Polypeptides having cellulolytic enhancing activity and nucleic acids encoding same
View Patent ↗The present invention relates to isolated polypeptides having cellulolytic enhancing activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods for producing and using the polypeptides.
1. An isolated polypeptide having cellulolytic enhancing activity, selected from the group consisting of:
(a) a polypeptide comprising an amino acid sequence which has at least 95% sequence identity to amino acids 18 to 240 of SEQ ID NO: 4;
(b) a polypeptide which is encoded by a polynucleotide which hybridizes under at least high stringency conditions with (i) nucleotides 98 to 821 of SEQ ID NO: 3, (ii) the cDNA sequence contained in nucleotides 98 to 821 of SEQ ID NO: 3, or (iii) a full-length complementary strand of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 mg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 65° C.; and
(c) a polypeptide which is encoded by a polynucleotide comprising a nucleotide sequence which has at least 95% sequence identity to nucleotides 98 to 821 of SEQ ID NO: 3.
2. The polypeptide of claim 1 , comprising an amino acid sequence which has at least 95% sequence identity to amino acids 18 to 240 of SEQ ID NO: 4.
3. The polypeptide of claim 2 , comprising an amino acid sequence which has at least 97% sequence identity to amino acids 18 to 240 of SEQ ID NO: 4.

The polypeptide of claim 1 , which is encoded by a polynucleotide which hybridizes under high stringency conditions with (i) nucleotides 98 to 821 of SEQ ID NO: 3, (ii) the cDNA sequence contained in nucleotides 98 to 821 of SEQ ID NO: 3, or (iii) a full-length complementary strand of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 mg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 65° C.

The polypeptide of claim 4 , which is encoded by a polynucleotide which hybridizes under very high stringency conditions with (i) nucleotides 98 to 821 of SEQ ID NO: 3, (ii) the cDNA sequence contained in nucleotides 98 to 821 of SEQ ID NO: 3, or (iii) a full-length complementary strand of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 mg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.
6. The polypeptide of claim 1 , which is encoded by a polynucleotide which has at least 95% sequence identity to nucleotides 98 to 821 of SEQ ID NO: 3.
7. The polypeptide of claim 6 , which is encoded by a polynucleotide which has at least 97% sequence identity to nucleotides 98 to 821 of SEQ ID NO: 3.
8. The polypeptide of claim 1 , which comprises the amino acid sequence of SEQ ID NO: 4.
9. The polypeptide of claim 1 , which comprises amino acids 18 to 240 of SEQ ID NO: 4.
10. The polypeptide of claim 1 , which consists of the amino acid sequence of SEQ ID NO: 4.
11. The polypeptide of claim 1 , which consists of amino acids 18 to 240 of SEQ ID NO: 4.
12. The polypeptide of claim 1 , which is encoded by a polynucleotide which comprises SEQ ID NO: 3.
13. The polypeptide of claim 1 , which is encoded by a polynucleotide which comprises nucleotides 98 to 821 of SEQ ID NO: 3.
14. The polypeptide of claim 1 , which is encoded by a polynucleotide which consists of SEQ ID NO: 3.
15. The polypeptide of claim 1 , which is encoded by a polynucleotide which consists of nucleotides 98 to 821 of SEQ ID NO: 3.
16. The polypeptide of claim 1 , which is encoded by the polynucleotide contained in plasmid pTter61C which is contained in Escherichia coli NRRL B-30813.
17. A detergent composition comprising the polypeptide having cellulolytic enhancing activity of claim 1 and a surfactant.
18. The composition of claim 17 , further comprising one or more of a cellulase, a protease, a lipase, a cutinase, an amylase, a carbohydrase, a pectinase, a mannanase, an arabinase, a galactanase, a xylanase, an oxidase.
19. The composition of claim 17 or 18 , which is formulated as a bar, a tablet, a powder, a granule, a paste or a liquid.
