Abstract

Disclosed are compounds, compositions and methods for treating various diseases, syndromes, conditions and disorders, including pain. Such compounds are represented by Formula (I) as follows: wherein Y, Z, R 1 , and s are defined herein.

Claims (271)

1. A compound of Formula (I)

wherein

Y and Z are independently selected from a) or b) such that one of Y and

Z is selected from group a) and the other is selected from group b);

Group a) is unsubstituted C 6-10 aryl, Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of thiazolyl, pyridinyl, indolyl, indazolyl, benzoxazolyl, benzothiazolyl, benzofuranyl, benzothienyl, imidazo[1,2-a]pyridin-2-yl, quinolinyl, quinazolinyl, furanyl, thienyl, oxazolyl, pyrazolyl, and benzimidazolyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally

independently substituted with one to two substitutents selected from the group consisting of bromo, chloro, fluoro, iodo, C 1-4 alkyl, C 1-4 alkoxy, and trifluoromethyl; and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, quinolinyl, pyridinyl, thiazolyl, benzimidazolyl, pyrrolyl, and pyrazolyl;

wherein phenyl of the C 6-10 aryl and the heteroaryl are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; trifluoromethylthio; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; (2-cyano)ethylaminocarbonyl; cyano; carboxy; aminocarbonyl; C 1-4 alkylaminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; di(C 1-4 alkyl)aminosulfonyl; morpholin-4-ylsulfonyl; —OCH 2 O— attached at adjacent carbon atoms; and NR c R d ; wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, cyano(C 1-4 )alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, C 3-8 cycloalkylcarbonyl, C 3-8 cycloalkyl(C 1-4 )alkyl, aminocarbonyl, di(C 1-4 alkyl)aminocarbonyl, C 6-10 arylcarbonyl; di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl;

s is 0, 1 or 2; provided that when s is 2, R 1 is independently selected from the group consisting of phenyl, C 1-3 alkyl, and C 6-10 aryl(C 1-3 )alkyl;

when s is 1, R 1 is C 6-10 aryl, C 1-3 alkyl, benzyloxymethyl, hydroxy(C 1-3 )alkyl, aminocarbonyl, carboxy, trifluoromethyl, spirofused cyclopropyl, 3-oxo, or aryl(C 1-3 )alkyl; or, when s is 2 and R 1 is C 1-3 alkyl, the C 1-3 alkyl substituents are taken with the piperizinyl ring to form a 3,8-diaza-bicyclo[3.2.1]octanyl or 2,5-diaza-bicyclo[2.2.2]octanyl ring system;

with the proviso that when Y is phenyl, Z is other than 3-(pyrimidin-2-yl)phenyl, 4-(pyrimidin-2-yl)phenyl, 2-(2-fluorophenyl)phenyl, 2-(4-fluorophenyl)phenyl, 4-(3-cyanophenyl)phenyl 4-(pyrazol-1-yl)phenyl, 1-ethylcarbonyl-3,4-dihydro-1H-isoquinolin-2-yl, 2-phenylethynyl, 6-bromo-imidazo[1,2-α]pyridin-2-yl,

and with the proviso that a compound of Formula (I) is other than

a compound wherein Y is phenyl, Z is biphenyl-4-yl, s is 1, and R 1 is spirofused cyclopropyl;

and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof.

2. The compound of claim 1 wherein

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of thiazolyl, pyridinyl, indolyl, indazolyl, benzoxazolyl, benzothiazolyl, benzofuranyl, benzothienyl, quinolinyl, quinazolinyl, thienyl, furanyl, and benzimidazolyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of thienyl, quinolinyl, pyridinyl, pyrazolyl, and benzimidazolyl, and pyrrolyl;

wherein phenyl of the C 6-10 aryl, and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; (2-cyano)ethylaminocarbonyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl.

3. The compound of claim 2 wherein

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of thiazolyl, pyridinyl, indolyl, indazolyl, benzoxazolyl, benzothiazolyl, benzothienyl, quinolinyl, quinazolinyl, thienyl, and benzimidazolyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, quinolinyl, pyridinyl, benzimidazolyl, and pyrrolyl;

wherein phenyl of the C 6-10 aryl, and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; (2-cyano)ethylaminocarbonyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl, and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl.

4. The compound of claim 3 wherein

Group b) is

i) C 6-10 aryl;

ii) heteroaryl selected from the group consisting of indolyl, indazolyl, benzoxazolyl, benzothienyl, and thienyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, and quinolinyl;

wherein phenyl of the C 6-10 aryl and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 alkenyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl.

