View Patent ↗The present invention relates to isolated polypeptides having beta-glucosidase activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods for producing and using the polypeptides.
1. A method for degrading cellulose- and hemicellulose-containing biomass, comprising treating the biomass with an effective amount of a polypeptide having beta-glucosidase activity and recovering the degraded biomass, wherein the polypeptide having beta-glucosidase activity is selected from the group consisting of:
(a) a polypeptide comprising an amino acid sequence which has at least 95% identity with amino acids 20 to 863 of SEQ ID NO: 2; and
(b) a polypeptide which is encoded by a polynucleotide which hybridizes under at least high stringency conditions with (i) nucleotides 58 to 2580 of SEQ ID NO: 1, (ii) the cDNA sequence contained in nucleotides 58 to 2580 of SEQ ID NO: 1, or (iii) the complement of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 65° C.
2. The method of claim 1 , wherein the polypeptide having beta-glucosidase activity comprises an amino acid sequence which has at least 95% identity with amino acids 20 to 863 of SEQ ID NO: 2.

4. The method of claim 1 , wherein the polypeptide having beta-glucosidase activity comprises the amino acid sequence of SEQ ID NO: 2.
5. The method of claim 1 , wherein the polypeptide having beta-glucosidase activity consists of SEQ ID NO: 2 or a fragment thereof having beta-glucosidase activity.
6. The method of claim 5 , wherein the polypeptide having beta-glucosidase activity consists of amino acids 20 to 863 of SEQ ID NO: 2.
7. The method of claim 1 , wherein the polypeptide having beta-glucosidase activity is encoded by a polynucleotide which hybridizes under at least high stringency conditions with (i) nucleotides 58 to 2580 of SEQ ID NO: 1, (ii) the cDNA sequence contained in nucleotides 58 to 2580 of SEQ ID NO: 1, or (iii) the full-length complement of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 65° C.
