Fusion antibodies
View Patent ↗The present invention provides novel antibodies. In particular, the present invention provides fusion antibodies comprising antibody heavy and light chain fusions. The present invention further provides multivalent antibodies comprising multiple fusion antibody chains. The present invention further provides methods of generating splice resistant antibody genes.
1. A composition comprising a multivalent antigen binding protein comprising at least two polypeptides, wherein each of said polypeptides comprises an antibody heavy chain comprising variable and Fc regions encoded by an antibody heavy chain gene from a parental antibody fused to an antibody light chain encoded by an antibody light chain gene from said parental antibody via a peptide linker so that said heavy and said light chain are expressed as a single polypeptide chain, wherein said antibody heavy chain has a C-terminus and said antibody light chain is fused at said C-terminus to said antibody heavy chain via said peptide linker, wherein said at least two polypeptides are joined via di-sulfide bonds.
2. The composition of claim 1 , wherein said multivalent antigen binding protein comprises at least 5 of said polypeptides.
3. The composition of claim 1 , wherein said multivalent antibody comprises at least 10 of said polypeptides.
4. The composition of claim 1 , wherein said multivalent antibody comprises at least 15 of said polypeptides.

A composition comprising a multivalent antigen binding protein comprising at least five polypeptides, wherein each of said polypeptides comprises an antibody heavy chain comprising variable and Fc regions encoded by an antibody heavy chain gene from a parental antibody fused to an antibody light chain encoded by an antibody light chain gene from said parental antibody via a peptide linker so that said heavy and said light chain are expressed as a single polypeptide chain, wherein said antibody heavy chain has a C-terminus and said antibody light chain is fused at said C-terminus to said antibody heavy chain via said peptide linker, wherein said at least five polypeptides are joined via di-sulfide bonds.
