peptidase IV, aminopeptidase, a cysteine protease, a serine protease, and a carboxypeptidase.

4. The method of claim 1 , wherein the peptidase inhibitor is a dipeptidyl peptidase IV inhibitor.

5. The method of claim 4 , wherein the dipeptidyl peptidase IV inhibitor is selected from the group consisting of thiazolidine, cyanopyrrolidine and pyrrolidine-3-sulfoxide.

6. The method of claim 1 , wherein the initial linker comprises a diamino acid.

7. The method of claim 6 , wherein the diamino acid is selected from the group consisting of homohomolysine, homolysine, lysine, ornithine, diaminobutyric acid, and diamino propionic acid.

8. A method for synthesizing a peptide-peptidase inhibitor conjugate having peptide comprising the structure: peptide-linker-peptidase inhibitor, wherein the linker is optional; wherein the method comprises:

a) coupling a peptidase inhibitor to a resin under conditions suitable to form a resin-bound peptidase inhibitor, wherein the peptidase inhibitor comprises a peptidase inhibitor core and an initial linker covalently bound thereto, wherein the peptidase inhibitor comprises a first amine and a second amine, and wherein the peptidase inhibitor is coupled to the resin through the first amine;

b) optionally coupling at the second amine an extended linker to form the optional linker, wherein the extended linker comprises an amine which replaces the second amine;

c) coupling the peptide, one or more amino acids, or one or more fragments of the peptide at the second amine under conditions suitable to form the peptide-peptidase inhibitor conjugate; and

d) cleaving the peptide-peptidase inhibitor conjugate from the resin, thereby providing the peptide-peptidase inhibitor conjugate, wherein the initial linker is formed by reaction of the peptidase inhibitor core with Boc-Lys-Fmoc-OH.

9. The method of claim 1 , wherein said initial linker is attached to an amine group of the peptidase inhibitor core.

10. The method of claim 1 , wherein the peptidase inhibitor is attached to a carboxylic acid group, an amine group, or a sulfyhdryl group of the peptide in the peptide-peptidase inhibitor conjugate.

11. A method for synthesizing a peptide-peptidase inhibitor conjugate having peptide comprising the structure: peptide-linker-peptidase inhibitor, wherein the linker is optional; wherein the method comprises:

a) coupling a peptidase inhibitor to a resin under conditions suitable to form a resin-bound peptidase inhibitor, wherein the peptidase inhibitor comprises a peptidase inhibitor core and an initial linker covalently bound thereto, wherein the peptidase inhibitor comprises a first amine and a second amine, and wherein the peptidase inhibitor is coupled to the resin through the first amine;

b) optionally coupling at the second amine an extended linker to form the optional linker, wherein the extended linker comprises an amine which replaces the second amine;

c) coupling the peptide, one or more amino acids, or one or more fragments of the peptide at the second amine under conditions suitable to form the peptide-peptidase inhibitor conjugate; and

d) cleaving the peptide-peptidase inhibitor conjugate from the resin, thereby providing the peptide-peptidase inhibitor conjugate, wherein the linker comprises a diamino acid.

12. The method of claim 1 , wherein the linker comprises one or more polyethylene oxide monomers.

The method of claim 1 , wherein the linker is selected from the group consisting of 8-amino-3,6-dioxo octanoic acid, [β-Ala] x1 wherein x1 is 1 to 5,[Gly] x2 wherein x2 is 1 to 5, [[Gly] x3 -Xaa] y3 wherein x3 is 1 to 5, y3 is 1 to 5, and wherein Xaa is any naturally-occuring amino acid, and [g-Abu] x4 where x4 is 1 to 5.

14. The method of claim 11 , wherein the resin is p-nitrophenyl carbonate Wang resin.

15. The method of claim 11 , wherein the peptide is selected from the group consisting of an incretin, an amylinomimetic, a glucose-dependent insulinotrope, a leptin, a cholecystokinin, a pancreatic polypeptide, and an analog thereof.

16. The method of claim 11 , wherein the peptide is selected from the group consisting of GLP-1, GLP-2, GIP, exendin-3, exendin-4, PYY, amylin, calcitonin, leptin, and an analog thereof.

17. The method of claim 11 , wherein the peptidase inhibitor is a dipeptidyl peptidase IV inhibitor, and wherein the peptide is selected from the group consisting of GLP-1, GIP, PYY, NPY and exendin-4, or an analog or derivative thereof.

18. The method of claim 11 , wherein the peptidase inhibitor is a dipeptidyl peptidase IV inhibitor or an angiotensin converting enzyme inhibitor, and wherein the peptide comprises the amino acid sequence of any one of SEQ ID NOs: 1-189, 190-232, 268, 269, and 274.