5. The compound of claim 1 wherein s is 0 or 1.

6. The compound of claim 5 wherein s is 0.

7. The compound of claim 1 wherein R 1 is phenyl, spirofused cyclopropyl, or C 1-3 alkyl.

8. The compound of claim 7 wherein R 1 is phenyl.

9. A compound of Formula (I)

wherein

Y and Z are independently selected from a) or b) such that one of Y and

Z is selected from group a) and the other is selected from group b);

Group a) is unsubstituted C 6-10 aryl;

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of thiazolyl, pyridinyl, indolyl, indazolyl, benzoxazolyl, benzothiazolyl, benzofuranyl, benzothienyl, quinolinyl, quinazolinyl, thienyl, furanyl, and benzimidazolyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of thienyl, quinolinyl, pyridinyl, pyrazolyl, and benzimidazolyl, and pyrrolyl; and

wherein phenyl of the C 6-10 aryl, and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; (2-cyano)ethylaminocarbonyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl;

s is 0 or 1;

R 1 is phenyl, spirofused cyclopropyl, or C 1-3 alkyl;

with the proviso that when Y is phenyl, Z is other than, 3-(pyrimidin-2-yl)phenyl, 4-(pyrimidin-2-yl)phenyl, 2-(2-fluorophenyl)phenyl, 2-(4-fluorophenyl)phenyl, 4-(3-cyanophenyl)phenyl 4-(pyrazol-1-yl)phenyl, 1-ethylcarbonyl-3,4-dihydro-1H-isoquinolin-2-yl, 2-phenylethynyl, 6-bromo-imidazo[1,2-α]pyridin-2-yl, and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof.

10. A compound of Formula (I)

wherein

Y and Z are independently selected from a) or b) such that one of Y and

Z is selected from group a) and the other is selected from group b);

Group a) is unsubstituted C 6-10 aryl;

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of thiazolyl, pyridinyl, indolyl, indazolyl, benzoxazolyl, benzothiazolyl, benzothienyl, quinolinyl, quinazolinyl, thienyl, and benzimidazolyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, quinolinyl, pyridinyl, benzimidazolyl, and pyrrolyl;

wherein phenyl of the C 6-10 -aryl, and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 alkenyl; (2-cyano)ethylaminocarbonyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl;

s is 0 or 1;

R 1 is phenyl;

with the proviso that when Y is phenyl, Z is other than, 3-(pyrimidin-2-yl)phenyl, 4-(pyrimidin-2-yl)phenyl, 2-(2-fluorophenyl)phenyl, 2-(4-fluorophenyl)phenyl, 4-(3-cyanophenyl)phenyl 4-(pyrazol-1-yl)phenyl, 1-ethylcarbonyl-3,4-dihydro-1H-isoquinolin-2-yl, 2-phenylethynyl, 6-bromo-imidazo[1,2-α]pyridin-2-yl,

and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof.

11. A compound of Formula (I)

wherein

Y and Z are independently selected from a) or b) such that one of Y and

Z is selected from group a) and the other is selected from group b);

Group a) is unsubstituted phenyl;

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of indolyl, indazolyl, benzoxazolyl, benzothienyl, and thienyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, and quinolinyl;

wherein phenyl of the C 6-10 aryl and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl;

s is 0 or 1;

R 1 is phenyl;

with the proviso that when Y is phenyl, Z is other than, 3-(pyrimidin-2-yl)phenyl, 4-(pyrimidin-2-yl)phenyl, 2-(2-fluorophenyl)phenyl, 2-(4-fluorophenyl)phenyl, 4-(3-cyanophenyl)phenyl 4-(pyrazol-1-yl)phenyl, 1-ethylcarbonyl-3,4-dihydro-1H-isoquinolin-2-yl, 2-phenylethynyl, 6-bromo-imidazo[1,2-α]pyridin-2-yl,

and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof.

12. A compound of Formula (I)

wherein

Y and Z are independently selected from a) or b) such that one of Y and

Z is selected from group a) and the other is selected from group b);

Group a) is unsubstituted phenyl;

Group b) is

i) C 6-10 aryl; or

ii) heteroaryl selected from the group consisting of indolyl, indazolyl, benzoxazolyl, benzothienyl, and thienyl;

wherein C 6-10 aryl and heteroaryl of Group b) are optionally independently substituted with one to two substituents independently selected from the group consisting of bromo, chloro, fluoro, iodo, methyl, methoxy, and trifluoromethyl, and are substituted with one additional substituent selected from the group consisting of

i) C 6-10 aryl; and

ii) heteroaryl selected from the group consisting of pyrimidinyl, thienyl, and quinolinyl;

