Abstract

A nucleic acid-delivery vehicle for delivering nucleic acids to target cells is provided. The nucleic acid-delivery vehicle comprises a plurality of nanoparticles; and a plurality of nucleic acids. The nanoparticles further comprises one or more signal peptides attached to the nanoparticles. The nanoparticles and the nucleic acids are agglomerated to form a nucleic acid-granulation particle having a dimension of at least 20 nm. Methods of making the nucleic acid-delivery vehicle and kits comprising nucleic acid-delivery vehicle are also provided.

Claims (29)

1 . A nucleic acid-delivery vehicle comprising:

a plurality of nanoparticles, wherein one or more signal peptides are attached to each of the nanoparticles; and

a plurality of nucleic acids;

wherein the nanoparticles and the nucleic acids are agglomerated by non-covalent interaction to form a nucleic acid-granulation particle having a dimension of at least 5 nm.

2 . The nucleic acid-delivery vehicle of claim 1 , wherein the nucleic acid-granulation particle has a dimension in a range from about 5 nm to 10 μm.

3 . The nucleic acid-delivery vehicle of claim 1 , wherein the nanoparticles are selected from metal nanoparticles, metal oxide nanoparticles, metal nitride nanoparticles, metal carbides nanoparticles, and combinations thereof.

The nucleic acid-delivery vehicle of claim 1 , wherein the nanoparticles are selected from gold nanoparticles, iron nanoparticles, iron oxide nanoparticles, titanium nanoparticles, titanium oxide nanoparticles, silica nanoparticles, alumina nanoparticles, zinc nanoparticles, zinc oxide nanoparticles, tin nanoparticles, tin oxide nanoparticles, copper nanoparticles, copper oxide nanoparticles, hafnium nanoparticles, hafnia nanoparticles, yttria nanoparticles, yttrium aluminum garnet nanoparticles, magnesium aluminate spinel nanoparticles, magnesium oxide nanoparticles, calcium carbonate nanoparticles, calcium phosphate nanoparticles, diamond nanoparticles, silver nanoparticles, and platinum nanoparticles.

5 . The nucleic acid-delivery vehicle of claim 1 , wherein the nanoparticles comprise one or more specific primers attached the nanoparticles.

6 . The nucleic acid-delivery vehicle of claim 1 , wherein the nucleic acids are deoxyribonucleic acids or ribonucleic acids.

8 . The nucleic acid-delivery vehicle of claim 7 , wherein the deoxyribonucleic acids or ribonucleic acids comprise one or more tandem repeat sequences.

9 . The nucleic acid-delivery vehicle of claim 1 , wherein the nucleic acids comprise one or more nuclease-resistant bases, nucleotide analogs or synthetic nucleobases.

11 . A deoxyribonucleic acid-vaccine comprising the nucleic acid-delivery vehicle of claim 9 , wherein the nucleic acids code for an antigenic protein, or an antigenic peptide.

12 . The deoxyribonucleic acid-delivery vehicle of claim 1 , wherein the signal peptides are selected from cell surface signal peptides, nuclear localization signal peptides, cell penetration peptides or combinations thereof.

13 . A method of making a nucleic acid-delivery vehicle comprising:

providing a first solution comprising a plurality of nanoparticles and a second solution comprising a plurality of nucleic acids, wherein each of the nanoparticles comprises one or more signal peptides;

mixing the first solution and the second solution to form a mixed solution; and

dehydrating the mixed solution to generate a nucleic acid-granulation particle having a dimension of at least 5 nm for making the nucleic acid delivery vehicle.

14 . The method of claim 13 , wherein the nucleic acid-granulation particle has a dimension in a range from about 5 nm to 10 μm.

The nucleic acid-delivery vehicle of claim 13 , wherein the nanoparticles are selected from gold nanoparticles, iron nanoparticles, iron oxide nanoparticles, titanium nanoparticles, titanium oxide nanoparticles, silica nanoparticles, alumina nanoparticles, zinc nanoparticles, zinc oxide nanoparticles, tin nanoparticles, tin oxide nanoparticles, copper nanoparticles, copper oxide nanoparticles, hafnium nanoparticles, hafnia nanoparticles, yttria nanoparticles, yttrium aluminum garnet nanoparticles, magnesium aluminate spinel nanoparticles, magnesium oxide nanoparticles, calcium carbonate nanoparticles, calcium phosphate nanoparticles, diamond nanoparticles, silver nanoparticles, and platinum nanoparticles.

16 . The method of claim 13 , wherein the nucleic acids comprise rolling circle amplification products.

17 . The method of claim 13 , wherein the dehydrating is carried out by evaporation, wherein the evaporation is selected from spray drying granulation, wiped film evaporation, freeze dry granulation, or wet granulation.

18 . The method of claim 17 , wherein the evaporation is carried out by spray drying granulation.

19 . A kit for delivering nucleic acids to a cell comprising:

a nucleic acid-delivery vehicle, comprising:

a plurality of nanoparticles;

wherein at least one nanoparticle comprises one or more signal peptides attached to the nanoparticle.

20 . The kit of claim 19 , wherein the at least one nanoparticle with signal peptide comprises one or more specific primers attached to the nanoparticle.
