Abstract

The present invention relates to novel humanized anti-IL-4 and IL-13 antibodies and fragments thereof and novel bispecific antibodies and fragments thereof that specifically bind to IL-4 and IL-13. The invention also includes uses of the antibodies to treat or prevent IL-4 and/or IL-13 mediated diseases or disorders, including allergic asthma and dermatitis.

Claims (24)

1. A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL-4, wherein said bispecific antibody or bispecific antibody fragment thereof comprises a variable light chain domain and a variable heavy chain domain, wherein said variable light chain domain comprises amino acid sequences SEQ ID NO:1 and SEQ ID NO:3.

2. The bispecific antibody or bispecific antibody fragment thereof of claim 1 , wherein said variable heavy chain domain comprises amino acid sequences SEQ ID NO:2 and SEQ ID NO:5.

3. The bispecific antibody or bispecific antibody fragment thereof of claim 1 , wherein said variable heavy chain domain comprises amino acid sequences SEQ ID NO:2 and SEQ ID NO:4.

4. A pharmaceutical composition comprising the bispecific antibody or bispecific antibody fragment thereof of claim 1 and a pharmaceutically acceptable carrier.

A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL-4, wherein said bispecific antibody or bispecific antibody fragment thereof comprises a variable light chain domain comprising amino acid sequences SEQ ID NO:1 and SEQ ID NO:3, and a variable heavy chain domain comprising amino acid sequences SEQ ID NO:2 and SEQ ID NO:4.

6. The bispecific antibody or bispecific antibody fragment thereof of claim 5 , wherein a peptide linker links SEQ ID NO:1 to SEQ ID NO:3, and a peptide linker links SEQ ID NO:2 to SEQ ID NO:4.

7. The bispecific antibody or bispecific antibody fragment thereof of claim 6 , wherein said peptide linker has an amino acid sequence consisting of SEQ ID NO:6.

8. A pharmaceutical composition comprising the bispecific antibody or bispecific antibody fragment thereof of claim 5 and a pharmaceutically acceptable carrier.

9. A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL-4, comprising:

(a) a variable light chain domain comprising the amino acid sequences of SEQ ID NO:1 and SEQ ID NO:3;

(b) a variable heavy chain domain comprising the amino acid sequences of SEQ ID NO:2 and SEQ ID NO:4; and

(c) constant region domains.

10. The bispecific antibody or bispecific antibody fragment thereof of claim 9 , wherein a peptide linker links SEQ ID NO:1 to SEQ ID NO:3, and a peptide linker links SEQ ID NO:2 to SEQ ID NO:4.

11. The bispecific antibody or bispecific antibody fragment thereof of claim 10 , wherein said peptide linker has an amino acid sequence consisting of SEQ ID NO:6.

12. The bispecific antibody or bispecific antibody fragment thereof of claim 11 , wherein said constant region domains consist of CH1, CH2, CH3 and CL.

13. The bispecific antibody or bispecific antibody fragment thereof of claim 9 , wherein said constant region domains consist of CH1, CH2, CH3 and CL.

14. A pharmaceutical composition comprising the bispecific antibody or bispecific antibody fragment thereof of claim 13 and a pharmaceutically acceptable carrier.

A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL-4, wherein said bispecific antibody or bispecific antibody fragment thereof comprises a variable light chain domain comprising amino acid sequences SEQ ID NO:1 and SEQ ID NO:3, and a variable heavy chain domain comprising amino acid sequences SEQ ID NO:2 and SEQ ID NO:5.

A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL-4, wherein the bispecific antibody or bispecific antibody fragment thereof comprises the amino acid sequences of RASESVDSYGQSYMH (SEQ ID NO: 8), LASNLES (SEQ ID NO: 9), QQNAEDSRT (SEQ ID NO: 10), HASQNIDVWLS (SEQ ID NO: 14), KASNLHTG (SEQ ID NO: 15), QQAHSYPFT (SEQ ID NO: 16), GFSLTDSSIN (SEQ ID NO: 11), DGRID (SEQ ID NO: 12), DGYFPYAMDF (SEQ ID NO: 13), GYSFTSYWIH (SEQ ID NO: 17), IDPSDGETR (SEQ ID NO: 18) and LKEYGNYDSFYFDV (SEQ ID NO: 19).

17. A bispecific antibody or bispecific antibody fragment thereof that specifically binds to IL-13 and IL4, wherein the bispecific antibody or bispecific antibody fragment thereof comprises the amino acid sequences of RASESVDSYGQSYMH (SEQ ID NO: 8), LASNLES (SEQ ID NO: 9), QQNAEDSRT (SEQ ID NO: 10), HASQNIDVWLS (SEQ ID NO: 14), KASNLHTG (SEQ ID NO: 15), QQAHSYPFT (SEQ ID NO: 16), GFSLTDSSIN (SEQ ID NO: 11), DGRID (SEQ ID NO: 12), DGYFPYAMDF (SEQ ID NO: 13), GYSFTSYWIH (SEQ ID NO: 20), IDASDGETR (SEQ ID NO: 21), and LKEYGNYDSFYFDV (SEQ ID NO: 22).

18. The bispecific antibody or bispecific antibody fragment thereof of any one of claims 15 - 17 that further comprises constant region domains.

19. The bispecific antibody or bispecific antibody fragment thereof of claim 18 , wherein said constant region domains consist of CH1, CH2, CH3 and CL.

20. A pharmaceutical composition comprising the bispecific antibody or bispecific antibody fragment thereof of any one of claims 15 - 17 and a pharmaceutically acceptable carrier.

21. A pharmaceutical composition comprising the bispecific antibody or bispecific antibody fragment thereof of claim 19 and a pharmaceutically acceptable carrier.
