Abstract

Novel isolated polynucleotides that encode plant transcription factors are provided, together with genetic constructs comprising such polynucleotides. Methods for using such constructs in modulating the expression of endogenous and/or heterologous genes are also disclosed, together with transgenic plants comprising such constructs.

Claims (49)

1 . An isolated polynucleotide comprising a sequence selected from the group consisting of: consisting of: SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421.

2 . An isolated polynucleotide comprising a sequence selected from the group consisting of:

(a) complements of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(b) reverse complements of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421; and

(c) reverse sequences a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421.

3 . An isolated polynucleotide comprising a sequence selected from the group consisting of:

(a) nucleotide sequences having at least 75% identity to a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(b) nucleotide sequences having at least 90% identity to a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(c) nucleotide sequence having at least 95% identity to a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(d) nucleotide sequences that hybridize to a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421 under stringent hybridization conditions;

(e) nucleotide sequences that are 200-mers of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(f) nucleotide sequences that are 100-mers of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421;

(g) nucleotide sequences that are 40-mers of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421; and

(h) nucleotide sequences that are 20-mers of a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421; and

(i) nucleotide sequences that are degeneratively equivalent to a sequence of SEQ ID NO: 1-591, 1183-1912, 1931-2106, 2371, 2374, 2377, 2385, 2387, 2389, 2391, 2393, 2395, 2397, 2399, 2401, 2403, 2405, 2407, 2409, 2411, 2413, 2415, 2417, 2419, and 2421,

wherein the isolated polynucleotide encodes a transcription factor.

4 . An oligonucleotide probe or primer comprising at least 10 contiguous residues complementary to 10 contiguous residues of a sequence of SEQ ID NO: 1-591, 1183-1912 and 1931-2106.

5 . An isolated polypeptide encoded by a polynucleotide according to any one of claims 1 - 3 .

6 . An isolated polypeptide comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 592, 594-850, 852-930, 932-951, 953-1046, 1048-1182, 1913-1930, 2107-2293, 2296-2368, 2372, 2375, 2378, 2386, 2388, 2390, 2392, 2394, 2396, 2398, 2400, 2402, 2404, 2406, 2408, 2410, 2412, 2414, 2416, 2418, 2420, and 2422.

7 . An isolated polypeptide comprising an amino acid sequence selected from the group consisting of:

(a) sequences having at least 75% identity to a sequence of SEQ ID NO: 592, 594-850, 852-930, 932-951, 953-1046, 1048-1182, 1913-1930, 2107-2293, 2296-2368, 2372, 2375, 2378, 2386, 2388, 2390, 2392, 2394, 2396, 2398, 2400, 2402, 2404, 2406, 2408, 2410, 2412, 2414, 2416, 2418, 2420, and 2422;

(b) sequences having at least 90% identity to a sequence of SEQ ID NO: 592, 594-850, 852-930, 932-951, 953-1046, 1048-1182, 1913-1930, 2107-2293, 2296-2368, 2372, 2375, 2378, 2386, 2388, 2390, 2392, 2394, 2396, 2398, 2400, 2402, 2404, 2406, 2408, 2410, 2412, 2414, 2416, 2418, 2420, and 2422; and

(c) sequences having at least 95% identity to a sequence of SEQ ID NO: 592, 594-850, 852-930, 932-951, 953-1046, 1048-1182, 1913-1930, 2107-2293, 2296-2368, 2372, 2375, 2378, 2386, 2388, 2390, 2392, 2394, 2396, 2398, 2400, 2402, 2404, 2406, 2408, 2410, 2412, 2414, 2416, 2418, 2420, and 2422,

wherein the isolated polypeptide functions as a transcription factor.

8 . An isolated polynucleotide that encodes a polypeptide according to any one of claims 6 and 7 .

9 . A genetic construct comprising a polynucleotide according to any one of claims 1 - 3 .

10 . A transgenic cell comprising a genetic construct according to claim 9 .

11 . A genetic construct comprising, in the 5′-3′ direction:

(a) a gene promoter sequence;

(b) a polynucleotide sequence comprising at least one of the following: (1) a polynucleotide coding for a polypeptide comprising a sequence of 592, 594-850, 852-930, 932-951, 953-1046, 1048-1182, 1913-1930, 2107-2293, 2296-2368, 2372, 2375, 2378, 2386, 2388, 2390, 2392, 2394, 2396, 2398, 2400, 2402, 2404, 2406, 2408, 2410, 2412, 2414, 2416, 2418, 2420, and 2422; and (2) a polynucleotide comprising a non-coding region of a polynucleotide of any one of claims 1 - 3 ; and

(c) a gene termination sequence.

12 . The genetic construct of claim 11 wherein the open reading frame is in a sense orientation.

13 . The genetic construct of claim 11 wherein the open reading frame is in an antisense orientation.

14 . A transgenic plant cell comprising a genetic construct of any one of claims 9 and 11 .

15 . A plant comprising a transgenic plant cell according to claim 14 , or fruit or seeds or propagules thereof.

16 . The plant of claim 15 wherein the plant is a woody plant.

17 . The plant of claim 16 wherein the plant is selected from the group consisting of eucalyptus, pine, acacia, poplar, sweetgum, teak and mahogany species

18 . A method for modifying gene expression in a plant comprising stably incorporating into the genome of the plant a genetic construct according to any one of claims 9 and 11 .

19 . The method of claim 18 , wherein the plant is a woody plant.

20 . The method of claim 18 , wherein the plant is selected from the group consisting of eucalyptus, pine, acacia, poplar, sweetgum, teak and mahogany species.

21 . A method for producing a plant having modified gene expression comprising:

(a) transforming a plant cell with a genetic construct according to any one of claims 9 and 11 to provide a transgenic cell; and

(b) cultivating the transgenic cell under conditions conducive to regeneration and mature plant growth.

22 . The method of claim 21 wherein the plant is a woody plant.

23 . The method of claim 21 wherein the plant is selected from the group consisting of eucalyptus, pine, acacia, poplar, sweetgum, teak and mahogany species.

24 . A method for modifying the activity of a polypeptide in a plant comprising stably incorporating into the genome of the plant a genetic construct according to any one of claims 9 and 11 .

25 . The method of claim 24 wherein the plant is a woody plant.

26 . The method of claim 24 wherein the plant is selected from the group consisting of eucalyptus, pine, acacia, poplar, sweetgum, teak and mahogany species.

27 . An isolated polypeptide comprising a DNA-binding domain, wherein the DNA-binding domain comprises an amino acid sequence selected from the group consisting of SEQ ID NO: 2279-2293 and 2296-2368.
