Abstract

Compositions and methods for producing aldehydes, alkanes, and alkenes are described herein. The aldehydes, alkanes, and alkenes can be used in biofuels.

Claims (15)

1. A method of producing an alkane or alkene from an engineered microorganism, the method comprising,

expressing a first gene encoding an acyl-ACP reductase that catalyzes the conversion of acyl ACP to a fatty aldehyde in said engineered microorganism;

expressing a second gene encoding a decarbonylase that catalyzes the conversion of a fatty aldehyde to an alkane or alkene in said engineered microorganism;

culturing said engineered microorganism in a culture media containing a carbohydrate carbon source under conditions effective to produce an alkane or alkene in the cell-free supernatant.

2. The method according to claim 1 , wherein said acyl-ACP reductase has a higher affinity for acyl-ACP as compared to acyl-CoA.

3. The method of claim 1 , wherein the engineered microorganism is selected from the group consisting of a yeast, a fungi, a filamentous fungi, an algae and a bacteria.

4. The method of claim 3 , wherein the bacteria is E. coli.

5. The method of claim 1 , wherein the alkane or alkene comprises a C 13 to C 21 alkane or alkene.

A cell culture comprising an engineered microorganism wherein an acyl-ACP reductase that catalyzes the conversion of acyl ACP to a fatty aldehyde and a decarbonylase that catalyzes the conversion of a fatty aldehyde to an alkane or alkene are overexpressed in said engineered microorganism and an alkane or alkene is produced in the cell culture when said engineered microorganism is cultured in medium containing a carbon source under conditions effective to express said acyl-ACP reductase and said decarbonylase.

7. The cell culture according to claim 6 , wherein the engineered microorganism is selected from the group consisting of a yeast, a fungi, a filamentous fungi, an algae and a bacteria.

8. The cell culture according to claim 7 , wherein the bacteria is E. coli.

9. The cell culture according to claim 6 , wherein the alkane or alkene comprises a C 13 to C 21 alkane or alkene.

The cell culture according to claim 6 , wherein the alkane or alkene is selected from the group consisting of tridecane, methyltridecane, nonadecane, methylnonadecane, heptadecane, methylheptadecane, pentadecane, methylpentadecane, pentadecene, heptadecene, methylpentadecane, and methylheptadecene.

11. The cell culture according to claim 6 , comprising an alkane or alkene having a δ 13 C of about −15.4 or greater.

12. The cell culture according to claim 6 , comprising an alkane or alkene having an f M 14 C of at least about 1.003.