wherein phenyl of the C 6-10 aryl and the heteroaryl substituent are optionally independently substituted with one to two substituents selected from the group consisting of C 1-4 alkoxycarbonylamino; C 1-4 alkoxycarbonyl; C 1-4 alkyl; cyanomethyl; C 1-4 alkoxy; one to three fluoro or chloro substituents; trifluoromethyl; trifluoromethoxy; C 1-4 alkylcarbonyl; C 1-4 alkoxycarbonyl(C 2-4 )alkenyl; cyano(C 2-4 )alkenyl; cyano; carboxy; aminocarbonyl; di(C 1-4 alkyl)aminocarbonyl; formyl; nitro; bromo; hydroxy; di(C 1-4 alkyl)aminosulfonyl; C 1-4 alkylsulfonyl; morpholin-4-ylsulfonyl; and NR c R d wherein R c is hydrogen or C 1-6 alkyl and wherein R d is hydrogen, C 1-6 alkyl, C 3-8 cycloalkyl, C 1-6 alkylcarbonyl, di(C 1-4 alkyl)aminosulfonyl, and C 1-4 alkylsulfonyl;

s is 0;

with the proviso that when Y is phenyl, Z is other than, 3-(pyrimidin-2-yl)phenyl, 4-(pyrimidin-2-yl)phenyl, 2-(2-fluorophenyl)phenyl, 2-(4-fluorophenyl)phenyl, 4-(3-cyanophenyl)phenyl 4-(pyrazol-1-yl)phenyl, 1-ethylcarbonyl-3,4-dihydro-1H-isoquinolin-2-yl, 2-phenylethynyl, 6-bromo-imidazo[1,2-α]pyridin-2-yl,

and enantiomers, diastereomers, and pharmaceutically acceptable salts thereof.

13. A compound of Formula (I)

selected from the group consisting of:

a compound wherein Y is phenyl, Z is 4-biphenyl, and s is 0;

a compound wherein Y is phenyl, Z is 4-pyrrol-1-ylphenyl, and s is 0;

a compound wherein Y is phenyl, Z is 4-(4-fluorophenyl)phenyl, and s is 0;

a compound wherein Y is phenyl, Z is 4-(4-methoxyphenyl)phenyl, and s is 0;

a compound wherein Y is phenyl, Z is 4-(2-chlorophenyl)phenyl, and s is 0;

a compound wherein Y is phenyl, Z is 4-(3,4-dichlorophenyl)phenyl, and s is 0;

a compound wherein Y is 4-biphenyl, Z is phenyl, and s is 1, and R 1 is 3-phenyl;

a compound wherein Y is phenyl, Z is 2-phenylbenzoxazol-6-yl, and s is 0;

a compound wherein Y is phenyl, Z is 6-phenyl-naphth-2-yl, and s is 0;

a compound wherein Y is phenyl, Z is 7-phenyl-quinolin-3-yl, and s is 0;

a compound wherein Y is phenyl, Z is 2-(4-methoxyphenyl)benzoxazol-6-yl, and s is 0;

a compound wherein Y is phenyl, Z is 5-phenylthien-2-yl, and s is 0;

a compound wherein Y is phenyl, Z is 4-methyl-2-phenylthiazol-5-yl, and s is 0;

a compound wherein Y is phenyl, Z is 4-methyl-2-thien-2-yl-thiazol-5-yl, and s is 0;

a compound wherein Y is phenyl, Z is 4-methyl-2-(4-methylphenyl)-thiazol-5-yl, and s is 0;

14. A pharmaceutical composition comprising a compound of claim 1 or 13 and at least one of a pharmaceutically acceptable carrier, a pharmaceutically acceptable excipient and a pharmaceutically acceptable diluent.

15. A pharmaceutical composition of claim 14 , wherein the composition is a solid oral dosage form.

16. A pharmaceutical composition of claim 14 , wherein the composition is a syrup, an elixir or a suspension.

17. A method for treating inflammatory pain in a subject in need thereof comprising administering to the subject a therapeutically effective amount of a compound of claim 1 or 13 .

18. The method of claim 17 wherein the inflammatory pain is due to inflammatory bowel disease, visceral pain, migraine, post-operative pain, osteoarthritis, rheumatoid arthritis, back pain, lower back pain, joint pain, abdominal pain, chest pain, labor pain, musculoskeletal diseases, skin diseases, toothache, pyresis, burn, sunburn, snake bite, venomous snake bite, spider bite, insect sting, neurogenic bladder, interstitial cystitis, urinary tract infection, rhinitis, contact dermatitis/hypersensitivity, itch, eczema, pharyngitis, mucositis, enteritis, irritable bowel syndrome, cholecystitis, pancreatitis, postmastectomy pain syndrome, menstrual pain, endometriosis, pain, pain due to physical trauma, headache, sinus headache, tension headache or arachnoiditis.
